it's the entire reason that we have a pc in the first place. You could have bacteria and mold growing on it, throw it in the pc and it would come out sterile. This is the point.
There are invisible mold spore, vegetative mold, bacterial spore, and vegetative bacteria present just about everywhere and on every surface including your hands and body. That's why we have a pc.

This is another interesting thing I'll start tomorrow now that I have my shit set up again.

Im going to start a thread on the topic of the effectiveness of our ppressure cookers. I will run a series of tests that should be pretty interesting and entertaining.  One of these will be to whip up a batch of agar in a media bottle and let it sit for a week to get nasty as fuck, then pc and wait to see if it re-contaminates.

Another great test of endospore destruction would be to make an LC broth with a handful of different types of grain in each LC. If there are surviving endospores they should readily reactivate in the broth after about a day or two, as endospore reactivation takes place they multiply quickly when in a nutrientrich environment. They're killed too quickly in the broth alone because it's a liquid but should hypothetically be protected by the grain if the survival hypothesis is correct.  If they survive the broth will make for rapid expansion of the bacteria. 

Should be fun.

I would read that. Would love to see the results. If you wanted to take it to another level you could run the same scenarios between pour and no pour.

difficult to do with the pour as it would be in a plastic petri. Im just going to use the media bottle as the vessel, the results should be the same for both scenarios.

I'm not saying you're wrong p9. Dead bacteria is dead bacteria afaik.

I'm just saying i'm probably not the only person that's gonna forget the brown sludge thing on one plate(of 20ish) is just dead bacteria. or worse, multiple spots on multiple plates.
Agar is cheap and easy. I can see the practical side of just making clean plates like bear's saying.
If you really got make the plates work, maybe mark them or something.

Do a write up and push the limits dude. I'll watch it.

I always thought the primary worry of letting things grow too much in a media even if you kill it afterwards is the potential inorganic byproducts they leave behind. Would be interesting to see if that really makes a difference post PC or if the heat just takes care of it all.

Any remnant or product of a living organism would be biotic and organic.
The opposite could be true though, living organisms could turn inorganic material into organic products.

So, why do you think (at least enough to do this experiment) there might not be complete destruction of endospores in a PC? Do you know of any extremophiles that could withstand these conditions? I'd assume that if there are any they live in pretty specific environments (geothermal vents, hot springs etc.) and aren't floating around our homes.

no Im on the side of complete destruction, I don't think that the endospores survive for a number of reasons, but there are others who believe that some small amount survive, and still more who are unsure.
Im no entirely sure if my test would be sufficient but it seems reasonable that it would be a good test.

I think that our experience tells us that endospores are totally destroyed because of methods like grain to grain expansion. Endospores protect themselves with many layered  protein coats that surround their core which make them thermophilic, however these spores reactivate in a suitable environment in a day or two and then double in number every 20 mins. G2g would be expanding bacteria exponentially along with mycelium.

Yeah, I mean in laboratory settings autoclaves that work similarly to pressure cookers (steam at 15 PSI) are used to sterilize media and have been for many years now. I'd guess if there were a chance this wasnt 100% successful it would be known by now but an experiment like the one you're describing would definitely be interesting.

Spores withstand 121°C for up to 12 min, and this has made the organism ideal for testing autoclaves that run on a time–temperature cycle designed to ensure the destruction of spores."

The argument against destruction isnt without some merit but I don't think it holds up to our experience.  Longer cycle times are required to penetrate the dense grain media, 2 hours as apposed to the 12 mins stated above, but for reasons mentioned earlier I think that 2 hours has been proven to be sufficient or else we would fail in some of our adopted practices, at least this seems reasonable to me.

This is why I want to run some tests to see how far I can push it.

No doubt I know by now you value clarity and directness where there's knowledge to be mined. But yeah meant chemicals, don't know what bacteria could produce but mycotoxins created by mold could be an issue.

Cursory searching seems to show that they're relatively heat resistant but I don't know what that actually means, maybe 25 minutes at 15 PSI is more than enough for most. Would actually be really curious to know more about how various chemicals change when sterilized, like could a fully pressure cooked jar of some molds still be potentially dangerous if opened?
