Well you got spore germination and some contaminant going on there.

I wud transfer some of that white goodness off the plate in the 3rd pic once it grows out a bit more.

Is that dirt on/in your plates? The green shit is obviously contamination.

I don't see green in any of the plates (probably bad lighting)
The black specs are from the soot from a butane lighter heating my butter knife in place of an inoculation loop.

Go for the white areas. Germ plates are generally more suspect. Things start looking nicer at transfer 2/3

Plate 3 is probably the only one I'd mess with if I was you. You can get more than enough off one plate to get things going

How did you inoculate these, with spore solution or from a print?


It it were me, I wouldn't use any of these and inoculate a new plate. Just inoculate one or two this time so you don't waste plates.

There seems to be mold that has sporulated in places, at least that is what those brackish looking spots look like to me on my phone.

from a print. I have some other ones that look a bit more promising. I'll try and get pictures later

Why does your agar look all clawed up?

Are you using an inoculation loop?

Sorta... I didn't have one or anything to make one with so after some searching on here I found some people who used butter knife in place of an inoculation loop so I gave it a try. Also, I feel like I am going to deep with it. I just ordered an inoculation loop set up (handle with replaceable loops) and gonna give it another go after watching some videos. I think using an actual loop will help cut down soot and probably will help prevent me from streaking too deep into the agar.

Really? A butter knife is the last thing I would use.
My first inoculation loop was a pice of wire bent into a loop and twisted with pliers.
No handle or anything,  just some wire that can scrap spore onto agar.

Yeah, a butter knife would be a terrible loop. A scalpel or Xacto would do much better.

People say you can cut some spores with them, but I've never seen it matter.

Update from the same set of dishes. Sorry for the heavy condensation.

This is the front and back of the one I think is doing the best. I used the 4 quadrant streak pattern for these.



Here some other ransoms I can see strings of what I think look like rhizomorphic growth. I really wish the camera could pick up on it a little better but it looks like strings reaching out.




This last one confuses me because it looks like healthy mycelium growth but it’s growing from where I touched my hot knife to cool down. I rubbed the spores down into the center of this dish

I would try and stay away from the edges of the plate when streaking/transferring

A butter knife, or wire work or anything really, but loops are cheap af and make life easier. Highly recommend it BHO

I actually just picked up a reusable handle with 50 replaceable loops. Also working on PCing some jars full of individually wrapped swabs. I've been meaning to PC the swabs for some time but havent got around to prep any grain to go in their with yet.

What do you mean when you say "streaking too deep into the agar"? Are you mashing spores down into it? Is that why the plates look so butchered?

Streaking spores should be a quick gentle wipe across the surface with as few spores as possible. It's not a scoop some up and mix like oatmeal type of affair.

Here is an update on my first ever agar. I’ve picked out some plates to transfer.

I used D3’s tek with the leather hole punch.



Also I’ve been noticing brown speckles in my freshly poured LME plates. Am I supposed to be filtering this stuff?
