LC to LC?

I don't see any reason why this wouldn't work but I'd love to know if it can't for whatever reason. say I've got favorable genetics in LC ready to rock, could I drip drop a bit of that LC into a fresh LC to expand it? would it be better to put that LC onto agar first and then that agar to LC?

gotcha, thanks for the reply sandman I was just thinking that may be the easiest way to continuously expand a good culture I like as long as I have a master plate/LC/slant stored away.

It's both the easiest way to expand cube myc and bacteria/ yeast/ trich/ ... If the LC is clean and your sterile tek is on point it'll work out wonderfully. I'd highly advise testing on agar first.

good point thanks verum done in front of a fh should give little problems i assume - only of course by taking sterile precaution I don't plan on doing this until i have my fh rockin and you mean testing my LC on agar every time before expansion?

Yes. I have used a shit ton of LC and test on agar every time. It's easy to spot a horrible LC but every now and then an LC can look good and be bad. Nothing worse than throwing an LC that looks good into a shit load of jars and then finding that it wasn't as good as you thunk it was. If your eye and tek are good, you can be pretty confident in your LC but it a risk I don't recommend taking and testing on agar only takes a second.
If one were to test an LC, find it clean and then shoot or pour that LC into a couple gallons, I'd say those gallons would probably be reliable but would still test them before going to grain. You can test them on grain instead of agar but agar is easier to see what's really going on..

Thanks for the explanation. What vessel would you use for an entire gallon of LC? I guess anything big enough to fit in the PC.. maybe an empty bottle of jack or even a growler bottle would work. If I nocced something that large could I pour it into a mason jar in front of a fh or would that be rolling the dice. I guess the only way to find out would be testing the receiving mason jar on a plate before using it - hmmmm after transferring some of the LC in the gallon to a smaller bottle - would it need to sit on the shelf for a bit to let any would-be contaminates grow out to show on the plate I test it on or could the test be done within the same session?
if thats confusing at all, just let me know and I'll rephrase

I like whiskey handles. My old set up was a pickle jar fabricated to use with the socerex automatic refilling syringe but more of a hassle. Nowadays I just pour from whiskey handle to grain.
As stated above, If the original LC is clean and tested and you have proper sterile tek on the lockdown you could be 94.57294573 % sure your subsequent LCs are clean. I wouldn't risk it though. I'd let them sit and colonize then test four or five days before I plan on using them.

Oh dope. I like the idea of those socerex syringes, seems like they take away almost all vectors for contamination granted you have a clean LC- why do you find them a hassle? I think the syringe is especially useful for noccing up spawn bags, and covering the hole with glue like I've seen Vinci and others do. but for jars, i imagine pouring is much easier.

I'd rather pour than poke any day. You can poke and tape but I don't see it being any easier, quicker or safer.
The Socorex is a hassle in a few ways. None are a huge issue in and of themselves but together it makes it much more of a chore than just pouring from a handle. If it's left to sit a few days after pumping Myc laden nutrified water through the syringe, The myc grows all around the mechanisms within and jams them up and causes all manner of problems. That is the biggest issue. It can be over come by tilting the LC jar and pumping air through the syringe or by being sure to pump a few squirts through her every other day or so but I've found just using a handle to be easier and smoother.

aye aye captain heard loud and clear. do you just eyeball your pouring amount - give it a 1-2-3 count or something? also using that much LC in a single container, do you find it bad practice to use it for multiple sessions? as in using it to noc up some amount of jars/bags whatever - and capping it up and saving the rest for another time. would you then want to test it for contaminates before the next use?

I just prep my grain dry enough to accept a bunch of LC and then pour it in there. Don't count or measure in any way. Just a bloop will do.
It's obviously safest to use it all up but I don't mind capping her and saving her for later. But, I'm not half bad with my sterile tek either. If I had ongoing contam troubles and wasn't satisfied with my sterile sessions I wouldn't be using liquids in the first place (they are just easier to fuck up than other things) and I'd certainly test again if I left her sitting around for any amount of time. Reason being, if one were to drop a little piece of shite in an otherwise clean LC and then pour immediately, that little pice of shite would only make it into so many bags. However, if one were to shite in an LC and then let her sit on the shelf for a week, that shite would exponentially expand and the entire LC would become a tornado of shite.
