Peptone an Yeast Extract Powder Question

What's up bitches? This is my first post but I must have been lurking here for well over 10 years. And I bet you never heard that before from a first time poster...

So I am getting into liquid culture and agar and the subject of peptone came up. (LC haters be gone, this is working for me.) I understand the basics of amino acids and such building blocks being used for developing mycelium. The guy who makes YouTube videos for Mossy Creek seems to like this stuff too, so I am giving it a shot. On to the problem:

I ordered very specifically "Bacteriological Peptone" as recommended by Mossy Creek. I also realize that "soy based peptone" is the stuff recommended in Radical Mycology. They seem to be very similar products.

So...I went to Amazon and ordered the "Bacteriological Peptone." They screwed that up and sent me "Yeast Extract Powder." The bottle looked identical and I put it up on the shelf until I needed it which was today. So now I am passed the point of no-returns and I am stuck with this stuff. Soy-based is on the way, but I would like to make some LC before that arrives. So it looks like Yeast Extract Powder is used to for similar purposes.

Does anyone have some experience with this stuff? Bacteriological, Soy, or Yeast Extract? I know that much of the contents of these products overlap, but could I use yeast extract until the other order arrives? Or is that a total waste of time and barking up the wrong tree? Searches on peptone and other key words do not turn up much other than hearsay and some really short posts with the only reply being something similar to "I don't know."

Honestly I personally make LC with 1.6% LME and nothing else and get crystal clear cultures that grow crazy. My personal approach to mycology is simple is best. There are some very precise and complex formulations and Teks floating around that can literally beat their simple 1 ingredient counterparts by hours. Literal hours shaved off of weeks worth of time. Patience is King here and adding steps and ingredients is not a substitute.

My point is Peptone works and some people swear by it but it's really not needed and shouldn't hold you back from making LC. All you need is water and sugar, the more stable the sugar the better.

I used yeast before I got peptone, idk if I ever used just malted. But peptone seems to be making a difference.

Why'd you get soy based? I got some of that, but for my cordyceps.

I'm going to do trials with the regular, soy and casein hydrosolate withcubes.
Also going to try glucose instead of maltectracts soon too. They got good results with it with cordyceps.

And honestly I'll probably just use it for my agar soon.LC just doesn't seem to take off like I ever hope.

The soy based peptone is mentioned specifically by the book Radical Mycology. He is specific on soy based as I think this was based on research. The book is well researched although his references on this are spotty.

I have never heard of "bacteriological peptone" until the guy at Mossy Creek started swearing by it. I tried to buy it, but was sent a bottle of yeast extract instead.

I keep my temps between 65 and 70 in my grow space. This works well because it is a basement and that's where it hovers without much effort on my part. Contams are about not existent, but things grow slowly. That's okay, I am not in a rush. But I started working with with mex stones. And wow, it takes over a month to colonize a jar. It wound up okay, but it is slow. Switched to LC and the shit rips right through a jar. That improvement was pretty good, but I was going to slowly introduce a couple new factors one at a time and see how it went. Peptone seemed like a good choice. Until they sent me the wrong bottle.

But in the end I might wind up increasing the temp. Summer may do that for me anyway.

Peptone Is use in liquid culture just as body builders uses protein powder after a workout.

My opinion It's not necessary at all.

My gripe with nutritional yeast and peptone is that you never know If you have a bacterial bloom due to the cloudy liquid that never clears up.

Keep It simple..

If you ever have time to experiment Try 4% broth:

1000ml. water
20 grams malt extract
20 grams dextrose (corn sugar)

I do small batches in small jars. 140ml. per jar.

In eight days your lc is thick and ready to use.

Well the going opinion seems to be just don't bother with it. But I already ordered the bottle, so you know that is not happening. So I'll  just have to experiment.

For the record my stone producing LCs are just made from my grain boil. I boil oats in water, coffee, and gypsum. I save the rest of the liquid, load it up into more quart jars, sterilize them and put them on the shelf. When I want to make LCs I break one out and put 100 ml of the above mentioned boil water along with 400 ml of tap water and a stir bar into a quart jar. That's it. This group of stones will get nothing but that. I'll do the next set with peptone and see what happens.
