Agaricus Campestris Multispore Germination [Re: Farm3r] * 2
    #28302175 - 05/01/23 12:34 PM (8 months, 24 days ago)

Just in case anyone comes looking in the future!  Multispore germination of Agaricus Campestris spores.  Media is PDYA w/ addition of 4g/L of powdered activated charcoal.  ~9 month old Spore print from a wild specimen.  Scraped about 1cm square section in 2-3ml Sterile H2O and soaked for 4.5 hours for one plate and 24 hours for the other.  No significant germination variation as a result.  3 drops per plate in different areas. Not sure if the activated charcoal was necessary, but my first attempt at germination failed.  The first plates where streaked with an inoculation loop (microliters of spore solution...).

Hope to be posting pictures of A. Campestris fruit in another couple months!

Turkey Tail is starting to fruit. Reishi getting taller. Next round in Jars. 4 Shiitake, 4 Maitake, 2 Reishi and 2 Turkey Tail. Shook Turkey Tail this morning. It looked almost completely colonized already. Very fast. Shiitake and Maitake take a long time compared to the others.

Wild collected exidia recisa. The smell of the mycelium is slightly reminiscent of moth balls.

It's amazing the spectrum of smells with mycelium from different mushrooms. Shiitake and pioppino smell terrible to me. Cubensis mycelium smells divine. Chestnut mycelium smells just like methaqualone. Weird stuff

Cordyceps a few days after being exposed to light . My tablet camera does not do the color justice.
Interesting to note, the mycelium starts changing color after only a few hours of light exposure.

First time I’ve fruited these oysters that I cloned from some I bought at the supermarket. Third time’s the charm but I think I’ve got the whole process sorted now.

They were just labelled “oyster mushroom”, but I think they might be some kind of pearl? Super excited!

A cordyceps jar I grew out awhile ago,
(I think it was something like 27g wet, 6-7g dry)

and the new bulk grows based on the same LC (inoculated bulk grow bag 4/5, transferred to 3 bins on 4/14, introduced light after recovery on 4/17)

My post is at the bottom of this thread regarding ratios.

It's brown rice with additives (per NorthSpore recommendations...basically a pinch of gypsum, bit of nutritional yeast, multivitamin, maybe one or two other things, as a broth mixed to whole brown rice. I just left out whatever was too hard to get (peptone, maybe a few other things). I doubt any adjustments to the nutritional value of the substrate actually contributes to its growth and moreso relates to its active compounds but who knows.

In any wase, I did a bulk bag to 30% or so, broke up, let it recover to full colonization, then opened it up and spawned it to 3 different tubs,  let it barely recover up, then sent it to light.

My biggest issue with the whole process is finding the correct water ratio (for this guys post, which detailed just venting in the PC to cook the rice in the bag with the broth). I like that for this nutritional additive grain, but the ratios everyone else seems to be using absolutely doesn't work for me, regardless of jar or bulk. WAY WAY WAY too wet. So I'll be experimenting there. Glad I could recover this grow by removing a bunch of water though.

It did still end up "sweating" quite a bit of liquid during it's recovery colonization and oranging stage. Like it is almost as if during recovery fully colonizes in the tub it tightens up the substrate and squeeze out water, so I just took a syringe with blunt 18g and removed pooling water from the edge. Now looks much more appropriate. There were some "dark" areas of grain, but those are more like ultra tight clumps of grain I didn't break up appropriately than any sort of spreading contamination.

Smaller bin:

Larger bin 1:

Larger bin 2:

Larger bin 2 close:

(my two syringes have some small amount of albino mushroom production in their initial jars, I'm not concerned about the tiny little blips of white)

If those albinos actually grow out to full sized fruits, CLONE them! Would be awesome if they grew out and the culture stayed albino. Terrestrial fungi wants almost $100 for a syringe of his albino variety. Based on a paper I found, the albino strain tested produced more antioxidants.
I need to get my jars prepped for my next cordyceps batches, the LC is just about ready. I have 5 cultures I’ll  be growing side by side. I’m going to be using the brown rice and broth recipe from North Spores’s article again, plus some live hornworm. I just got a mini fridge and the inkbird temp controller is on order, so I’ll be able to grow these through the summer.

Pic tax- Lions mane LC test plate that I let sit out. For the hell of it,  I cloned a piece of LM from the grocery store directly into the LC using joesex poke method. I even did it in open air

Hehe well you know where I got my syringe from. Granted I didn't order an albino variant, just the cheapest one. Got sent two. One of the two seem more multi genetic than the other, but both have produced a few albinos. I cloned one and it's on agar now. I'll do some set n forget jar grows for clone experiments and post on here. For syringe 1 it had much better and faster growth, so I continued with that for this bulk grow. Syringe 2 was varied, so I cloned the biggest boy, the biggest cluster, and the biggest albino.

These pics were like 6 days before harvesting. Like I said they were sloooww. Like 10 days slower than the other syringe.

Jar 1:

Jar 2:

These were done around 70-74 f in my living room next to my offbrand aerogarden. Probably got lucky since my GF let temps hit 78 a few times.

Multi strain cordyceps run under way.
Left to right is Asian commercial strain AS-4, MMX3 from fresh fungi, then beast and high five  from AP Gold

Then I only had 4 viable hornworm pupas, I infected one per strain, but the syringes got dropped and fell out of labeled sleeves by the time I got to these so I have no idea which is which lol
