New to LC - Can you take a look and let me know your thoughts on contamination?
    #27307102 - 05/13/21 02:00 PM (3 years, 8 months ago)

I don't use LC because I grow very small batches of many different things, but I've been wanting to grow King Oysters regularly so that gives me a reason for going with LC.

These are my first two LC jars. I did the 4% LME formula, PC'd for an hour, knocked up with a small chunk from a clone in my SAB. My sterile technique is good - I haven't ever put tissue from a clone onto an agar plate and had it contaminate on me, and I used the same basic technique to knock up the LC (rip stem in half with gloved hand, then use sterilized scalpel to carve out a small sample from the center of the stem, being careful to avoid having non-sterile items above the split stem or LC jar, etc).

So both jars, same formula, tissue from the same clone. The only difference is that I put one of those protein shake stainless steel whisk balls into one of jars to help break up mycelium when shaking. But these jars look completely different. I do see a mycelium blob steadily growing in each jar, but the jar on the left looks very cloudy compared to the jar on the right so I'm assuming it's bacterial contamination? I read about waiting to see if the mycelium wins the war and the jar clears up but I didn't see anything about how long that could take (jars were knocked up on 5/3).

Thanks in advance!

I'm no expert in LC, just getting my first ones set up so bear that in mind. But I'd personally say bacterial in the left yes. One thing I will suggest regardless is switching to one piece plastic lids. They make it much easier to inoculate.

Thanks - yeah I have been using two piece solids for LC and 2 piece with 1/4" holes and a single large SFD with great success so far, but from all the things I've read from the TC's on just using unmodified plastic lids for everything including grain jars, I think I might give it a shot.

we can say it looks funny or good, there really is know way to tell unless you put it to agar or gains.

The color difference is rust.

4% is wayyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyyy too high for LME LC. Try 0.1-0.5% for clearer cultures.

Clone to LC is very very risky. Like don't do that risky. Go to agar then LC.

Thanks for your reply. Shouldn't be rust though as the whisk is stainless steel. I thought 4% was the gold standard for LC! I found that from all my searches. Doh!!!!! As I added the powder I kept thinking to myself "man this seems like an awful lot" but trusted what I read. Great info thank you, and thanks for the note on going agar to LC - had no idea on that one either but it does make sense as to why it would be less risk!
