Hi all, requesting thoughts on what's going on here.

I left this jar of golden oyster on oats for a long time after full colonization. Probably at least 3-4 weeks, I got distracted by school and life and stuff. It looked happy, strong and white a few weeks ago.

Maybe bacteria surrounded by heavy metabolites? Lid has 2x sticker style SFD. Maybe the myc weakened after going too long in the jar allowing the bacteria to take a foothold?

I've read that mushrooms make metabolites under stress and also when the jar has been fully colonized for a while, but it's surprising that it's that much, and that there are black splotches too.

I'm having trouble identifying healthy mycelium, it's probably too early to tell for the lions mane, but do these plates look fine to every one? My technique for putting these on a petri after flaming the needle wasn't great, I didn't wait for it to cool down enough and many spread culture all over

Pink oyster (looks a little wispier than I'd like, but I think it's OK):


Lions mane, the typical hard to see mycelium:


Ganoderma, the spot at the bottom right worries me a bit, but they both look like very similar types of mycelium:



Finally, I know for sure that this is contaminated, that white layer on everything isn't condensation, it's something growing on the plate, and the dot in the middle I'm pretty sure is ganoderma mycelium. Does anybody know what the white layer is though? I had the same thing on another plate, maybe yeast?

Yeh most likely yeast, it will have a sour smell if you open it.
Shine a light from the back on the two spots of growth, pretty sure there's a contam piggybacking there.

So, the surgery I performed two pages back in this thread appears to have gone pretty well! I think I should introduce fruiting conditions in like, 4-7 days? Please advise.

About the tape: I HAD polyfill in the holes, but after watching some videos, decided that it was unnecessary so I closed the lid (not snapped shut) and have a lot more humidity now. Stopped opening it all the time to spray. The spikes of mycelium have decreased. Entire thing feels like a gigantic block of mycelium.

Oh yeah it's terrible hahaha.  I have another tub though for my next attempt.

How will I KNOW it's ready for fruiting conditions (more FAE)?

Thanks, but I don't think so, since I have a perfectly healthy plate from the same syringe. Or maybe I do have a contam there and don't realize, here are some pics:

how did you sterilize the scalpel between transfers? if you used flame or torch did you let it get red hot? looks like mold to me.. or to wet of agar at best.

Hey everyone, what might be causing contams on the edge of plates? I think in most of my cases I poured them wrong (agar got on the edges), but I'm sure I poured this plate in the picture right and still got contams:



Could it be how I'm storing them? I don't think it's my sterile procedure inside my SAB, since it would be localized contam, not distributed relatively evenly along the edge of the plate. Could it be that I'm putting the petris inside the sleeve and the slight pressure from the air is pushing contams in? I also stored the plates upside down in an effort to control condensation, could it have been that?

I think it could be any of the following:
- plates were dirty to begin with, prior to pour (a tear in the bag)
- your parafilm or whatever you use to keep em' shut isn't keeping airflow out, it may also be torn or warped
- agar PC problem (not long enough / at the wrong PSI)
- like you said, if you use a SAB and pour all your plates, and store them back in the original sleeve which is now not air tight that could be an issue
ive seen people (d3monic) store their plates back in the sleeve in front of a fh but not too sure if it has the same success rate inside a SAB, unless you maybe close up the arm holes :shrug: either way I wrap my plates as soon as they cool and have never had an issue with contam like that

Thanks, I might just start wrapping them after pouring/solidifying. It's a pain, but I've lost a good amount of plates to contam like that already.

I'd just like to say that I did eventually rid myself of all of the contamination using 1/100 bleach solution and scooping up a couple of sporadic spot of slime mold.

Hi all, first post... sorry if this is not the proper place... I've recently started this new hobby and would like some advice on possible contamination.

I inoculated some standard PFTeks a few weeks ago (they were too cold for the first week, so I think nothing happened). However, now I'm seeing the following from most of my Tek's (which I believe is good, note green is just reflection on jar)



And this is what the 4% honey liquid cultures looked like for most of them (not sure if this is right)



However, one of them has the following Tek and LC. I orinally thought the floaters were carmelized honey from the pressure cooking, but they've been continuing to grow... Can anyone let me know if they think this is contamination or growth?

Liquid culture to agar first attempt.
These are results since May 9th evening. Cubensis B+. Wait to grow out for longer until full plate of stronger white mycellium before transferring to next agar plate (I've got two spare remaining) or take a transfer of the whitest section now at this stage. Signs of any contamination here?

Or simply no need to repeat transfer and go straight to grain?
