No growth after two weeks in fruiting chamber

About 6 weeks ago I inoculated two small autoclaved PF Tek bags, after about 1-2 weeks I shook them up and another 2 weeks later one of the blocks appeared to be fully colonized and not growing anymore. Although one thing I noticed is that, compared to other blocks or BRF cakes that are fully colonized, everyone elses appears to have mycelium completely covering the surface so the block looks just white and fuzzy, but my mycelium never grew in anywhere near that thick, it was certainly visibly white, but you could also very clearly see all the grains and what not, and the surface was rough from the PF tek materials and not fuzzy from mycelium like ive always seen.

I then put it into my SFC with hydrated perlite on some foil, I fanned and misted it 3-4 times a day keeping it around 18-22 celcius and almost two weeks later, the only growth was some super super tiny small white dots (primordia i think) which were around after 1 week, then after about 10 days I noticed a tiny bit of blue contam, and 2 days later it had took over a large portion of the block and I considered it lost and binned it.

What happened here? I don't believe I ever saw contam during the entire colonization process, so it seems likely that something happened during fruiting? Any response will be greatly appreciated!

did you sterilize those bags yourself or buy them that way?

Could be that it was birthed with uncolonized parts that grew contamination after the fact.  Did you take pics along the way?  Pics really help us see what exactly is going on.  I highly recommend taking pics as you go in case situations arise like this.. especially if you’re new to the game

Sorry, I didn't manage to take any pics, but it seems like that could be a possibility. As I mentioned  I inoculated two bags, the second one I still have in the bag because there were parts on the bottom that weren't colonizing, guess I mustn't have shaken it up good enough. I've shaken it up again to see if I can spur some thick mycelium growth, but is it bad that I've had it colonizing for almost six weeks now? Keep in mind the block is only about 1kg so its pretty small. Maybe I just got some really shit genetics and it takes forever to colonize.

I bought the bags pre-sterilized by an autoclave because since this is my first time I wanted to make it as simple as possible so I didn't fuck it up, but I guess I managed to find a way :frown: Hopefully  I have better luck in the future.

I’m assuming you used a multispore syringe to inoculate?  If so, they could very well be bacterial.. that would explain the slow growth/uncolonized areas.  Usually you can’t see bacteria, but you can see the affects of it.. was there metabolites (yellow liquid) or a lot of heat that the block produced with lots of condensation inside the bag? 

Bacteria will weaken myc and once it’s birthed, that weak myc can’t fight off competitor molds and that can also be why you didn’t see contamination until after birth.

I managed to get my hands on a cubensis LC syringe, I feel pretty confident that it wasn't contaminated,  but who knows. And I don't think I saw any yellow liquid, maybe some muddy colored liquid but that wouldve just been from the mix, and I can't remember exactly, but I think I might've seen condensation once or twice, but it'd be there one day and gone the next.

seems to me there are 3 possibilities

1- the bags were not well sterilized. i bought ready-made bags once and 6 out of 10 were contaminated. never again.

2- the LC was contaminated. ive never bought LC so i dont know

3- you contaminated them when you injected your LC. did you flame the needle well? inject them in a still air box?

i hope not but it sure sounds like your other bags are dirty too. if there are parts that dont want to colonize, or if the myc struggles to recover after you shake them up, bad news

just my 2 cents but i think a nice simple pf tek with a MS syringe is the way to go for your first grow. also i would highly recommend trying a humidity chamber instead of a sgfc. its much easier to look after and you can repurpose it if you want to do a monotub later on

I made the stupid mistake of wiping my needle with isopropyl alcohol after flaming it so its possible that couldve been the cause. As for the LC, I've made two of my own liquid culture solutions with that syringe I had. One of the solutions grew pretty well, until it filled about 1/4 of the jar, then it stopped, which I'm guessing is because I messed up the measurements of corn syrup, but the second one seems to be growing well so far, so the LC syringe doesnt appear to have any obvious signs of contamination, I guess I won't be able to tell until I try and use my LC's

you can see bacterial contamination in LC but mold hides in plain sight. im not saying thats whats happening but its good to know.

i dont think wiping your needle with iso was the problem...id still put my money on dirty bags
