My first sets of agar- ever
    #27297966 - 05/07/21 01:03 AM (2 years, 8 months ago)

Working with my first batches of agar, finally. Thankfully 41/45 of the plates look to be contam free so far.

my current goal is to grow out some fruit ASAP to get a clone to work off of instead of isolating/testing monocultures one by one. I'll get to that as well- but my priority is to get some consistent flushes going first.

these plates below are from a blue meanie print, and are the first to take off between the 3 strains i got on agar now. These are just 7 of the 12 or 13 i have- and they all look more or less the same as this. I know im supposed to transfer the aggressive myc out- but it looks pretty even and all over the place currently.  What would you do- and why?

thanks! much love

Let them go a bit longer and take samples from the healthiest areas and transfer to agar again.

Okk, i'll be able to work on this in the next ~50 hours im assuming that wont be too late.

Once i've transferred the healthy myc over to the second set of agar, given they grow out clean, i could go to grains afterwards and test? Or am i still looking to xfer multiple times even if i'm not trying to isolate right now.

If i understand correctly, the first transfer would be to eliminate the slower/weaker colonizing myc in the first round, and also possible contams that you can't see? i just read that germination plates could look clean but still have contams present

thanks for the reply

Most people I see recommend transferring 3 times. So when you transfer to the next plates would be T1. Then T2, and T3 they put to grain. I've put some T2s to grain but only the best looking ones. My roommate on the other hand tries to put T1 and T2's to grain all the time no matter how uneven they are. He ends up with like 2 out of 10 jars surviving on average. Super impatient and it bites him in the ass.

Ive had what appeared.to be clean germ plates, but i always make.at least one transfer cuz contaminated germ plates are common. For me that wud be the only reason why. Im against narrowing genetics.

ive been wondering about this.
Isnt diversity what encourages the most healthy species?
Like if strains are degrading over time that has to be a sign of something and if there is a problem w lets say B+ starting to become unstable and shit for a lot of people because its essentially being incested into Queen of England genetic diversity a responsibility for us to recognize as a bad thing and try to avoid? lol idk mayb im overthinking it
