Waiting for a master slant to colonize wood before refrigeration?
    #27288738 - 05/01/21 06:37 AM (3 years, 8 months ago)

The wood sticks in my master slant are maybe an 1/8 inch below surface at its closest point near top of tube.  Should I wait till it colonizes the surface and then dig lower before refrigeration or will it colonize further in fridge?  I'm using carbon agar so I won't be able to see it penetrate into agar so not sure how to handle this.  Thanks.

I like to allow my culture to fully colonize the exposed surface of the agar, then allow for it to travel under the bottom of the tube before refrigerating. I find that this helps with revival should your slant remain in the fridge for a very long time. After years of temp fluctuations and possibly drying a bit, the underlying mycelium is better preserved if shit goes sideways in the fridge.

If you were doing this with Pleurotus it would continue to expand in the fridge but not so with cubes.
