My Grain is not colonizing
    #27287393 - 04/30/21 02:15 AM (3 years, 8 months ago)

Hey I hope this is the right place to post this. I inoculated 3 jars of popcorn and 3 jars of rye with a couple different strains of lc. It’s been almost 3 weeks and nothing. No contams, no growth. I soaked, boiled and pc’ed. Let cool 24 hours, 1cc of LC in each jar. Polyfil lids. I am so frustrated. Where did I go wrong? Is it the Lc? I left the syringes in the fridge but let them warm to room temp before using.

Did you purchase LC or did you make them? I'm 0 for 1 with LC onto grains and 0 for 1 with LC onto agar.

Not pink oyster LC right?

I bought them. But I can’t believe 3 different LCs didn’t work... like it’s gotta be something I’m doing wrong. Even though I was clean af and followed the tek to the T. I’m thinking of getting some spores and going to agar first. I’ve just been a little intimidated.

I've also heard some people refer to spore syringes as LC because they didn't know the difference. ( not saying that's the case here op ) unfortunately I don't use LC so I don't have much to add Kat.

Edit: also agar is the way to go. It seems intimidating but isn't that bad after a couple go's!

Did the grain not soak up enough moisture?  I don't see even a tiny bit of condensation.  I'm a noob but could they just be too dry maybe?

Im guessing these were spore syringes as well as he states buying them. Could be a terminology issue. Whatever it is, putting it to agar is what should have been done first anyways.

You might be a noob but I think you're right sir. Not because of no condensation, though. Those grains looks like they have a lot to go yet before they are properly hydrated, particularly the popcorn.

Nothing delays leap-off of an LC more than underhydrated grain. If you still have LC left I'd start over.

Maybe avoid the popcorn altogether as it's notorious for being more difficult to hydrate and stick to that wheat (you said it's rye, but that looks like wheat to me).

Stop soaking and do this next time.

This too. Just now I reread the op a bit slower and it says it's been 3 weeks and no growth yet.  Those grains look undone and dry as a bone and OP only used 1CC of "LC", both these things in conjunction can delay leap-off a hell of a big deal, but there's the possibility it could be spore solution that has not germinated yet.
Either way, that grain looks too raw to bode well even if the solution/LC took.

I like a nice dry jar myself but to each his own. I agree though, 1cc of lC is not how to roll, hell I pour mine when I use it now and have no idea how much comes out into the jar but its a lot lol. This is where starting dry is good, if you pour a couple tablespoons worth or more in the grain just sucks up the excess.

You like grain like that? I'm not even saying it looks on the dry side from here. It just looks undone, not boiled nearly enough to be viable as spawn.

Yeah this is the hobby of to each their own, I'm an LC user and I know my spawn colonizes way faster if it's properly hydrated. Never understood the approach of getting the grain on the dry side so I can use more LC, it's cringe worthy imo. Nothing but trouble from underhydrated grain ime.

I don't mean dry as in not hydrated. I'm not sure what that stuff is op posted, and I don't know if it's done because I can't see inside it.

Possibly, I tossed the first batch bc there was too much water pooling and tried again. They have seemed to dry up a lot tho

Thanks, I’m going to cook longer and make sure they are more hydrated. Also, was using liquid culture in a syringe. No spores

You should also try pre poured or poured agar plates imo
Or get some spore prints from a sponsor

I'm not a pro by any means, but in my experience with stalled grain it could all be blamed on too little water in the grains. I have used premade lc and had great success.

I think his grain looks good. Those look like properly hydrated grains if you zoom in.
