Myc, Clone, or Grain Bit?
    #27287559 - 04/30/21 07:28 AM (3 years, 8 months ago)

:facepalm3:What would be the preferred source of myc to save as a master going into the fridge?
In my case, "preferred", is the same as what produced the spores that I used.
Keeping the original characteristics that I got from someone.

    1. Take a piece of that myc that went to spawn and make a master plate.

    2. Take a clone from a worthy cluster of preferred size. RR picks a clone,    Masters and Slaves - verum subsequentis

    3. Grab a grain from the spawn jar and isolate it.

    4. Start from the same print, swab,or syringe (God forbid!)

I'm a little confused because I've been picking up stuff like, "master the original myc"
or "take a piece of grain and put it to plate". I'm getting kinda lost...

Here's a reference to a  thread  with a paper about preservation in vials.

And the link to the paper from 1995 that documents successfully storing mycelial plugs of many species for 7 years in 2ml vials of sterile water at 5C. They also note  "Refrigeration  may  not  be  necessary. Boesewinkel (1976), Smith (1984), and Jones et al. (199I) have  maintained isolates at room temperature."

It seems that if you put your culture in water with no nutrients at all it just goes to sleep. If you try to store it in solution with nutrients, it creates metabolites that eventually self-destruct the culture.
