Microscopy, for admirers of tiny things
    #27272997 - 03/28/21 01:19 PM (1 month, 7 days ago)

Here's a nice mass of mycelium fresh outta the LC jar. Grown on just a tiny bit of blackstrap and tap water. Inoculated with a piece of agar and it appears that some motile non-pathogenic bacteria rode in on the inoculum. Not a huge deal since this was the first successful LC I've gotten some growth in, just getting my practice in before I can acquire sterile tissue. Enjoy!

There's some funny lookin swimmers that hitched a ride on the mycelium I used. the little round things floating are some cocci (bacteria) and stuff. My lc started out clear, and now two weeks later has mycelium growth but is a little cloudy. I figured it has some contamination. Neat to see!

I was wondering!

I wonder if you could check for contaminates this way rather than growing out on agar? Mod, tc?

What are the brown spots in the micrograph?

Mycelium was cultured in blackstrap molasses solution. It picked up a lot of the color of the growth media.

You might discover a contamination using a microscope, but it would not make a good go-to method for checking to see if a culture is contaminated or not.

Bacteria are so tiny compared to hyphae that they could easily be missed.

Not too mention you can only view a single drop at a time.

That certainly does look like it could be a little cluster of bacteria that are not in focus and so have taken that circular shape, or possibly yeast that are in focus.

But it almost seems like the proportions are off compared to the size of the hyphae. What magnification are you viewing from? And we're the possible bacteria all moving in one same direction or were they clearly moving independently of any current? It is helpful to discern an air bubble from a yeast or bacteria cell if they are moving in different directions or of course if they are rotating.

What microscope do you have?
