I used syringe pricks on agar with SAB. The first time I tried the growth was wispy and nice and have inoculated grain successfully. Weirdly this hasn’t but the number of bits have multiplied :shrug:
(Excuse my son’s dulcet tones)
Any ideas... Chuck?

I don’t use the queens English for three reasons:
1. I have English as a second language
2. I’m dyslexic
3. I don’t like what the queens of England represents

I’ve attached a pic of both. The scratches on the agar is where I have scraped with a syringe needle to scoop up the myc with the needle bevel into the needle shaft

Well shit, that explains a lot. I'd love to tell you your plate is clean but most of it is gone so....... shrug.

It's good to note, any amount of Bacteria, Yeast or Mold will go absolutely bananas in a sterile nutritious liquid. For this reason it is safest to only take a tiny transfer from a plate when noccing up LC. Even plates that you think are clean can sometimes having lurking little shits. Grabbing a tiny piece lowers your chances of snatching one up.

The parts of your plate I can still see look pretty good. I'll be curious to see your LC in a few days.

So over all when a plate is dense white like the OP's plate.

Is it a better idea to transfer until you get more "strandy" plates where contamination is less likely to "hide"?

So in a way less is more?

Smaller defined strandy wedges > dense white wedges?

To my VERY limited knowledge there are just some types of mushroom mycelium where this is easier said than done.

As a rule of thumb most definitely agree the smaller the transfer the better. Pick the nicest, least questionable little piece at the boundary edge and move it over. If you grab a square inch then you are bringing over whatever is under there. To microorganisms a corn kernel is like 1000 acres.

No. Not really. There are a lot of ins and outs. Hard to really explain cause it just takes some getting used to looking at dirty and clean cultures. It just clicks after a few thousand plates or so. Just kidding.... kind of. I still get confused sometimes but that's when you just snatch up what you hope is best and see what the next plate has to offer. Always steer away from anything chaotic and disorganized looking.

Pics are great and very helpful to study but nothing is like transferring and looking at your own, up close and personal. I like looking from the back side of the plate with a light behind it. Makes certain things really clear.

so I’ll stay well away from the mirror then.

Thanks Verum.... I’ve got loads plates with equally rich growth... I’ll be less of a prick with them then.

If there is further growth I can always test the culture on an agar plate, while I take smaller scratches next time round.

Gonna try my hand at lions main and other medicinals next. Will start with PFtek with those guys to get to know them personally first
