Interesting research paper I found - Cleaning up bacteria from agar plates

Apologies if this has already been posted on the forum.

After being frustrated by continuous bacterial contamination on my agar plates, I had a look at ways of cleaning this up, it always seemed to be bacteria and never mould in my case.

It seems some people opt for the antibiotic agar route but this isn't really the most sensible option. I came across this interesting paper in my search: https://journals.plos.org/plosone/article?id=10.1371/journal.pone.0224635

It seems to be a Chinese research paper(?) that is peer-reviewed and open-access, so make of that what you will :grin:

I didn't fully understand the paper but what I gathered is you place contaminated spores/hyphae on a new agar plate. You place a sterile coverslip on top of it, and after a while, clean uncontaminated hyphae will emerge from the edges of the coverslip and continue growing. Please correct me if I have this horribly wrong. There seem to be a lot of interesting diagrams and results and such. If this is legit then this could be a fantastic addition to the community and make agar work way simpler :sun:

A coverslip is essentially a tiny piece of glass used in microscopy if you are unfamiliar with them

edit: To clarify, I properly understand it now. The point is that you cut a little ditch/hole in the agar and put the contaminated shit in there, the coverslip lies on top of the hole. The myc can apparently grow out of the hole but the nasty shit stays trapped in the hole.

Nice. It's basically the same as our sammich tek. Kinda like the hot pour as well. I haven't really tried either much but thanks for linking the paper.

Nice share. I've just ordered some coverslips to attempt cleaning up some contaminated MSS with this method.  Bought both glass and plastic versions.  Might find it tricky handling the glass coverslips without contaminating them or breaking them.

I didn't have much luck with my gentamicin agar plates, I might've messed up the ratio or I bought the wrong shit. I bought some gentamicin pills aimed for aquarium treatment.  I didn't try the hot pour method, I just transferred wedges. Still get green mold and some slime bacterial stuff growing. But I finally got some clean growth transferring to plates with H2O2 added.

Anyway, I'm excited to try this new technique.

Good luck. A lot of lab equipment from lab/educational suppliers comes pre-sterilised but if not, I'd just smother it in 70% iso to be honest.

Interesting technique.
I can see some instances where this technique wouldn't work.
Such as when your contaminant is another fungi, or it is a bacterium that can penetrate agar (such as motile bacteria in a lower concentration agar)
