hello everyone! here's the situation
Some months ago I tried agar culture, turned out to be a failure. I don't know exactly why but I suspect mainly 2 things:

1st. maybe this product i'm using(blue lid) is not apropriate for fungi spore germination...

2nd. maybe the proportions(water/agar) are incorrect, the label says "suspend 22.5g in 1l of water. Since i don't have the means to calculate this porportions I just mixed a tea spoon in 1 normal water glace(cream/darked white color)...

along with the product i'm using is another I haven't tried yet
share your knowledge with me please.

-the product's label I used says it contains Casein-peptone glucose yeast extract agar for microbiology as you can read wich i think is good for fungi.

-the concentration is not too low since I've tried it high first(golden color), if it is incorrect it's because it is still high or very low(not likely).

-there is no contam problems, all very well sterilized, glovebox work. Still I've done the experience and bacterial and other organisms grow on this agar.

-the spores I'm using are ok...(for sure)




P.S. - I know this is not exactly basic stuff, but it isn't exactly advanced too, so if the responsibles for the site think this is missed placed please feel free to move the post.

I would say the container on the right is more suitable for plate mycelial growth. Does the one on the left even contain agar? 22g is no way equal to a teaspoon...even a heaped one. I'm guessing you came by these products rather than bought them right? And the last time I checked a decent set of scales could be picked up quite cheaply online at pawnshopper.net

nop but it contains peptone beef extract dextrose and it's used on water treatment analysis(sewers), this means it has the purpose of feeding some bacterea, who knows it can also feed some fungi.

I sure didn't buy it. It was given by my lab prof, and the funny thing is that he knows what's it for :P

How are you preparing the agar? like cooking wise? do u PC it? what psi? what temp? how long? if u overcook it the sugars carmelize and nothing will grow on it.

yep, it might be taht too, I sterilized it for 20 min. the prob may be the amount of solution for that time, if it caramelized is due to it's volume...

what are the signs of caramelizing? how does the cooked agar looks like? jelly, solid, semi-rigid, dark, whitish?
