I waited too long and my only viable culture has now grown out to the edges of the plate. Is there any reason not to just transfer a chunk from somewhere in the middle region or should I still go for the very edge even though it hit the wall? Any advice would be appreciated, thanks

Those slices missing reminds of these and some others I've seen. There's was a discussion about those in this thread a few days back. Consensus seems like meshed in mold that is a SOB to clean up which I can agree with IME

Either way count me as #4 for run it. Your plate looks much better than this piece of shit I have posted here

I don't see any mold...
Cube cultures do that all the time. There is so much "mold" talk thrown around here. If you have a microscope, take a look at the sectors that are thin. You can see mold. It does not ever join together. It will run in different directions and is obvious. Lots of metabolites are visible up close and the hyphae are often a different size.
In thousands of plates I have never seen mold "pizza slice" in a culture like that. I have seen many many cultures with mold and enjoy cleaning cultures very much. I also grow mold on purpose regularly (Trichoderma, Aspergillus, Rhizopus, Saccharomyces, etc)
If it looks like pizza it is just colonies separating. It is not different organisms.
Some of my best cultures came from wedges of those thin whispy pizza slices. Often if you transfer them they grow thick or more "normal" from then on. A colony will choose to invest energy to a select group of community members who are the chosen "work horses" of the whole community. This is affected by environment and habits of the culture.
Here is a burma culture that pizza's out like crazy no matter what.

Here are the fruits from that culture.

Each culture has it's habits and those habits change over time within a given culture. By providing certain sequences of events you can coax different responses out of a culture if you know it's habits and potentials. Just like raising any living thing.

Hello all, I was wondering if most of you store you're plates lid up(agar closer to ground) or lid down (agar closer to you) after takes a transfer or streaking spores? Coming from a microbio/chemistry background, I am used to storing plates lid down to minimize condensation. However, I noticed that my myc was growing very fluffy on my plates instead of a nice 2D plane. I flipped the plates and sure enough the new growth looks nice and flat. Is this a known occurance?

Yeah but look at them fruits I've also been trashing plates with missing slices. I'll probably send this ghost to grain no matter what the T3 looks like.

We also do this in the cell bio lab, but with plates that are stored in an incubator. Not sure if the incubator has temp swings that cause condensation. Storing myc plates in my kitchen seems to cause very little condensation long as they were dry when I inoculated. Even when I have plates with a lot of condensation on the lid from lazy pouring, it seems to stay on the lid pretty good and not affect myc growth

Pic one looks like it's got something meshed in. I feel that way because the growth is uneven and kinda disorganized in a few places.
Pic two just looks like mold to me

Hi been a member a long time but first time posting. I'm sure I've read about this but can't find it. What's a good recipe for trying to clean up plates? I'm thinking just lower nutrients so the cantam has less to thrive. Am I thinking right?
Thanks Much

I think you’ll find that many folks use the same recipe for growing/spawning/cleaning/germing. . .
I use 1.5% agar agar and lme. . . Germs great. . . A2G beautifully. . . Cleans up no problem. . .
