I've been having troubles identifying contamination on my agar plates. After switching from full strength grain soak agar to your lme recipe I've had only rhizo growth and it's helped me a lot being confident on whether or not things are clean. Your tek is the best thanks man! Definetially a keeper I will be doing all my agar work your way from now on!

Well I am still working on this putting coloring agents in plates and taking the time to let the condensation settle infront of the FH. Still not quite where I want it but its better than the last batch.

Im still working on how much to put into each bottle. I used 1/2 Tsp and each bottle poured out to a little over 300 ML. But this is the lightest plate also.

Not sure if this has been said elsewhere in the 21 pages of comments so forgive me if so.

Before doing this tek, I was adding my dry ingredients and water to my media bottles, microwaving until they boiled, then I would PC as normal.

Plates looked like this



I started doing this tek because i'm using 1000ml media bottles now and cannot fit those in the microwave plus using a saucepan is just all around easier and more convenient.

The only thing I do differently than you is use a higher nutrient ratio (15g to 500ml water) and my media bottles are coming out of the PC looking like this...



Plates now look like this



It's not the end of the world but is certainly making it much harder to spot satellite contams.

Would love to hear any thoughts, as stated earlier I didn't read all of the comments so maybe someone else has already experienced this and found a resolution. Apologies if so

Oof, that’s really dark and a ton of sediment. I’ve never had a plate come out like that. You’re using the same brands agar agar, LME and doing a light pre-boil and stick blend/whisk before adding to the media bottle? That’s a really chunky sediment

For ~ 300ml I use roughly this much charcoal powder



Which equates to around 3-3.6g

Same agar agar. I'm using Briess instead of LD carlson LME at the moment but will switch after i've finished it.

Light pre boil and I whisk in the ingridients, making sure to stir often as it simmers for 3-4 mins.

I don't think the LME is the issue as I can show countless examples of crystal clear plates with my old method as described in my first post.

Not sure what it could be, might get a stick blender but I don't think that's the issue.

Reminds me of a grain water run. Maybe bring water to boil before adding dry ingredients and boil less than a min. I can’t really imagine it’s being over boiled but 🤷🏽‍♂️ How long are you PC for 15-20 min?

That's exactly what I do as per the tek instructions.

Bring to a light boil and then slowly whisk in the ingredients. Had the same results with sediment when I lowered the nutes back to what you suggest in the OP.

I'll try to boil for less time and see what happens.

I PC for 20 minutes.

Distilled water, yes.

Probably not the first to try this but I put a lil bend in my stainless steel straw and now my disgusting hands never have to hover over the plate. Also I found it cuts down my SAB hand movements significantly :thumbup:




No healthy plates were harmed in the making of this video :cool:

Yeah thats a lot!

In my anecdotal experience when i add boiling water straight to large amounts of dry ingredients sometimes i see more sediment,  seems like the quick temp change on the malt changes it into something w a more caramel texture that doesnt dissolve as readily.  Filtering it removes most of it

i usually first disolve the dry ingredients in a bit of cold water, then add boiling water, then microwave on low to maintain a boil for a min or so, then filter through cheesecloth. Not really necessary ofc

Yeah C10, I do that when making gravy. I mix the dry ingredients with cold water and then slowly whisk into boiling water. I should try this approach when making plates. I have way to many poured plates at the moment because I was working on CHarcoal Powder levels and so I guess we wait a month?
