AGAR FOR DUMMIES


Hello Shroomery. I have been reading here and afew books on mycology over the years. I have been having very good success with the SIMPLEST tek i could come up with using others methods combined. So i decided to share my tek to help others master agar EASILY!!

With this method you can make your agar and start doing transfers all in less than an hour!

With the $25 of ingredients you can find at most grocery stores. You can make enough agar for 200+ cups! This is enough AGAR for months! A SAB may cost a little extra but it is RECOMMENDED! 200 2oz pp5 sauce cups is about $9 at my local grocery store. With all of the rest of the ingredients you will spend about $25 and have more agar than you need.


This is for educational purposes. Only use this TEK with legal strains.

Things needed :


Agar Agar
Instant Potato Flakes
Coffee
Karo Light Corn Syrup
A small Jar to pour from
Cheap 2oz plastic PP5 Sauce Cups from grocery store.
Hydrogen Peroxide 3%
Optional, 10ml syringe with needle.
70% ISO Alcohol
A SAB
Gloves
Food Coloring



This is the prep.


ALSO i should add that i do my pour process in a SAB!! ( UN-Modified SAB )

I prep my SAB, wiping it and the area around down completely with a paper towel soaked in 70% iso alcohol. I also use gloves in my SAB.

Before getting started, i fill one of the plastic sauce cups with hydrogen peroxide 3%. Another cup with 70% ISO alcohol. Before putting peroxide into the syringe, i suck up a sauce cup of 70% ISO alcohol until full, give it a shake and then squirt out the ISO alcohol into a sink.( Now the syringe should be pretty clean!) Then i suck peroxide into the 10ml syringe until it is full. ( If you dont have a syringe. Just fill a 2oz sauce cup with peroxide, put the lid on and set it aside until the step it is required to be added into the mixture. )

I also clean my small jar (holds about 500ml,so it works PERFECT) i wash it in the sink with dish soap. Then i dry it and pour a splash of alcohol into it and swish it all around inside of the jar. Then i pour it out and allow it to dry upside down on a clean cloth on the counter. After dry i place the lid on the jar until i am ready to use it.

After i do the prep work, its onto the 15-20 minutes of cooking and im DONE!

This is the cooking process.


Make a cup of coffee for step 3 beforehand!


Step 1) Put pot large enough to hold 500ml water on stove. Start burner to warm up pot. Add 500ml water to microwave safe bowl/jar. (I use a small Jar.) Microwave for 2 minutes. Then slowly pour the water into the pot on the stove. ( SLOWLY & CAREFULLY!! STEAM CAN BURN YOU!!) Doing this with microwave will make the water boil in the pot in less than 5 minutes.


Step 2) After water is boiling add 1 to 2 teaspoon of potato flake to the water and stir for about 2 minutes. Add 1 drop of blue or green food coloring.

Step 3) Pour just a small amount of coffee out of the cup into the pot of boiling water. Probably about 100ml of coffee. ( I just eyeball it. About a shot glass worth of coffee.)

Step 4) Add 1 teaspoon of Karo Light Corn Syrup to the boiling water and stir it in well.

Step 5) add 1 teaspoon of Agar Agar into the boiling mixture. Stir in as you pour in the agar. Try to pour the agar powder in slowly. If you dump the whole teaspoon of agar powder into the pot it starts to clump up fast. ( 1 teaspoon agar = Soft Agar / 2 teaspoon agar = Thick Agar )

Step 6) Mix in the agar until it is fully dissolved and the consistency of molten glass. Should only take about 3 minutes at a rolling boil.

Step 7) Turn off burner and in the sink, CAREFULLY pour the agar into your small jar. I do this immediately after it comes off the stove. Carefully place the lid onto the jar to keep out contams.

Step 8) After about 20 minutes , touch the jar. If you can hold it for about 10 seconds it should be cool enough to proceed to the pouring stage.

Step 9) Wipe everything down with a paper towel soaked in alcohol before putting it into the SAB. EVERYTHING! Your jar with fresh made molten agar! Your stack of plastic sauce cups! ( Dont wipe sauce cups one by one, just the stack of cups and lids. ) If it goes into the SAB, it gets WIPED down thoroughly!!!

I even wipe down my gloves with 70% ISO alcohol EACH TIME they re-enter the SAB!

Step 10) Open your jar of molten agar in the SAB, pour in the 2oz sauce cup or squirt in the 10ml syringe of hydrogen peroxide into the molten agar. Put the jar lid back on tight. Give the jar a good swish to fully mix the peroxide into the agar mixture.

Step 11) Pour agar into the bottom of each cup. As you pour into each cup. Place the lid immediately after the pour. Be very careful not to reach over the sauce cup while putting on the lid. Its best to have everything set up in the SAB so that you can pour each cup and then grab the lid and place it on without ever reaching over the sauce cup. Pour quickly and carefully. After adding the peroxide the agar will start setting very fast! 

Step 12) FINISHED! You can allow it to cool and set now. Afterwards you can store in the refrigerator for afew weeks until ready to use. I however prefer to use them immediately. Because it seems that in doing so,while the peroxide is still active in the agar it helps keep my transfers clean! Because  the peroxide should kill any bacterial contams on contact if the rest of your procedure was sterile! 

IF YOU WANT TO USE SPORES WITH THIS AGAR. YOU MUST WAIT AT LEAST 2 DAYS BEFORE PUTTING SPORES TO THE AGAR. BY THIS TIME THE PEROXIDE SHOULD HAVE CONVERTED INTO WATER. PEROXIDE WILL KILL SPORES BUT NOT MYCELIUM.

Day 10:

This is the cup on day 10! All of the cups are pinning in vitro!
This cup has two little pins with red caps!


TUBs from rice inoculated with this agar TEK. Spawned 2-4-21. 4 days old 40% colonized. 2:1 spawn to coir


Day 10 Tub :
I added the smaller tubs to a larger shoebox to help with fruiting conditions & get a better flush. 

My phone camera kinda sucks. But this one you can see pinning plus afew fattys coming in.

The yellow is from the lighting in my room . Its all white to the eye. Not a contam.

Will be ready to harvest the first flush in  the next few days.



Day 16. HARVEST DAY!!!

With this AGAR method. You can go from agar to mushrooms in about 4 weeks!



This isnt the best/recommended way of making agar. But it works if you need it!

How can boiling for five minutes get rid of the bacteria? With my low-psi pressure cooker I get contamination even if I PC under twenty minutes. Are you sure this works?

Positive!!! Before i did the little test, where i put a drop from my contam MSS. I left the agar in a drawer for a week. There was no growth on the agar whatsoever . It wasnt until about 3 days after adding the spore solution that the yellow bacteria was very present. I think that peroxiding the agar at the end before i pour has alot to do with my success. Also the 2 minutes in the microwave before being poured into the pot probably eliminated alot of the bacteria as well. I may add some pictures of my cups soon.


Edit : My first agar attempt, i didnt use peroxide at all. And i steam sterilized the jar of agar for 90 minutes. The agar had bacterial growth within 3 days. PDA

My second attempt,i steam sterilized then i used peroxide at the end before i poured into cups in a SAB. These dishes did well, but the rice i transferred to them from jars was still slightly bacterial. So as the mycelium grew so did the bacteria. PDA

Third attempt was coffee agar NOT steam sterilized, then peroxided before the pour. Took transfers from the other agar dishes, i believe i mixed this agar too thick and it didnt take very well. But there was no other growth in the cups.  Coffee Agar

Fourth attempt, I made Potato Dextrose Coffee Agar with the TEK above. No steam sterilization. Just peroxide added right before the pour. Then after all jars were poured and not even fully set yet. I took transfers of colonized rice from my jars and dropped them right into the center of the agar. They sank in slightly since it was still half set. It has been about 3 days and the mycelium is starting to take the agar! Its starting to branch out! And i see no yellow around the mycelium like i noticed with a previous transfer from the bacterial cup/dish.

I should also add that these some of these cups have been shelved for afew weeks with no growth other than mycelium and the yellow bacteria that was transferred in by me. So im very confident that these new cups ive just made will do great. As i stated the mycelium is already starting to shoot out strands of mycelium!

Here is one that is showing growth well!

Transfered from Brown Rice Jar on 2-4-21

Jar had a yellow bacteria from the MSS that was used from the beginning.
The jar was innoc. with agar. Shouldnt have innoc. jar with bacterial agar. But it went tormentous and took the whole jar.  1 of 3 jars didnt colonize well due to bacteria. The other 2 ate the rice up bacteria and all. So i then took transfers from this rice to agar on 2-4-21 . And its looking like its doing well, and that it kicked that bacteria's butt!



 
Notice the bubbles, indicating that peroxide was killing bacteria on the rice as the agar hardened. But it only had bubbles around the rice. May need afew transfers to be 100% clean. Time will tell. But these cups all look great so far!

By now there would be noticeable bacteria growing on the surface if my process was bunk . As my first agar attempt had some clear slimy stuff streaking across the surface within 3 days of pouring it. and i steam sterilized that batch! BUT i didnt use peroxide that time. I feel like the peroxide makes a BIG difference in the end.

It WORKS.

As long as you have a good sterile technique and use peroxide in the end before the pour. You should be GOOD to GO!!

I live in a dirty/dusty old house. So for my agar to be this clean, is a god send. LMFAO.

Very enthusiastic write up.

I would not use bleach (Clorox wipes) around H202 for an "anyone can do it" tek. It could possibly explode if they contact, but it WILL produce toxic chlorine gas if they contact.

That forms hydrochloride acid in your lungs and will do permanent damage.

I'd replace the bleach with strong soapy water in a spray bottle. That kills contams also. Or better yet, use your ISO. You already have it and it's better than bleach or soap.

As for the value of the tek, my guess is you've been very lucky and this isn't going to be repeatable for long.

A lot of people will probably jump on here to bash it if this thread gets very long.

There's a reason people here are religious about the almighty pressure cooker - It consistently works.

I have a recommendation. You don't say how big a sauce cup is. How many ml, or ounces, in case people can't find that and need to substitute. I can't picture in my mind what you're talking about.

I would edit your post to be more precise with this.

I'm also really not clear how much 3% H2O2 solution is going into how much agar mix. Is it 10 ml into 500 ml water plus 100 ml coffee in a container that can hold 500 ml? If so it's running over.

I'd for sure replace "water" with "distilled water." Less chance of contams. Tap water can be nasty in some places. Esp since you live in an old dirty house (why not clean it?), your pipes may be growing stuff in them, and even some city water has a little live bacteria.

Thank you for your critique. Man it really works! ill add more pics of the growth in a few days to show progress. Im still new to agar. But i have made it this way 3 times now with no contams in the agar after 7+ days, other than the yellow bacteria that is piggybacking some of the mycelium. But these new transfers look healthy! I will add the cup size and amount of peroxide to the post.

If clorox wipes have bleach, you are putting people in danger with that part.

Edit: looks like they don't have bleach (unless they have different formulations), but they have
Alkyl C12 – 14 Dimethylethylbenzyl Ammonium Chloride

but the fact that it's a cleaner with "-Chloride" would make me not gamble that it won't form chlorine with H202.

Any chemists here know?

Sorry for the double post, but...I just read this part. 7 days??

Some contams can take longer than a week. Some are faster than mycelium but not all from what I've read from others on here.

I'd wait to at least get to the point of inoculating grain with the agar before writing it up. Really, I'd wait until it's all the way to shrooms, then repeat all of it a few times to really be a useful tek.

One of the factors that makes something science is that it's repeatable. This site is basically a bunch of citizen scientists...at least the people writing teks that get shared.

But I really do love the enthusiasm.

Uncle Ben's has onion and garlic. Those might slow things down. Plus yeast and salt. Bad news. And that rice is processed, right? That strips nutrients. Use regular brown rice anywhere you use rice. Or better yet, rye or wheat berries.

Did you do the plate in your avatar? It looks great, and I don't guess it was done this way. If that's really yours, I'd stick to whatever you're doing. Though I'd take it to the next step before it gets all the way to the edge.

Thank you for bringing this to my attention. I use ISO sometimes, but lately ive been using the disinfectant wipes. I wipe everything with a wipe and place it in the sab. After afew minutes it all dries off . Actually ive wiped the needle down with a clorox wipe and then squirted it into the agar jar!!! oops. I was under the impression that bleach was better than ISO. But if ISO is just as good i may substitute it in my TEK.

No the picture wasnt mine :frown: . Im just enjoying agar .

I have some dishes from 1-17-21. No growth. Its still clear agar with a flashlight. One has some slight yellow bacteria leaching out from around a transfer. The agar that that transfer came from was infected. I think it would have grown better. But i used like 2 and a half teaspoons of agar and it came out way too thick.

Edit : Just looked at dish in picture day 3. It has grown 1cm more in 7 hours! I think this is the sweet spot. Its loving this agar. The coffee and potato is probably pretty tasty to the mycelium after eating up all that nasty brown rice. lol

I would seriously use a different photo. It looks like you're claiming it's yours.

Everyone on here who has an avatar that's not the moon or a wizard or their cat, people who have an avatar that's actually of growing stuff, they actually grew it, and it's a point of pride.

I don't think it's a written rule but I read this site a lot every day and that's the feeling I get.

Like my avatar is a pic I took through my microscope of spores from shrooms I grew.

You having someone else's kick-ass prefect Petri pic as an avatar kind of comes off like "stolen valor."

Again, I really do love your enthusiasm, you remind me of a young me (or even me when I was brand new on this site).

But a noob telling everyone "do it my way" when you haven't even finished your first run with the new tek isn't going to make friends on here.

I'm have some notes for a tek, but it's at least a year away from sharing.

I would highly recommend that you at least stop replying to every noob with a cut and paste reply saying stuff like "This doesn't have to be hard, read my tek!" Your "tek" isn't even done. It's a work in progress, you haven't even proven your premise yet.

Go read the replies.  People who've been here a while are replying to you with "WTF?" type things.

I hope you stay around here a long time, you're full of energy and that's great. But you're doing it wrong, little brother.

Solid critques from nektar.

Nice write up but I'd consider moving this to your journal untill you've gone spore to fruit with this tek.

You're pushing it very hard on the getting started forum without any real results to back it up. Run a grow, write a log, and use that as the basis for the tek.

I'm also a little curious as to how this is easier than normal agar?

Im not knocking anyone elses TEKs. Theres tons of ways of doing things. One thing ive noticed, is everyone does things a tad differently. But if it works for you, run with it! Im just sharing my little experience with the community, hopefully we can all learn from it and expand on it. Thats what these forums are all about right? Sharing information for the future of mycology.


These cups look amazing . After doing afew agar runs, and seeing mycelium preform on brown rice in jars. IM VERY confident with this agar. I have a tub from this agar TEK from agar made 4 weeks ago that was done with brown rice steam sterilized. It is taking the coir well.

I hate to say it but by the sounds of your approach you're just rolling the dice.

Steaming grains for 45 mins isn't going to sterlise them.

Have you got some photos of the tub?

People usually do uncompleted teks on their journal, not on "getting started." Your journal is kind of a sand box.

There is so much wrong about what you're doing on here. Just go read the replies to your comments every place you've posted your link. You can find them by going to "THREADS" near the top right of the page.

Well, it takes me about 25 minutes to finish making the agar. By 45 minutes im in the SAB pouring into the cups. By 60 minutes, im transferring mycelium into the cups. With peroxide still active in the agar, i feel this is beneficial to mycelium, especially mycelium that has a bacteria piggybacking it. So i feel its easier and faster, and im a rookie. Im not asking for a cookie. Just sharing my baking trick. Like substituting butter with oil. Its not really advised, but it works . LOL

Why the rush?

Feelings aren't the best grounds for posting a tek. Photos of harvests are.

Don't get me wrong, I want you to succeed.

But you have to appreciate that if you post a tek that flies in the face of convention, with zero results to back it up, you're going to have trouble convincing people that it's of any value.

Posting it in the getting started forum is unfair. Take it over to mushcult and have it looked over by the seasoned grower. Posters here don't know better. It's a place to suggest tried and tested teks that have years of consistent results to back them up.

Ideally keep shtum until you've got a couple of decent harvests under you belt.

At the moment, it's just a fun idea, not a tek.
