I think grain would have to be pretty expensive before cost effectiveness becomes a real issue. I know many of us are cheap bastards but this is a cheap hobby relatively. If I converted my automotive hobby to mushroom goods I'd have a lab the size of a home Depot.

1 looks suspect to me, bit too thick on the growing edge.

Maybe some dodgy patches on 2 and 3. This is about a week after shake no.2 though so the growth is spread out. The other side's are pretty much covered.

What do the trained eyes say?

After running a bunch of rye and following teks which are great guidelines.

I realized that grain can vary from batch to batch (ex.sliced rye grains- how dirty etc.) so preparation should be adjusted accordingly.

This last batch of rye I washed the rye with a couple of drops of soap (thx Smartattack) soaked for 4 hours and boiled for 10 minutes until some rye burst but more importantly I stopped the heat when the majority were a little past al dente (thx p9).

While the rye/ grains are steam evaporating they are still "cooking" so by the time they are dry they should be pretty well hydrated.

Just rambling here but I hope this helps someone out. I'm a noob so please take this info accordingly.

I was having decent results with oats. I'd say 80% success. Rarely contams that I could see - usually if there was an issue it would show up as a stalled or very slow jar with no other obvious symptoms.

Working with pan cyans instead of cubes now. I was having the same success rate on oats with pans as I was having with cubes.  I decided to give WBS, and then Millet a try just to see if those would work better for me than oats, and I haven't had a single successful jar with those new grain types.

The three jars on the left are 6 days old, pure millet, PC'd 4 hours following a millet tek. The three jars on the right are 3 weeks old. They are still moving along but painfully slow and the mycelium looks too thick and creamy to me. All were knocked up from clean-looking agar cultures.

These are G2G transfers where I transferred some oats into these jars which are 80% WBS and 20% rye. These jars are 3 weeks old as well. Just not moving at all.

Any ideas? I feel like my sterile technique is pretty good. Huge sab, sprayed with soapy water, floor sprayed with a sanitizer, wire rack over the floor. I put foil over my jar lids when I PC them and leave the foil on until I put them in the sab. Everything gets wiped with ISO. Instruments torched. I use unmod plastic lids and also metal ring lides with SFDs and 2x 1/4" holes punched in the lids. I wipe them down with ISO and only hinge one side of the lid up an inch to drop the agar in with my sterilized xacto knife. I keep hands from going above anything and continuously wipe my gloved hands with ISO. I'm very new to G2G transfer - all my experience has been with agar. To do the G2G I was sterilizing metal spoons wrapped in foil in a 400 degree oven for 30 mins or so, cooling, then unwrapping the foil in the SAB and using the spoon to spoon out a bit of grain from the master jar into the destination jars. If these jars are bacterial, I have no idea how the bacteria is getting in them.

Here are two examples of plates I would consider worthy for transfer to grain:

Well, 2 x 1/4" holes seems excessive, you may be drying out your grains which may be contributing to the slow colonization; but yeah some of that mycelium looks pretty thick along the leading edge.

Bacteria is user error: so either you failed to purge your pc correctly/failed to cycle correctly, or introduced bacteria  in some other way.

In the top pic the first 3 jars look suspect, the rest seem to be ok. Hard to tell.

Your mixed jars look super weird, some of your oats look dried out, very light in colour.

Thanks for the reply. I do feel like my grains have been getting a bit dry in the jars and think that could be causing some of my stalling issues. I'll switch to 1x 1/4" hole on the metal lids, but the odd thing is, even with unmod plastic lids screwed on tightly, I still feel like those are drying a bit too. When I was using oats, I was getting them really dry after boiling (per what I had read in some teks). But I think I was getting them too dry. Then when I switched to WBS/Millet, I tried to do the same thing and think they were WAY too dry. For the millet-only jars in my pic (the ones with the thick white mycelium), I actually followed the millet grain prep tek in your sig, which had me loading the millet into the jars fairly wet, but it all absorbed after PCing and the grains look great now - plump, and the jar doesn't look wet. And I did the full 4 hour PC per your tek so I'm sure it was sterilized properly. I vented for 10 mins but per your comment, I'll start doing 15 just to play it safe.

Agree on the mixed jars. I those are too dry. They were the ones where I did cold soak and dried them out probably too much.

Thanks again for the help.

Maybe this will help.

Left jar cracks millet tek came out almost mushy where I could not break up some millet clumps after the PC cycle. Clumping was most likely caused from my jars being in the water (tek says to get jar above water)

Picture was taken about 16hrs after a shake.

Right jar p9. My mistake was most likely from drying the millet a tad too much before PC cycle. The mycelium has just slowly expanded In a death star fashion which I just shook.

As you can see the left jar has colonized ok after the shake at 30% even through all the clumpy wet millet.

Right jar I just shook cause it's been about a week. We'll see if it helps a bit colonization.

I guess the question would be how long did you soak the millet and how long did you set aside to dry hindsight?

I soaked for 17 and iirc 30 minutes dry time

Next time I will soak longer and I'm gonna dry the millet for 15 minutes.

My soaks are almost always 18 hours, sometimes ~20'ish. I let drain in my sink for approximately 20 mins while I prep my work space, turn my hood on etc for my days culturing work.

I load wet to the touch, it clings to my hands and to the glass. Make sure that your millet isn't too dirty, I recently ran a particularly dirty batch of millet in the same manner as I do with clean stuff, meaning that I didn't rinse them clean....all unwashed/dirty grains contaminated.

If you have filthy grain I suggest a good rinsing until rinse water turns clear.

Yeah, sorry I soaked longer than I said. I followed your tek to a T so thinking back, it was 12 hours I believe.

Atomsplit, in prior attempts I got the grains dry to the touch before loading into the PC (actually spreading them out on a table on a sheet and putting a fan on them like I do for oats), but per P9hu7's tek, the problem with that is that you don't get full hydration of the grain during the cold soak so you need to load them in wet. I drained exactly 20 mins and loaded them in to jars from there. They were definitely wet on the outside but it all absorbed after I took the jars out of the PC and let them cool and settle.

And yes, I do clean the grains VERY well. I didn't do that with oats but all the WBS teks instructed washing and I just carried that over to millet because the millet I have is about as dirty as WBS. Usually this means about 5 rinses in the pan.

Hey Shroomery. Just wanted to verify if I have a contam riding along my grain jars.

Looks like mold but since it's near the agar/mycelium I have a little hope it's not.

Thanks in advance!

:frown: thanks p9. Is this most likely from my culture or sterile technique?

It's growing on the bottom of your culture, mold won't survive the pc so it was post pc cycle; if it was a faulty filter it would be growing on top, maybe it was on top and has outrun your mycelium, but it's likely that it was poor technique: deposited via turbulence, fell from scalpel, hovering your hand, etc.

Mark your jar lid so that if it happens again you'll know it was the lid.
