Made these the other day... maybe I confused the agar and lme... I doubt it though. Bacterial maybe? Haven’t been inoculated yet

My malt extract broth Is always cloudy until the cultures starts chowing down.

After about eight days it'll get crystal clear.

Another tip I'd like to give is to not give up on a cloudy LC.  Cloudy LCs are normal if one uses malt even from day one because the malt has solids form from the heat (no, I don't know the chemistry behind this).  But if the cloudiness doesn't resolve right away (say a few days), it still might be fine if you see mycelium blobs forming.  The reason is that mycelium can kill very minute amounts of bacteria.  In the end, a good LC is not cloudy, though, since the mycelium will sequester all the malt sugar fragments and kill all bacteria that may be in there.  I have had cloudy LCs work that I am positive had either bacteria or yeast growing in them (from a smell test), but the chances of stalling the bulk substrate (or spawn if you go that route) is a lot higher with a cloudy LC.

I’ve been wondering why my lc stalls sometimes in my grains and this totally answers it. Thank you

It’s more gelatinous almost.. maybe I messed up. Here’s a pic if I spin the media bottle the solution stops spinnin within a second or 2. After shake air bubbles remain In solution. Like a heavy mineral oil

I’m just going to assume I fckd up and grabbed the wrong powder.

Yeah, I'm putting my money on you mixing up the powders and putting agar in there instead of LME. That gooey sheen is a giveaway.

Without wanting to derail this thread, Blue Helix's substrate that he sterilises and then inoculates  with LC — the grains are added dry to the substrate — they aren't boiled or soaked at all first?

Presumably they hydrate in the sterilising cycle?

Ok, this was def agar in there but now that gives me an idea.. probably been done before and since I don’t see it already being done I assume it doesn’t work well but Imma try anyway.

I did 350ml distilled water, .75g agar agar, and what I assume was probably close to .5-.75g lme.

My hopes is that the solution will be viscous just like above where I can inoculate like a LC but pour like a slurry once inoculated.

That is a really cool idea, D3monic. I can't wait to see the results.

There is no grain prep at all with this method. It's just mixed in with the sub and everything is
hydrated all at once.

Got it, thanks. I think I’m going to try this a substrate in bottles and inoculate with LC.

Yeah the agar in the lc is called ssli, like most cult stuff it takes a little dialing in based on your brand but it works great.

I think I liked ultra soft plates better though.

Yeah, it's sometimes called direct inoculation. You can search mary fairchild's grows or
Blue Helix to find it. Or you can click the "Cultivating" link in my sig. I use the method as well.

Blue Helix touches on it in this thread in a few posts:

https://www.shroomery.org/forums/showflat.php/Number/26977601#26977601

But also in stand alone threads:

I searched the thread but I didn't really come up with anything definitive, so I'll ask:  what are the cues you want to look for when deciding if a LC is ready to test out?  Is it just clarity and colonization?

I have a pasta jar half-full of undiluted grain water that I PC'ed and dropped some myc from a clean plate into.  I swirl it a few times a day (there's a spring from an old cocktail shaker in there to break up the blob) and now after a couple of hours it settles out to something that looks very clear to me (no visible sediment).  The liquid is about 50% colonized after 10 days.  Should I let it colonize further, or is that enough to test it on agar?

(I know a picture would help but I'm having trouble getting a clear enough one.)
