Have been slacking on cult stuff, so I had to get my ass in gear today...
Took transfers with the last of my red plates from the PFC clone, B+ clone, and Pink Buffalo that are all looking great. These are all going to grain and possibly LC tomorrow as well.
Took a couple transfers for my LAGM project too.
Made a bunch of agar bottles so I have enough pre-sterilized to store + a couple jars of broth to play around with.
Just poured some new plates, and I'm about to get busy on some grain before I conk out. :stoned:

That's the spirit!  Just forced myself to go harvest wild grass stalks for muda bottles in -5f after working like 12hrs on and off and it was definitely fighting gravity to do it lol

Heart = Can a cap be too small to print? if my first flush is breaking veils when caps are about the size of a 10p (a quarter I think?!) if it even worth it? should I wait for the 2nd flush and hope for bigger?

do you guys always keep a MS culture around on agar? Just to quickly go back to the diversity? Or if you were going back to MS would you just start from fresh spores every time?

I have some projects on agar pretty much all MS and the grows will be hopefully have clones taken, but once I move to grain or LC I find myself still making xfers on ms plates when they get close to the edge. generating more and more ms plates lol 

Its like I keep up with agar work just to keep plates going and from being overgrown, maybe I should look into slanting or something to hit the pause button on plates while I fruit out the culture? :shrug:

I thought about using pastyplates or the holy grail containers poured thick to use as short-term slants, but if I am thinking 6 months or less, could I just double wrap a plate, ziplock it and toss it in the fridge?

Once clones are taken and I have clean clone myc, I would like to forget about the MS stuff until I see the clone sucks or I want to go back in the gene pool for something.

Cam, what I plan on doing this time around is doing 3 shoeboxes at a time with one quart jar grain spawn each. Each quart takes 4 plates, or in my case baby food jars. One ms agar, one T1, one T2, and one with oats for the T1 to begin colonizing. I will pitch the Ms and T1 as soon as I get a wedge on grain, and either save the 3rd agar for the T2 or a clone later depending on how fast it colonized. Then I use the grain to G2G a full mycoquart and all the inoculation points is as good as lc in my book. This process allows me to try different genetics on a monthly basis while limiting waste and it only takes a few hours a week.

This is also good for trying to cross strains because you can do two boxes that are control specimens, and one that is the potential cross, to see if any of your fruits are new phenotypes

I just had a bunch of spore plates and T1's that grew out and stuck em in the fridge. If I don't like what my T2's do, I'll bust out the T1's and if I'm still bit satisfied, I'll go back to the germ plate.

My ultimate goal is to have a working clone for each of the varieties I plan on running.
A slant, and a working plate I make LC from. I'd like to see how far the LC can go too with LC - LC inoculations
To bulk out the chosen 4, the path I planned on was: Agar->LC, LC->Presealed bags via syringe in SAB, 1 bag = 1 mono

Right now we have MS cultures of:
Rustywhyte
Penis Envy
Texas Yellow Cap

A clone of:
APE - in LC now and will be going to grain this weekend I hope

I would love to have a clone of all 4 varieties to rotate out. I'll have a white/reggy of the classic cubes and the same duo for the stronger cubes 

Those 4 will take up real estate in my 8 monos (I need 8 more), and I can run small shoebox grows of others for prints to build my library for trades and start giving back in giveaways

So much is a matter of preference its hard lol, this is where I really wish my girl was into it so she can help me "decorate" in a sense when it comes to cult and what/how I'm growing

Cam, I'm exactly the same with plates. I started with 20 from a single MS syringe and did like 8 transfers from the best and then 4 more from each and then 4 more from each and then 4 more. I'm fruiting the 4th gen now but still continuing to transfer even though I've cloned a 4th gen fruit.

What I'll probably do is get rid of everything before the clone either by chucking it in spare tubs whilst I wait to get the clone to grain or by just throwing it away. All good practice for me as a rookie but not how I'll operate going forward.

Thinking I will clone this big guy wearing the quarter hat. Nothing great, but biggest\baddest of this flush and I'm in a hurry to get some choice genetics again. I always took clone material immediately after harvest but I kinda recall a discussion a while back about waiting a day or two before cloning or in some cases even partially dehydrating first.
Thoughts? Anyone have any favored procedures when cloning fruits?
Not looking for a how to; more just personal preference or insight. A discussion, if you will, about general cultivation practices. (Haha, see what I did there...)

I'm harvesting today too :smile: just took another 140g, that's 210g now in total from 4 cakes. 6 still finishing flush 1 but I don't expect them to produce as much in weight.

Would you wait and dunk them all together? I've put the harvested ones back in the chamber for now

I'm trying to wrap my head around why its known that "less is more" when adding inoculant. And that the moisture content gets thrown off if too much is added to larger bags... but if you scale up 5cc (quart jar) to 30cc (4q Myco bag)... wouldn't that have the same effect on the moisture content?

I usually just dunked and put them back into fruiting as they became ready. even if I start all the cakes at the same time it seems like I always get some discrepancy between them before it's over.
This time however, I think I may be more excited for the dunk than the harvest. These are the test cakes for my vacuum hydration experiment, and I'm very excited to see how it works on the later flushes.
I kind of f***** up because I honestly didn't think it would work this well and I didn't track individual cakes after they went into the fruiting chamber so data may be a little bit limited, but still very anxious to see how this plays out. The results so far I suggest that is little as 10 to 30 seconds in the vacuum chamber is enough to fully hydrate the cakes on the first dunk. I know an overnight or 24-hour dunk isn't that long or taxing to do, but if I can throw these in a jar, vacuum for 30 seconds, and put them right back into fruiting then less time is spent waiting and potentially more fruiting before going green.

also clockwork, just wanted to say that you were kicking my ass as far as weight goes. Only harvested three cakes so far but those three only gave me 120 g

yeah lol that was the thought, but didn't know if it was worth keeping that T1. Cause if I am running the clone all the time it should be nothing to get fresh spores that will pry germ really fast for going back to MS

Check it,Its no secret an even surface thats correctly packed helps promote an even pin set.

This tub was weighted with a grate on top for the first 60 hours of colonization. Then the grate and weight was removed.
Its kinda hard to see cause they are albinos but everything that looks like a lil knot is actually already a pin.
Seems the scoring and weight helped the pin set for this MS tub.
