Hey guys I’m new here. I bought a grow kit and this is my first time with a buddy of mine. We got some instructions from the guy we bought the grow kit from but here’s the start. Taking more pics in a hour or so. any advice would be helpful.
When to mist? Fruiting? Infections?

Doctor Mario's ROARing Apes Grow Log 1/19/2021Cut a swab into 4 pieces and loosely buried it into a plate. Great things to come. 1/31/2021Buried the other swab tonight. The last plate didn't do anything except for grow some pretty pink bacteria in one spot.
Maybe the other swab will do somethin for me.2/3/2021 The 2nd swab germinated somethin. Still not sure if it's myc. Looks kinda funny. Took transfers anyway.

Two plates for the APE swab: one contam and one a light circle of growth?? Next time I use a swab going for more plates for that swab.
14 January, 2021:Well... Progress is progress, no matter how small... 20 January, 2021:From five plates with four wedges ea. Transferred two plates, 4 wedges ea. of SW APE for T2. Well I think I'm learning. I'm realizing it's a numbers game. I've started putting four wedges per plate. So last one was 20 wedges, so it's like 20 plates! But only five. And out of that I managed to go T2, two plates for 8 chances at a winner! Also I've started looking at the growth with a strong magnifying glass from the back as the growth shows better. Learning to look for straighter, better organized. 21 January, 2021:Ok, swabbed four plates, including the swab tip last one. So, second try. Nothing to see yet... 28 January, 2021:So has anyone else looked at their swab doing nothing in the plate and ripped it out and shredded it to three other plates? 7 days old And just look at the contam coming from behind where I held the swab. Then I had a second thought that I should have put a drop of water on each. Anyway just a noob larnin 'bout swabbin! (ya really lumpy green agar, more larnin! ) 10g 2% Agar, 7.5g 1.5% LME, 500ml 12/30 pretty hard Split 2 jars grn & blu/red 🍒 325ml 28 January, 2021:

Eh, it's only been 4 days and burying swabs seems to always be 3-5 days slower than loop streaks. I'll give it another 5 days and if I don't see anything, I'll put a drop of sterilized water over each part of the buried swab and see if that makes em pop.

Rolled one swab to two PastyPlates. Temps will be in the mid to upper 60s for germination. I will keep this thread updated once I have growth.

I took Roar’s advice and tossed a space heater in my germination room to get my temps up. I noticed a tiny white spot on one of my plates yesterday which if it’s not a contam, would be 3 days to germinate. It’s slightly off center of where I swabbed.
Now that some of the condensation has cleared up in my plates, the exact center of the plate where I first made contact with the swab is starting to look like germination may be starting there. I will know for sure by the end of the weekend.
2/1/2021
The small dot on one of my plates hasn’t changed at all since my last post. No growth, no color change, no nothing. Still no idea if it’s a contam or spore germination. I will monitor. Nothing on the other plate.
In the meantime, I took the remaining swab and streaked a few more plates and buried the swab in one of them.
Hoping for some success on this round.

We have liftoff! The only growth I see is in the plate where I buried the swab.

You might wanna find a warmer spot to set them until you get some germination... APE are notoriously slow and a lot of people have some difficulty getting germination from swabs in general... just my 2 cents tho... hopefully you get something to pop...

Thanks for the swabs, roar!
I put one of the swab tips in the bottom of measuring cup and repeatedly sprayed it with 30ml of distilled water from a syringe, an extreme bastardization of faht's tek. After a few cycles, I put a drop in an inoculation loop and streaked a pair of plates. Then I cut the tip in half and streaked another pair of plates. All 4 plates were 1% agar, 1.5% LME. I'll hold onto the other swab, in case this doesn't work out.
I'm also tracking this grow in LAGM2021.

Just saw this pop up. I’m jumping in on this, my swabs are from Roar through a trade. Took some effort to germ but I have 8 colonizing grain jars now. All have been shook and should be ready for spawn in the next couple weeks depending.

Recovery on 23JAN
I shook again yesterday to speed things up a bit. Recovery looks good, no pic yet.
All jars were noc’d with small agar wedges and took forever to colonize, I would recommend using larger or more pieces for faster colonization.

Was frustrated at germing plates with swabs over the past couple months and got tired of just burying the whole swab in the plate so I started to use a fresh sterile 28g needle to rip a tuft of spore laden cotton off the swab which I then drag down into the middle of a thickly poured plate so it's sammiched around with agar like the tip of a buried swab would get.
Works like a hot damn.

I didn’t document a timeline on germination
I am having good germ luck with an 8g lme 8g agar to 400ml distilled water recipe in combination with dipping the swab in sterile water first. Another good tek with swabs has been using this brf puck recipe and taking a few strands off the swab and shoving it in the puck.

1/31/2021 Buried the other swab tonight. The last plate didn't do anything except for grow some pretty pink bacteria in one spot.
Maybe the other swab will do somethin for me.

The small dot on one of my plates hasn’t changed at all since my last post. No growth, no color change, no nothing. Still no idea if it’s a contam or spore germination. I will monitor. Nothing on the other plate.
In the meantime, I took the remaining swab and streaked a few more plates and buried the swab in one of them.
Hoping for some success on this round.

I got my swabs on agar 3-4 days ago, I had to medivac one myself. I think I'm starting to see (good) germination now though

I've been thinking about this a lot lately, and with how many people are expressing issues with swabs and germination, I wanna try swabbing dehydrated fruits to see if that might help eliminate some of the contamination issues... its just the start of an idea so don't burn me at the stake here... lol

I had some bacterial issues with my swabs, but it absolutely could be user error. My cleanest swabs have all come from fruits I grow in bags. Open/harvest/swab in front of the hood and then let 'em dry for an hour or so with the hood running. Might be overkill, but it works real well for me.

My roarkell APE jars are going to be spawned sometime this week to 17qt tubs. One jar looks a little off from the rest... will update with pics later in the week. Doing my biggest grain jar run to date starting tonight. I’ve never had 30 grain jars going at once... dippin them toes. Thanks LAGM2021 lol

Plz let us know how the swabbing of dried fruits goes but I have to say that dehydrator air blowing all over the gills can’t be better than swabbing and waiting a week or more to dry out the bacteria if there’s any. Do you guys rinse your swabs in sterile water before streaking? Those two things seem to help me when I make spore solution from my swabs

despite knowing I probably should, I'm always too lazy to get sterile water into a syringe but have had good luck since switching to super soft agar for swabs. I used to go through swabs and get nothing but now I get germination on like half my plates at least

Wait a week or more? Are you talking SAB? My swabs seem to do just fine with some time in front of the hood. Then I'll just slip em back into the sleeve and close em up with micropore tape.

Well I don’t use a SAB or hood, so I just throw them back into the sleeve right away and wait at least a week to use em... been working out. That’s how I’m getting my crosses. Your way sounds legit

But I'm out of swabs, so that'll have to wait. There's just one little hopefull piece left, all the other plates have contamed out.

Made up some sub this morning for 6qts of the APE that are ready. I’m following Fahts 16qt PE tubs for these.
RK APE 3 in the wide mouth is the one that was looking slightly off from the rest but now I’m not sure. I’m just going to spawn them all either tonight or tomorrow. 2 myco qts per tub staying with their numbered counterparts.
The RK APE 2’s were shook on the 31st and are coming around. They’ve been lagging behind the entire process.

First signs of germination today, 9 days after using swab.
The first two plates I rubbed/streaked with swab. Nothing yet.
Third plate I cut off a very small piece, finally turning white around the edges.

Alright dudes/dudettes I spawned 6 of the 8 jars this morning to some 17qt tubs following fahts tek.
The grain broke up in the jars easy and smelled fantastic. I only had 1 gallon bags on hand and they worked just fine.
Three tubs that all look like this. I wasn’t sure about the initial hole placement and had only drilled one tub as a control. It was perfect considering these will be cased at 100%. So I drilled out the rest and went on my way. Cheap 8gal trash bags used for liners, about 2-3 inches trimmed off all around.
Now we wait...

Dang sorry about your luck Mario, swabs are tough. What's your agar recipe? I've been having really good luck lately using a water:agar ratio of 1L:15g, might even go softer soon like 12-13g.

The last swab threw some crazy fast mold. Like super super fast shit. I've got 2 more swabs that I'm gonna try but I might wait until next month. I'm pretty sure that I'm moving in about 38 days. All projects are gonna come to a halt.

I tried the same ratio of agar last night for some other swabs, coincidentally, and found it was too rigid to press the swab in and have the agar squish around it like jello or vaseline would. instead my agar would crack and split (but my agar could be stronger per gram or something too). That's the consistency I'm going for now with my swabs, like soft jello, trying 12g to 1L today. Definitely worth a shot IMO, I've found I prefer softer agar across the board now basically. shreds up super nice when dropping into grain as well

We have liftoff! The only growth I see is in the plate where I buried the swab.
For those that have buried swabs in the past, do you ever see a very light, almost transparent ring of myc on the surface as myc grows within the agar? It’s hard to see in the picture but the white is likely mycelium on the surface. If I hood the plate while shining a light on it, I can see what looks like a circle of myc on the surface spreading out from the swab. It’s just super faint and almost looks like a circle of dry agar rather than bright white myc.

Roarkell, what’s the timeline from spawn to 100% colonization? I know APEs can be much slower... and at day 6 from spawn mine seem to be doing OK. I will grab a pic later and update. What’s throwing me off is I spawned another 2qts to a fourth tub. It’s 4 days behind but it’s almost caught up to the other tubs in just 2 days. It’s also from the jars that were lagging behind the whole time. I’m hoping the tubs aren’t stalling out.

I’ve only had one other failed APE attempt. Is anyone casing these? I was planning on it, but since searching up and down on APE I’m seeing no one really cases them.

Well I guess I’m going full experimental on these. I did not top layer and I pre mixed a bunch of casing that just needs hydration and pasteurizing. Since these are the only pe variety I have going I will case 2 tubs at least. Just when I feel like I got shit figured out, I go and try and grow these. Updated earlier post with pics

Wow, those turned out excellent! I'm in the process of moving and everything is being put on hold. All my stuff is about to get packed up and I'll spend the next few months replacing drywall, painting and installing carpet. Its so depressing but I'm also upgrading from a closet grow to a dedicated room. So that's fuckim sweet.

So one plate is mold city and the other one has a tiny spec of germination on the swab but there's bacteria also colonising just to the right on the tip. Gonna have to try and scrape that bit of myc off onto a fresh plate

Day 9 for three of the tubs. They look alright. Not much more growth since the other day, but it is APE after all. Took the mp tape off the 1/16” holes around the tubs. Glad I have other projects going on

T1 has grown out quite uneven. Hoping there isn't any underlying contam. I know there is a water drop on the surface from lid condensation dripping down, that hasn't grown any (yet....)
Low photo quality from cheap no-pour containers.
Going to split this one up into several T2's tonight.

To be honest I cant remember. I'm pretty sure it was early on and like 3 or so transfers from spore plate. I know my planet was a clone at least, same one I still got going.

Just learned about this thread. Major hype thanks roar.
Currently I have:
One mono 10 days from spawn:
A bunch of plate pin clones, looking from decent to good so far on agar:
And a bunch of somewhat but not hopefully not overly bacterial spawn:
Couple questions:
Anything I need to do differently to APE in a mono vs regular cube varieties? I just upped the FAE a bit and will likely be misting more often, depending on how much I decide to up the FAE. But otherwise can I just let it rock and keep surface conditions good?
How common are plate pins for APE? I had a bunch on my germination plates that I cloned, they didn't look exactly like normal pins but were undoubtedly little white blobs lol.

Lenz, that tub looks great. Far better than what I’ve got going on. I didn’t do a top layer at spawn but I’m not sure it would of helped me. Thinking I’m dealing with something bacterial. I’ll let the pros answer all your questions since everything I’ve been doing lately has put me into feeling like a total noob again haha! Def keep us updated on that mono win or lose

Looking back on photos of the spawn, all but one jar looked bacterial. Thanks to p9hu7’s clean spawn checklist for the insight and keeping me humbled. Thought, well it smelled good! Now I know, classic mistake. I cut a few corners on my pc cycle, and I’m pretty sure that’s what got me. I will let the tubs sit for a while, as I don’t need them right away and see if something can fight it’s way through for clone material. Now I see why it was taking FOREVER for the jars to colonize.
Reposting the pic of spawn... I can now clearly see issues based on p9hu7’s checklist. Sparse uncolonized grain, wet spots etc. IMO the RK APE 3 reg mouth looked the best and should have been spawned separately. The 2’s were always days behind and should have been tossed. If mushrooms have taught me anything, whether in cult or the experience they give, it’s to LET GO haha!
Excited to see everyone else’s grow, and hopefully the checklist can help shed some light on people in the same position I was in.

What's the consensus then? you guys seem to be scraping a piece off and burying some of the tip (just the tip mind and only for a minute). Probably going to try a bit of everything tbh.
The agar I have poured right now is 2% light malt with a bit of yeast, a pinch of gypsum and a whiff of rye flour.

Here it is: Not bad for MS to BRF to coir. I swabbed the snot out of 3 of the biggest fruits so I can do agar transfers to oats in the future. I want to get to a point where they all fruit at once instead of over the course of a few days or weeks. Anyway, I just pulled around 280g from a mini tub. At this point in the game, I will take it.

My roar plates are heavy on the contam side at the moment although I opened a couple of prints in the SAB before swabbing the APE so that may be to blame. Just waiting for that spec of myc to get things moving.

I have little experience with swabs, but I think you just described the best method. All I've done so far is just streak a Z pattern. So far, it's worked with MM, PE, and Ghost. There's weren't many colonies from any of these varieties though so I'll see what they even produce once fruited. No luck with this APE yet, so I'm gonna try pulling apart the cotton and burying it like you described. Trying to finish up some other projects first so I have the plates available.
Would love if others chimed in with other useful methods. I wouldn't mind trying a few to make sure I get growth

I slice the tip of the swab into four pieces with the scalpel then bury the slices into the surface of the plate. Hasn't failed me yet.

Ok, I can't visualize how you do it. Can you describe a bit more.
I've been thinking since the swab is "one and done", I would have a small amount of sterile water, stick the swab in and stir it up. Then innoc boo coo plates!

Think of it like you want to use a knife to sharpen a pencil. The swab strands are wrapped all the way around so make vertical cuts before you try to shave the pieces off. The center of the swab is hard due to the stick being there so only try to cut the soft outer part. I get lots of contam when trying this. Small pointed scissors works better for me to cut the littlest pieces off the tip.

Nothing makes me happier than a plate that looks like that (well of course except the entire plate looking like that with no contam.That is the apex of "hell yeah"). It makes me happier than a good harvest because I know that I have a great chance at a nice harvest when I see those fingers! The more I get into this the more I like the process more than the harvest. I may try my hand at Oyster, Reishi, or Lion's Mane.

Looks good! I have some little clusters of knots starting to show up. I almost tossed the tubs a while back. Kept them around till I absolutely need them or they throw some green. Spawned on the 12th of feb and no green so it’s most likely slightly bacterial. Only one tub has pushed out the smallest dot of metabolites. So they aren’t as bad as I had originally thought. The sub has shrunk quite a bit. If/when they flush I will have to bottom water.

That’s the word on the street. Hence why they will be tossed if I need the tubs or I see green/overactive metabolites. But for now they’re doing way better than anticipated and are away from my main grow space. 3 weeks is tomorrow and I just noticed the smallest knots forming. Will probably see the first pin in a week.

Thanks Dal, no I haven’t tried them. Got my ass handed to me with Melmak. Probably the closest to ape I’ve had.

I just finished a BRF to coir grow that took the following (from my notes): March 1, 2021 harvested the first flush of APE o APE tub first flush: 257 grams wet 8 grams dry (16 dry) 94% water to fungus ratio 63 days from inoculation to harvest 42 days from inoculation to birth 22 days from birth to harvest Bottom watered for 24 hours and put back to FC

So I’m seeing knots at just under 3 weeks. Not too many but hopefully one of the clusters develops and I can grab a clone to work with. We’ll see what happens. I will post pics if/when there are pins

Well it looks like those knots were contam. They started to change a crazy yellow. Pretty confident that is Aspergillus.
I think the source of the tam could be my pellet stove. When I drop a bag of pellets in the hopper there’s a decent amount of dust that comes up. Tubs have been tossed. Not sure I will be revisiting APE anytime soon. That’s 2 for 2 on the fails. Good luck to everyone.

Been too caught up with shit the past few months to have anything great going on. Finally back full steam and looking down the barrel of hopefully around a half dozen APE tubs in the coming week. This tub is from some early finishing spawn. Excited to be back with these bitches.
Oh and I should clarify, these aren't roars APE, they are mine but I wanna play. 🙂

I made a couple T1 plates after germination and they slightly colonized the wedge and just went dormant for 2+ weeks. I was about to toss them and then all of a sudden they started growing out. Very irregular, but growth nonetheless. My temps are on the low side (65-70) but I’ve never had something take so long to jump off an agar wedge.
Has anyone experienced something similar with APE? I’m going to keep transferring, but at this rate it’s going to be fall by the time I have something to spawn.

That's what I see people report so often. This is why I won't let this culture stop, and it's the first one I ever did, got the lottery on that one. I'll be distributing swabs before too long, maybe someone will get lucky and that trait will come to them.

So I'm goin to revive this thread, I hope no one minds. I had this growing earlier this year when everyone else was and got it all the way to grain but had to stop because I was too busy in the summer. So, after a long old battle with bacteria ruining all my grain jars except for one, I had to put it in the fridge and pray it didn't die. Until now that is.
Anyhow, long story short, It's alive and I just put it to bulk + a couple of agar plates for storage -- coir and verm at 4:1 substrate to grain. I have no idea if it's a viable strain. fingers crossed.

The box here is about shoebox size because I only had one jar of spawn by the time the bacteria was done with the rest. Hoping to see some pins soon. there is one little knot looking spot in the middle. I drilled some holes and stuffed them with polyfill after this picture was taken. room is a steady 20 deg C with white LED lighting 0n for around 9 hours a day.
Anyway, so far so good.
