I missed out on LAGM. This could be fun too.
Jan 24: Agar plates swabbed, last one cut small strand into.
Feb 2: First signs of germ on strands Feb 4: Growth spreading!

Feb 5: First Transfer Feb 13: T1 growing uneven
 Uneven growth made for stalled out T2, time to backtrack to germ plate
Feb 17: another T1 plate made Feb 22: looking better. Second Transfers cut.

Two plates for the APE swab: one contam and one a light circle of growth?? Next time I use a swab going for more plates for that swab.
14 January, 2021:Well...  Progress is progress, no matter how small... 20 January, 2021:From five plates with four wedges ea. Transferred two plates, 4 wedges ea. of SW APE for T2. Well I think I'm learning. I'm realizing it's a numbers game. I've started putting four wedges per plate. So last one was 20 wedges, so it's like 20 plates! But only five. And out of that I managed to go T2, two plates for 8 chances at a winner! Also I've started looking at the growth with a strong magnifying glass from the back as the growth shows better. Learning to look for straighter, better organized. 21 January, 2021:Ok, swabbed four plates, including the swab tip last one. So, second try. Nothing to see yet... 28 January, 2021:So has anyone else looked at their swab doing nothing in the plate and ripped it out and shredded it to three other plates?  7 days old And just look at the contam coming from behind where I held the swab. Then I had a second thought that I should have put a drop of water on each. Anyway just a noob larnin 'bout swabbin! (ya really lumpy green agar, more larnin! ) 10g 2% Agar, 7.5g 1.5% LME, 500ml 12/30 pretty hard Split 2 jars grn & blu/red π 325ml 28 January, 2021:

Eh, it's only been 4 days and burying swabs seems to always be 3-5 days slower than loop streaks. I'll give it another 5 days and if I don't see anything, I'll put a drop of sterilized water over each part of the buried swab and see if that makes em pop.

Rolled one swab to two PastyPlates. Temps will be in the mid to upper 60s for germination. I will keep this thread updated once I have growth.

I took Roarβs advice and tossed a space heater in my germination room to get my temps up. I noticed a tiny white spot on one of my plates yesterday which if itβs not a contam, would be 3 days to germinate. Itβs slightly off center of where I swabbed.
Now that some of the condensation has cleared up in my plates, the exact center of the plate where I first made contact with the swab is starting to look like germination may be starting there. I will know for sure by the end of the weekend.
2/1/2021
The small dot on one of my plates hasnβt changed at all since my last post. No growth, no color change, no nothing. Still no idea if itβs a contam or spore germination. I will monitor. Nothing on the other plate.
In the meantime, I took the remaining swab and streaked a few more plates and buried the swab in one of them.
Hoping for some success on this round.

We have liftoff! The only growth I see is in the plate where I buried the swab.

You might wanna find a warmer spot to set them until you get some germination... APE are notoriously slow and a lot of people have some difficulty getting germination from swabs in general... just my 2 cents tho... hopefully you get something to pop...

Thanks for the swabs, roar!
I put one of the swab tips in the bottom of measuring cup and repeatedly sprayed it with 30ml of distilled water from a syringe, an extreme bastardization of faht's tek. After a few cycles, I put a drop in an inoculation loop and streaked a pair of plates. Then I cut the tip in half and streaked another pair of plates. All 4 plates were 1% agar, 1.5% LME. I'll hold onto the other swab, in case this doesn't work out.
I'm also tracking this grow in LAGM2021.

Just saw this pop up. Iβm jumping in on this, my swabs are from Roar through a trade. Took some effort to germ but I have 8 colonizing grain jars now. All have been shook and should be ready for spawn in the next couple weeks depending.
 Pre Shake (20JAN)
 Recovery on 23JAN
I shook again yesterday to speed things up a bit. Recovery looks good, no pic yet.
All jars were nocβd with small agar wedges and took forever to colonize, I would recommend using larger or more pieces for faster colonization.

I'll hop in when I get my Roarin' swab. I've been having troubles with germinating swabs, poured from softer agar yesterday... Will report back!

Was frustrated at germing plates with swabs over the past couple months and got tired of just burying the whole swab in the plate so I started to use a fresh sterile 28g needle to rip a tuft of spore laden cotton off the swab which I then drag down into the middle of a thickly poured plate so it's sammiched around with agar like the tip of a buried swab would get.
Works like a hot damn.

About how long until you guys are seeing germination? Mine still aren't growing anything except for a really small spot blue bacteria.

I didnβt document a timeline on germination
I am having good germ luck with an 8g lme 8g agar to 400ml distilled water recipe in combination with dipping the swab in sterile water first. Another good tek with swabs has been using this brf puck recipe and taking a few strands off the swab and shoving it in the puck.
