First time working with agar

I’ve never used agar or “grown” agar. I bought some prepooured plates from a vendor. I mainly wanted the plates for cloning. Anyway I decided to use two plates on a spore syringe. For days I didn’t see anything. Now I see mycelium growing from the spores. The one plate has a orange/brownish dot in it. Is that a contam?

Yeah that small dot is bacteria most likely. And it does look like you have some viable myc there. Let it grow out a bit to about the size of a nickel and take some xfers 👍

Make sure your xfers are small. You really only need just a pinhead size of tissue for a xfer to work so less is better here. You can place 3xfers on each dish in a triangle pattern to save space and dishes

Looks like they are germinating.

Yes the orange is contam. I may be wrong here, but it looks like something bacterial. So you don't need to worry too much as it will spread across the agar and won't sporulate like a mold would (? would be awesome if someone could confirm or deny this ?) Just make sure when you come to transfer you take a piece of myc on where near the bac.

I'm thinking that has to be a contaminant, I'm thinking bacterial. You'll know soon cause it'll start growing pretty quickly. Doesn't mean you can't take a transfer from it though.

In my experience bacteria doesn't expand on a petri dish

I have those 2 dishes sitting in a 6qt shoebox with the lid one. Should I remove the one with the contamination? Could the contamination transfer over to the other??

Also thanks for all the reply’s... I never did agar work so I really don’t know what I’m doing. I did watch a couple of YouTube videos. I’m excited to start cloning.

No need to worry about contaminating other plates. That won't happen unless you have lids of both dishes off and your sterile tek is poor. Make sure you wrap them with saran wrap or parafilm before you put them in storage while they grow.

Like I mentioned above. Bacteria is very common to see on dishes and I have yet to see it expand like mold does.

Thanks! So wrap them with plastic wrap? They don’t need some air exchange like colonizing jars?

Also I have some LC syringes. Can I use LC in the dishes?

It advised that you test LC on agar before shooting it into grain. LCs are prone to contam, and it's hard to tell if they are dirty. Dropping a bit on agar can let you know whether it's clean or not befor going through all the steps to prep grian.

I have ape LC that I’m looking forward to spawning. Thanks for the info. I’m gonna test it on agar today. If it’s clean it will grow? If it’s dirty I’ll see contams?

If it is clean and grows, I can use it on some qt jars?

If it's clean, it'll look like healthy myc

If it's dirty... well it'll look contaminated. Normally it's pretty obvious

If it's healthy you can use it on whatever you want to go with. A little goes a long way

Depends on culture and how much agar, whole plate? slice? whole plate sliced up? I use crack plates or 60mm petris so personally 2-3 weeks and I leave my plates intact.

No, more likely it will form colonies the resemble a string of pearls rather than the bacterial blob growing in size.
