PLEASE HELP I just can’t seem to figure this out

Okay so I’ve been working in making my own agar going on week 3 now.... not trying to do anything special, just get a good recipe to grow mycelium.... I’ve been bouncing back and forth between PDA and MEA but not having much luck..... for some reason a lot of my plates are turning out looking like this...

Any idea what’s up with the blotchiness and what I could do to fix it??

I sometimes get 3 out of 20 plates with that I toss them. Flow hood. Improve your aseptic technique. Watch that "home mycology" YouTube channel he is a sab guru.
Good yo bounce back and fourth agar to keep the culture dancing and guessing and fighting. According to mckenna the way to stop senescence is to switch up the agar. Is that true anyone?

Thats bacteria.
Those splotches are from air or falling bacteria getting into the plates during transfer. Tighten up your sterile technique and you should be fine. You can also transfer from that plate into a clean plate if you need to.
That being said, Look up D3monic's Punch Tek, works so much nicer than trying to slice a piece of agar and then transfer it.

Agree. I'd like to add that condensation in the plate makes this contamination much more likely: bacteria have an easy time penetrating when they can travel through water. OP should review their pouring process.

Is that the one using a hole punch (for leather)? I tried that but found flame sterilizing the punch is quite hard. Also half the time the agar piece would end up stuck inside the tool. To each his own i guess

If you are using a SAB I suggest using no pours. Pouring requires a flowhood. If you are using a flowhood it is either your technique or you aren't getting the agar sterile. That is a lot of bacteria.

Pouring does not require a flow hood. Sure makes it easier tho But I poured agar in a sab for 2 years.

It may not require it but it's such a pain without one you may as well run no pours. I'm talking for efficiency sake here. By all means carry on.

I've just started cultivating again after a 7 year hiatus and use a sab for pouring. I have practically no contaminated plates. Sabs are great, you just gotta take your time and spray it down well.

Not here to argue, saw that plate and said to myself "he's moving too fast or bumping the top" flow hood makes it 1000x easier to pour and no pours add 30 minutes to the process but you do you.
