I'm a major proponent of grainwater agar, and just successfully cloned a local lion's mane onto it!  The way I've seen it, if I would get good growth on grains I should expect It on the grain boil water, it can be sterilized practically as long as I want like grains, and nutrient saturation is comparatively less important and far less likely to be any kind of detriment like grains. I've used exclusively wheat, ryegrass seed, or brown rice boil waters for several years. Other recipes have to work great as they have for years, as I usually had fine just going with malt extract for several years before, but I've grown to love relying on grainwater.

After capturing water in some bowl or pan under the strainer, I set it aside to cool a while and settle. After some time there will be a clear difference between top and bottom layers. I carefully gradually pour off the top water leaving the thicker starchy later in the bottom and either use this for some more novel projects or rinse it down the drain. The clearer top water is allowed to fully cool in refrigeration and settle, again pouring away from new sediment.

For longer storage or for the clearest agar, I fully freeze this grainwater which converts many dissolved starches to resistant starch and creates more precipitation from the grainwater when thawed, even sped up in a microwave. Once again I slowly pour away from sediment and then enjoy the clearest agar or liquid medium I've gotten from working grainwater.

It's a little bit of work, but with a little side effort now and then while preparing grains I avoid having to buy, ship, or carefully measure an outside material. Now if only I could produce agar-agar!



My lion's mane was very successful despite a rather mature specimen. Contaminations were able to show strongly, and it reminded me that trying a few dishes with agar-agar only is a good idea for potentially dirty clones.  I ended up having a pick of several apparently clean initial dishes though! The mycelium is very classically Lion's Mane, tho the dishes (or in my case, containers) tend to show the more explosive maturation when I occasionally boil some hardwood sawdust into the grainwater. I find this unnecessary for most grow-outs.


I'd say be ready with other materials you're able to trust too, but grain water agar IME is amazing!

I also have a very 'mature' LM fruit that I'll be cloning. Got it from a guy that had it in his fridge for a week. Certainly not edible, but I should be able to get some clean culture when I rip it open

If you want to do all that work go for it. But don't think you are saving money doing it. Time is money as they say. I just bought 1kg mea for $15 I have to wait for 2 weeks to get it though.

I use grains as spawn for my actives, so I have lots of grain water stored in the fridge. Just got to mix it with agar and PC. I don't mind the sediment at all!

But since I got you here, SHROOMSISAY, wanted to say you're a legend! Your contributions to the gourmet forum have helped me understand LM much better.

If you make it around to this thread again, I have a question for you: How long will a big chunky lions mane be good to culture sitting in the fridge?

I came back to check this thread to add that, while I get consistent results with grain water, lion's mane (and I think this clone might actually be bear's head tooth, or Hericium Americanum) does act on it similarly to how it acts on grains. Kinda slow, and the aerial mycelium visible is not very dense. I leave the dishes out on the shelf colonizing for quite some time before they ever end up needing refrigeration. I do recall it growing a bit faster and being more prone to spontaneously pin on malt agar.

Went and checked a chunk of mushroom I had left in a container in the fridge just in case, and it did NOT look good for cloning. I'd say more than a couple weeks and stop hoping for too much luck, but it would depend on the health and maturity of the mushroom.

Thank you for the kind words!! On your question, I really wouldn't know. I would do them ASAP. But if you have something you want to clone throw it on some agar and see what happens you have nothing to lose.
