hey everyone. i got some colonized grain jars from a friend and dtarted to isolate them on agar i was very happy that i was making a monoculture and my grows are gonna be better. but i was actually isolating some pretty small fruiting genetics even though they looked superion in agar. so i decided to go for a MS run and then clone the best fruits to agar so that i know what genetics im isolating, and after a few runs ill have the desired monoculture. now my question is that how can i get the most genetics available for the first gerow so i can select the one i want from a broader set of genetics ? i know spore syringes are famous to do so but what about spores to agar ang then to grains ? it seems safer and simpler to me , but is gonna still give me a broad set of genetics ?

Spores to agar and yes you will still get the same widespread genetics

aside from lookin pretty a TRUE monoculture dont mean dick as u found out. i think way too often ppl spend entirely way too much time on agar. i want to get on grain as fast as possible. i dont even bother to grow any plate/culture out any bigger than a qtr and im always on grain by T2 never later than T3 if u cant get a clean grain worthy culture by then you really should go back to the drawing board or review your process. clones are what i am after and i want my initial genepool as large as possible to choose potential clones from. to much time and too many transfers on agar only serves to work against you as you can be transfering away from something you may want without even knowing it. and the larger you let a culture grow closer to the edges of the plate only increases your chances of not having a clean culture when u do go to grain. a pretty plate dont mean fuck all. i have had ugly ass plates put out amazing killer fruits and vise versa. you will never know til u fruit it. so why not just start fruiting as soon as possible. then once you have a clone that is good and has potential you can start taking transfers and fruiting every 1 out in small contained grows while always saving part of it to go back to if its somethin u like. eventually you will end up with the best possible fruiter but it has nothing to do with it bein a monoculture

Aw yes thats true, I spent way too much on agar. U know I loved the shit and I was overdoing it haha.. but now I'm getting my agar and spores ready to reset everything.

They look healthy.I wanna go to grain no further that T2 so as not to isolate any genetics, but on the other hand there is also another purpose to transfers and it is to make sure we are not getting any contam into grains and to do so one needs to transfer a very small pice of leading edge . But im afraid by transferring a small sample im gonna isolate some genetics and dont get enough large broadest of genetics to start with ! And i really touched everywhere on agar with loop when I was putting spores on it so I shouldn't trust these initial plates, though my loop was flame sterilized and they pretty look healthy. So how do u think I should go about making second transfers ? Should my samples be bigger than usual (when were isolating frome other genes and contams) ?!

there is so many sets of genetics in a transfer its insane. take a small transfer or several and put to new plates n grow it out a lil bit. thats how u tell if its clean. hell thats the main point of using agar. contams are fairly easy to spot when u know what ur lookn for

Aw good, Actually, contamns were not my problem, Yeah they are visible. i was worried about unwantedly cuting down on the number of genetics i was going to grow. but im apparently just over thinking it, as u say its alright, thank u

I put everything to agar first to make sure there are no contaminants. Spore syringes work fine with PF Tek, but I'm not chancing it with tubs.
Why don't I trust spore syringes? It doesn't matter who made them, they are still coming from spores which were growing on the underside of a mushroom, which certainly wasn't in a sterile environment. You can always just put things to agar first before putting it to grain. It won't cut down on the genetics if you aren't isolating.
I also love agar because you can store it and you don't have to wait for it to germinate every time you want to make a run.
