Just got my two 500ml media bottles in the mail!! I'll be pouring plates today or tomorrow to get ready for the spore drop. The real trick is going to be to not get fucked up on New Years eve so I can inoculate before bed since we're having a few neighbors over for New Years brunch and everything needs to be out put away before then. Can't wait to start this log. Cheers, mates!

I didn't get back in the country till Jan 17 last time.
Although for those wondering about starting late, it was cool for me
because I was watching everyone go to agar. So I ended reading all about
before I did it.

2021 LAGM
January 3
Today I finally got around to pouring some MEA for some Nepal Chitwan spores. I chose this variety simply because it was the freshest print I had at my disposal. The print was part of a trade with some rando awhile back so we'll see how dirty it is. I ended up swiping spores on 10 plates with an inoculation loop.
 It's worth mentioning that I forgot to tighten the lid on my media bottle for like 20 or 30 minutes after taking the cover off my PC to let everything cool down. I'm eager to see if I end up with a bunch of contaminated plates. January 11One plate is showing a few spots of germination, will update in the next few days with pics. January 21It looks like I have some shit riding along with my chitwan mycelium on the t1 plates from the 11th, and I don't yet know how I'm going to proceed.   January 22I did swabs on the print and buried them in some plates, also took some transfers from the shitty t1 plates. The growth I've been observing really doesn't look like I can do anything with it so I'm probably wasting my time. There was an ok looking germ plate that I might be able to get something from, so I did a few transfers from it. March 5Here is one of four or five t2 plates from that germ plate I previously went back to. The growth looks ok enough, so I'm going to noc up 6 jars and do some transfers.  I'm still really bad at taking plate pictures. The condensation on the lid doesn't help any. Right now some wheat is being prepared so the jars should be inoculated some time tomorrow morning.

I started my agar at midnight on 1.1.2020. I'm using Telephone brand this time. When pouring, I realized I mistook the old bag of agar for LME. So I waited until it was light out and poured plates of LME agar, some with 2mg honey per 500ml.
Few if any of the agar recipes posted here specify liquid or dry LME. Liquid is more common in the home brewing background I came from and I had to guess that mycologists would work with dry LME.

Something interesting looking among the gunk on one of the APE plates.

Puerto Ricans germed a couple days ago.
Two spots of something white germing on one of the P. adiposa plates.
The first APE plate that looked like it might have mushroom mycelium is looking...someone tell me how it's looking? Anything salvageable? I have Furan-2 to try in my next agar dishes.
Here's what another gunky APE dish looks like.
There might be some growth in this dish with is more uniformly and has less apparent bacterial growth.
While starting a Lions Head project 1.4.2021 I noc'd up one jar of grain with the APE MSS. If all else fails maybe this will allow me to get a spore print to take to agar really late.
1.14.2021
Row 1 APE day 13. The leftmost is the dish is the one that was so colorful recently. Looks like the mycelium is winning the battle.
Row 2 Puerto Rican day 13. Is the largest growth the best to take a transfer from or do I want to prioritize vigor and symmetry?
Row 3 APE day 10. Tried to noc up plates from the contaminated syringe by just getting the tip of a needle wet with spore solution and making scratches in the agar. One point of germination and no nasty bacteria that at least I can see as in the other dishes started from the syringe.
Row 4. APE rescue. I transferred to agar some growth from one of the WBS jar that also had a spot of mold. Hope I get some APE and spores!
Unpictured. P. Adiposa day 13. One small slow growing area of growth that looks to my newbie hopeful eyes more like mushroom than mold. No other germination points. Thinking about starting more spores.
Unpictured. With the last APE spore solution I started a BRF puck. The lid had sealed to the jar in the PC so it sucked the approx 6CC remaining in the syringe. It does not appear to be too wet and has germinated nicely.
Will be making more plates for transfers tomorrow or this weekend!
1.19.2021
Using new batch of LME plates with FURAN-2, 6g LME/8g telephone agar / 500ml distilled H20. Let the dishes site for four days in SAB before using them, very little condensation.
Took T1s from the once beautifully contaminated plate. A slice from where the battle was fought and two from a late germinating spot.
More T1s from other plates. How clean does the plate started 1.4 look?
T1s for the Puerto Ricans.
Giving up on the P. adiposa, perhaps will try again soon but have enough projects started for now.
1.29 APE T2s
1.30 PR T1 progress on day 11. Ready for T2 or not yet?
2.10 Had a pacer put in a few days ago. Behind on transfers!
But a few of my APE t2s look good!
2.22 PR to T3 yesterday.
APE plates are one transfer ahead.
2.26.2021 The APE T4 plates had too much condensation to photograph. I was sloppy on both Puerto Rican T3 plates, dropping the sample in a couple places. Maybe I need to try using two scalpels...or watch more videos by TCs.
3.3.2021 Plates looking better and growing faster each transfer. Plates cooling right now for the next transfer.
I've again failed to germ P. adiposa from a spore print taken from grocery store fruits. This plate looks to have two small areas that look close enough to mycelium to try a transfer; with this plate I just held a drying cap over the plate and tapped.
3.5.2021 One of the growths on the Pholiota adiposa germ plate looks much like the light, wispy myc I got from a Hericium erinaceus clone. So am taking a transfer and seeing what happens.

Got some wbs I boiled, rinsed and pc for 2 hrs (I got an electric pc that only goes to 11 psi, so I pc for extra time)
And knocked up a couple jars of wbs with the T1s .
Also mixed some karo and water in a 100ml media bottle and through a couple transfers in it.

OK SO ITS BEEN A MINUTE, AND IVE GOT A COUPLE GOOD LOOKING JARS, THE RW IS ON ITS 1ST TRANSFER BECAUSE I HAD TO START OVER WITH IT. BUT THE ABM (ALBINO BLUE MAGNOLIA) I INOCULATED A COUPLE GRAIN JARS FROM THE LC I MADE IS ALMOST DONE COLONIZING.
And i had a decent agar culture from a swab i mislabeled but i think is malmak, and i put it to some wbs:

LAST BUT NOT LEAST HERES MY NEW RW PLATE I TRANSFERED FROM A BACTERIAL PLATE:
Ill take 1 or 2 transfers from that before i go to grain, update in a few days.

Took a PE syringe that i've been holding onto forever now and finally put it to some plates. I started with 8 plates and hoping to get a little something from at least one to continue my adventure. Will be starting other varieties here soon.
1/11/2021: Got some PE germination!
1/14/2021: Took a whole bunch of transfers from the PE germination, got alot more than I thought I would!
1/20/2021:
Have moved to my T2 transfers, all is looking well so far. Wispy growth concerning but is organized. Will be taking a few T2 and then T3 to grain to see how it goes!
 
1/28/2021: Have taken T3 on all my plates will be dropping to grain soon.

Variant: Unknown -02JAN21- noc’d 3 lme plates via loop from MSS. One drop on two of the plates and the other got away from me a bit. All got a little zig-zag action.  -15JAN- MSS to brf puck -27JAN- Well here’s a breakdown of what I ended up doing:   Made this biopsy tool using a 5/16” leather punch and 1/16-309 stainless TIG wire and the wife’s hair tie (she had a brand new pack)  Suspected mold circled in red. Pan germ in blue.  Bottom side of germ  Screenshot of the transfer. I will admit it was stuck to the TIG wire a bit but came loose with a little turning action.  Then I split transfer into 4 sections with flame sterilized blade  What’s left in the puck All in all I’m happy with what went down. Took about 20 minutes to make the tool up. I will keep it in the myco tool box -3FEB-  Growth update Species: P. Cubensis
Variants: Maz, Whytegold, WGxTOC, WGxBlueMeanie, APEU Mazatapec-02JAN21- 2 lme plates via loop from on of my own spore prints. One I tried a zig-zag from 12 o’clock down and the other just kinda swiped across the middle.  -11JAN- No germ on the maz yet -23JAN-    No germ on original maz plates. I just harvested two fruiting tubs and swabbed fresh spores. -29JAN-  Took a couple T1’s -3FEB-  Fresh spore T1’s update -05FEB-   The T1’s looked so good they went to grain. 1/2 plate per qt jar. -18FEB- Spawned 4 shoeboxes -27FEB- Pins baby   -05MAR- Started harvesting... they’re coming in at all different rates  -06MAR-More harvesting     Whytegold-11JAN- I received an awesome package from ‘FK’ today and in it was a print labeled “Whytegold”. I haven’t read anything about this on the forum yet which has me pretty excited. It’s a rusty looking print (I didn’t get a pic of it). Used a sterile scalpel, scraped the print and jammed the spores into the agar. Not the best technique but I’m just not having any luck with the streak lately.  -16JAN- Germination on both plates.  This plate was streak/sliced with a knife so I’m not 100% on all the growth. Looks healthy enough. Will let it grow some more before T1’s.  This plate was one single jab with a healthy amount of spores on the tip. No satellite germination has me happy. Will let grow out a little more before T1’s as well. -21JAN-  4-T1’s of Whytegold and 4 APEU -24JAN-    T1’s are taking off nicely and I have my eye on some strong areas for T2’s -3FEB-  Whytegold T2 update (4 plates on the right top and bottom) The two on the left are APEU T2’s. These will be going to grain tomorrow. -4FEB-   Whytegold transfer to 8 grain jars. First time using half a plate per jar... improvements to be made on speed. We’ll see how these go. Guess APEU is tagging along in this WG thread. Transferred to 4 grain jars. Dipped into my second batch of jars and noticed I over boiled a bit, a little clumpy on the shake.  -18FEB- Spawned 8 shoeboxes -27FEB- Fully colonized and about to pin  -01MAR - 05MARPhoto dump uploaded in reverse chronological order      -07MAR-Harvested 142g wet of random mature fruits from several tubs. Cloned one.    Whytegold x Blue Meanie-25JAN-  Swabbed plates -29JAN-  Took three T1’s. If they’re clean they will go to grain from here to get the most genetic variability. I see other options at cross points but ran out of plates. -22FEB- Spawned 2 shoeboxes -06MAR-Knots and a few light/white pins could be something? Could just be Whytegold Whytegold x TOC-25JAN- Swabbed plates -29JAN-  Took two T1’s. If they’re clean they will go to grain for most genetic variability. I see other options at cross points but ran out of plates. -22FEB- Spawned 1 shoebox -06MAR-Decent amount of orange pins. Some good clusters. Thinking I have TOC, no cross. Species: P. Baeocystis 
GAME OVER -02JAN21-
2 lme plates via scraping the gills with a sterile blade onto the agar. I used the blade to push around some of the material on the agar. Donor is an urban harvested fruit out of the Seattle area. Pic shows dried fruit on top of the unopened maz print

Something has germinated. Both plates have an identical spot of something mysterious and it’s fucking fast.
-05JAN-
The growth has shot spores... 
-06JAN-
Tossed both plates, there’s no saving that mess. Not sure if I will try again. Hindsight: Should have taken prints after I found them.

Var: x7x (f2), Melmac, Gandalf  1-2 streaked plates with x7x and Melmac 1-5 first germination on the x7x   Still waiting on melmac 1-9-21no germination on the MelMak streak plates but did get some on the dunk.. along with some bacteria off to the side and a satellite probably from when I cut the tip off the swab with dikes over the plate.  I transferred the dunk to a new plate and took xfers of the growth coming off it.  x7x ready for its first xfers as well though I’m a little leary on the growth. What you guys think?  1-18Took some T2’s today.  1-25Taking T3’s of the x7x f2 and the MelMak   1-31 Finally got around to streaking some plates with gandalf   x7x looking like it will be good to go to grain soon as it grows out a lil more  2-10 Took transfers and sent 2 plates of MelMak to grain. Possibly to use as masterd, will use a T4 plate for lc  Gave a buddy a Gandalf plate, pretty sure it was the only one to have germinated. 2-26-21x7x plate pin  Spore under magnification   Sending the f4 plates to grain today   Debating if I'll send these melmak or make LC or both... not quite happy with how they look yet.   Forgot about the Gandalf swab plate. Looks like I got something to work with even if it looks sketch

The only spores I currently have are in a known bacterial B+ syringe, but this will be the excuse to finally try to make some antibacterial agar.
If all goes well, my first GT tub will be fruiting early in the year, so I can make a spore print. I also have grain colonizing with B+, AA+, and Tamp.
If you want some prints that are much cleaner let me know! Mail is still moving surprisingly fast...
