Plates swabbed!  Well, 2 were swabbed and one accidentally got a squirt from the syringe so we'll see what happens there.  Never used swabs before, and they are homemade, and using grain water agar that I had no recipe for, so this whole thing is probably going to be a dumpster fire.

My "Let's All Grow Copious Amounts of Bacteria 2021" grow is off to a flying start. 




Made up 2 more plates this morning, hopefully I caught the one mushroom spore in the syringe this time.

edit:  Looking more obsessively closely at my plates I noticed some wispy growth that could possibly be myc, so I took a chance and grabbed a couple of xfers to a new plate.

Xfers seem to be growing, so maybe my gamble paid off?  Hard to get good pics and can't upload at the moment anyway, but hopefully I'll know by the weekend what I'm working with.

3/4 plates and all 3 jars have already started growing hair.  (4th plate is probably just a slow grower.)  WTF.

Noc'ed up 4 new plates from an Ecuador syringe.  Hope it's not garbage too.

I totally failed at this last year (thanks school closures), so would love to give it another shot.

I have a KSSS print I haven't used yet so this seems like the perfect opportunity. I also have one that was kindly sent to me, labelled "GYM" but I no longer remember what that is - maybe we will find out if I successfully grow it!

Update 4th January

Last night I finally got spores swiped (well, scalpeled) to agar :smile:  For spores I like using these pint-size PP5s as no-pour agar plates, they're nice and roomy. They take up a lot of space so I did only one for each print. KSSS and a Gymnophilus. The KSSS is labelled as an in-vitro print so it might actually be very clean, however there is a good chance my poor sterile technique may have negated that. We shall see!

Update 21st January


Finally I have something to update! Up until now, the condensation of the plates has made it pointless to take any photos for you guys.

I have transferred from the germ  plate of Gymnophilous, which had what I believe is very nice growth and I didn't see anything that looked like bacteria or mould. I could be terribly wrong, but I guess we'll fine out :smile:

I couldn't find my BRF for the no-pours, so I blended up a little grain I had soaking, then strained it and used that for the agar nutrient. Should be fine?



I had a new bottle of food colouring and underestimated its strength - the agar is VERY blue lol. Here's a photo before they went in the PC.



I got 3 transfers from the Gym (actually 4 but the first was a disaster) and swiped some TWC to two plates.



I am low on jars and the places I'd usually get them have all sold out. I could buy them off Amazon but that's not a cheap option where I am. So, I went to a cheap supermarket, got 3 x 670g jars of pickled beetroot for 85p each, am eating my way through it all and hopefully they will be empty by the time the cultures are ready for grain!

Update 4th January

My Gymnophilous T1 plates have been doing well and ready for another transfer. The myc really seems to be munching the blue colouring from the plates!



TWC has germinated on agar, looks fairly clean as well. One plate was full of mould so I got a little worried, turned out there was like a 3mm hole chipped out of the side of the pot.




I've lost my scalpel (shit, I really don't want that laying around the house somewhere) so can't do transfers til it turns up or I order a new one. I used a sterilised spoon recently to scoop colonised plates into grains jars.


Can anyone advice on what to do with the Gymnophilous please?
Can it just go to WBS etc as normal or does it need some kind of wood added to the grain jars?
Will I need hardwood for spawning, and will some kind of woodchips or bark do the job?
Do woodchips need to be sanitised?
Will it want fancy conditions to fruit, or will some kind of shoebox be fine?

Gymnopilus is a genus of fungi... ive got a print of some gym. lutefolius that I've been meaning to run... :hmm:

Put me down for a few random cubes and the gym lute... maybe this is the push ive been waiting for... lol

Took me a while, but I finally gathered everything to join the fun. Made 500ml black agar last evening and poured it into 24 holy-grail plates. I've tested both the original approach (placing as many as possible in a jar) and Josex' approach (just wrapping every layer of plates with foil), got to admit I prefer the no frills one. The less that can go wrong, the better.



21.03.2021


The agar dishes survived a week of sitting around. I took six with the least amount of condensation for inoculation in the SAB. Put two self-made swab halves on two of them and used an inoculation loop on the other four with a vendor print. Doing a zigzag streak was tricky because the holy grail dishes are relatively deep, so I ended up bending the metal piece a fair bit and digging it into the agar. For the last two I've noticed I could just lightly hold the loop on one point and spin the dish around to make a circular streak.



26-28.03.2021


The most promising dish (read: fastest growing), on day 5 and 6 respectively:



The milky part is bacterial growth (been there since day 1, smooth and milky), whereas the wispy part is mycelial growth (became apparent on day 5, faint and stringy). On day 7 I took transfers from the mycelial parts. Four agar chunks from this dish, one chunk from two others. The remaining three have been dumped for not showing anything else than bacterial growth. Transfer work was lots of fumbling. The agar chunks didn't want to fall off the scalpel and when they did, they didn't land on the center of the dish, let alone on their face. I hope for the best

I was worried about the sideways landed pieces not germinating, but I was worrying for nothing, day two and they're almost completely covered in fluffy white growth now. I expect them to do the jump to the surface soon.



23.04.2021


I'm glad this place is back again. A bunch of things happened in the meantime, I ended up doing four agar transfers and prepared 18 pint jars. Made a bunch of in-progress pictures, but decided to only keep the last few ones because once I managed to separate the healthy mycelium growth from the bacteria, there wasn't that much of a change. I think four transfers is overdoing it (spent more than a month on agar work), but it was so fun I didn't mind.

Since Shroomery was down, I had to look up some grain jars guide that was indexed by archive.org, so I went for Bod's writeup and studied Stamets on how to do agar to grain transfers. I found the tip of cutting out a slice and halving it interesting, so I tried that out by halving a quarter piece every time. A bit fiddly to pick up both pieces with the scalpel, but it does indeed increase the chances of at least one piece not getting stuck to the glass or lid. It might speed up inoculation a bit that way. There's half of an agar dish left now as backup in case things go catastrophically wrong. I'm considering to do another run of holy grail dishes and just practice with other spores, like bisporus agaricus. I didn't consider that doing agar work can be this fun.

20-21.05.2021


I've let the grain jars sit a bit too long and found not only hyphal knots, but pale shrooms spawning inside them. After panicking a bit, I decided to pasteurize coco coir, waited for it to cool down, harvested the side pins, then spawned the grains with coir at a 1:1 mixture with no casing layer.

I kind of forgot about this thread and Shroomery in general. So here's pictures of my second flush:



At this point I'm pretty sure my vendor screwed up and sent me Albino A+ instead of GT. But it doesn't matter really. Potency-wise they're good and when dried, turn into a silver instead of a golden color.

That one surviving tub gave me several more flushes, one every week. It's ridiculously tough. Here's my latest flush from today, which took almost two weeks this time:



I expect it to eventually give up though. Harvested 60g dry from that one tub. I can only wonder what if the other five didn't die on me...

I'm waiting on 2 new media bottles to show up from Amazon. They were promised to be delivered by yesterday evening but that has now been bumped to 12/26-12/30...As soon as they show up I'll be pouring some plates and getting ready for the ball to drop!

I'm debating on putting some Pan. cyan Hawaii to agar for LAGM to but I won't have any way to dry out my hpoo until Spring. That project will just have to wait

If you can't get them in time TigerStrike you can always grow along when the arrive. It's fine if you start a little late, or a lot late really.

I gotta say it’s really weird doing this again. Seems like last years wasn’t that long ago, and that was my first time ever using spores on agar. This year I’ve got all kinds of bad ass spores and a flowhood.

Finally did t1s. Prob gonna make some lc and drop some germ plate wedges to grain tomorrow.

Did some t2s and dropped a t1 wedge of Argentine to grain. Got a NecD lc about ready to use also.

Been a long time, necd is in fruiting and Argentine is popping some nice clusters I’ll prob clone.



Pesh is still on grain but almost there.

Atl7 is looking crazy I’ve never seen such aggressive myc with stone producers. Finally got some pins popping off.
