I'm a bit rusty and shaky in my sterile tek but it'll come back to me soon enough. Couldn't find my media bottles so used a rando glass bottle with a pp5 lid but it was a bit bigger than I'm used to at almost 1L; took longer to remelt and seemed harder to find that sweet spot of liquid yet palatable to the fingertips. As a result some of the plate are gonna be a bit lumpy and overfilled. Normally I would allow plates to cool for 24 hours in SAB before wrapping but I may need to move the SAB between now and next year so I will likely wrap them only an hour or two over cooling. As such, a few variants have been added. A CRS print was also found but did not appear to have any usable spores on it; to the point that it could not be discerned even where the print used to be on the foil. (the good kind)![]() ![]() ![]() ![]() 1 PR plate 1 GT plateThe PR is a surpise. It is from a swab made from an MSS about a year ago. It was funky as fuck in growth and from what I've read/heard it's still pretty unstable so I happy to be able to play around with it again. GT is a favorite. One of my first and most worked with. This is sort of my workhorse variant and can't wait to see what it puts out this time around. Current growth is 50%-70% the area of a dime so I will hopefully have time and motivation to make T1s tomorrow. T0 plates will be one time use but T1-T2 will be done on the same plate. T3 will be done on its own plate as it will go to grain. Excited to see signs I still kinda know what I'm doing! ![]() ![]() ![]() January 8th, 2021![]() ![]() ![]() ![]() So unless I just get antsy and can't wait, I'll make up some more (better) agar on Monday and make T1s and probably some more germ plates for anything now going by then. I've never had plate NOT germinate. Ever. From the first time I started agar. Plenty of dirty plates, but never just no growth. The AA+ has some bacteria but I knew it was a dirty print so no surprise there. While a couple of the others are a bit difficult to see due to condensation, I see no signs of contams in them yet and I can see spores. They are just holdin' out on me. ![]() ![]() ![]() January 9th, 2021![]() ![]() ![]() ![]() The Z plate that had germinated now appears to be mold. However, the GT and PR plates continue to look good, despite being quite light and wispy (as good as they can on this PDA). On the upside, It looks like I have germination in both PFC plates. These were noc'd via MSS so kinda surprised they were not first to germ. Still very happy to see any favorable life in these guys right now. ![]() ![]() ![]() January 14th, 2021![]() ![]() ![]() ![]() Made a couple T1s and started more T0s. Mostly just wanted to get everything on MEA instead of PDA. A bit of ups and downs so I'll try to detail them separately. -GT -AA+ T0 -Z -CR -PFC -AA+ The Bad: Working in less than ideal settings; lighting was terrible and myc was quite hard to see while in SAB. Took 2-3 wedges out of T0s for T1s but I am not 100% sure I actually got any myc. The Good: MEA plate came out looking great! Like looking through amber. Poured a full sleeve but used less than half so plates ready to go for next transfers. Of the PDA plates I pour and did not use, none of them are showing any contamination. I'll not likely use them for anything unless no other option but it is a nice reassurance that my technique hasn't completely gone to shit. ![]() ![]() ![]() January 17th, 2021![]() ![]() ![]() ![]() Finally getting some action. Not much to report on most but GT and AA+ T1s are looking great. GT is my workhorse and this print came from like 10th generation of my work with it. AA+ I only worked with shortly before just kinda letting it go and forgetting about it so excited to see those come back. AA+ ![]() ![]() ![]() Mini Update![]() ![]() ![]() ![]() While my plates are not like crystal, I really have no need to get them more clear than they are. While the mix and procedure is fairly customized to fit my needs and preferences I feel it's a pretty easy and straightforward way to get your plates within the top 5 percentile of clearest plates. LME-15g tap water-450ml
To serve a bit of a dual purpose, here's a pic of my GT T1 with some kickass growth on it. Of 20 plate sleeves, usually no more than 2 will have a bit of sediment like the one shown. I'm not too worried about10% having pieces like that, it's the cloudiness I hate. If you check back to my update on the 17th you can see how much that GT has grown in less than 24 hours and I am stoked about that. I may go ahead and start taking T2s today. AA+ is looking very similar. Z still seems to be on the struggle bus but my have germination on the new PFC T0. ![]() ![]() ![]() January 19th, 2021![]() ![]() ![]() ![]() Progress at last!! Here's the Breakdown: AA+ - 2 wedges each from 3 T1 growths GT - 2 wedges each from 2 T1 growths PR - R&R:pin mold; poke Z - R&R"pin mold; 2 wedges from T0 PFC(a) - 2 wedges taken from 0102 T0 PFC(b) - 2 wedges taken from 0114 T0 CR is still holding out on me. Thinking my swabs are just shit; still hold out some hope though. PFC(a) and PFC(b) germinated about the same time despite a 12 day different. I think this is due to (a) being on PDA and (b) being on MEA. Naturally, I took Ts from both. Both was noc'd with the same MSS made by me from a print that was also made by me. Z and PR are going to be a challenge. The T1s are Rescue and Recovery plates. There is some cube growth intertwined with some pin mold but I think I may be able to save them. Tried a somewhat modified poke method using tweezer for one, and tried to catch the outer most growth on wedges with the other. Time will tell. AA+ and GT are doing well it seems and already jumping off to the new plate. Very excited to have AA+ back after a couple years of it falling through the cracks. GT is my work horse and while I know starting over from a print can leave a lot of wiggle room for performance, I am cautiously optimistic that they will not disappoint. Also pictured but not part of LAGM are some PFC clones. These are some pins I pull off a recent BRF cake when birthed. They are the first fruits (FF) I have harvested in over a year. However, they were NOT handled properly. Exposed to open air, I picked them barehanded, and they sit in a square ziplock container for a few days. I did not even bother splitting them, though I did at least transfer them using some flamed tweezers. I expect lots of bacteria in the near future. I figured it was my best chance at building up my cultures and spawn quickly. 4 pins on 3 plates and 1 plate attempting a tweezer poke again. We'll see what happens. R&R plates: ![]() ![]() ![]() January 26th, 2021![]() ![]() ![]() ![]() Got a little impatient and my AA+ T1 was looking good so I took a chance. Put about 1\3 of plate into each of 3 pt jars of WBS. I got lazy though and didn't rinse grain well enough resulting in some lumps that refuse to break up. We'll see what happens. About to pour some more plates and make some more transfers. Was checking out some plate with my scope and while I am in no way trained to ID contams, there are some plates and sectors I've decided look especially good and am going to pick up the pace a bit. Basically, every plate I transfer from I'll let grow out and if it looks good it goes to grain. Meanwhile I'll be transferring best looking\fastest growth more quickly than before. This should give me more fruits sooner and better more fruits later. Finished making transfers around 7 or 8. Made the first few T3s and more T2s. I think only 1 T1 this round. Have been checking out some of the T0s that didn't germ with my scope. Spores look healthy as far as I can tell and see no signs of germination. These are relatively old spores and were not stored properly for part of that time but I will just keep waiting. Hopefully something will pop off eventually. Not sure WTF is going on with the CR T0. Got some growth that looked odd but hasn't changed color or put out obvious pin mold or bacterial growth. Just looks like unorganized growth under the scope (I am not a train mycologist. It's possible I am simply missing the identifying features). When slicing for transfer though my scalpel hit something in the agar. Plate was noc'd with a swab and growth is from area where I dug swab in so possibly a piece of the swab? Put it on a slide but can't tell for sure what the hell it is. Went ahead with transfer and we'll see how it plays out. All in all, a very productive day today despite some very dumb and avoidable setbacks early in the day. ![]() ![]() ![]() February 25th, 2021![]() ![]() ![]() ![]() Forgot to get pics before shaking. Will try to be better about getting pics are the grow progresses. Started today: 5 shoeboxes (AA+ & PFC) 1 mini(PFC) Started previously: 1 shoebox (AA+) Not sure where I went wrong.... I know I started more than 2 variants but somehow along the way I seem to have forgotten about some and am now only working with AA+ and PF Classic There is still a RustyWhyte germ plate to working and some z plates I can transfer from. Not sure what the hell went wrong with my GT. It was my go-to for years but having just terrible growth and contam issues with my print this time around. May just dig out a fresh print and try again. They hold a nostalgic place in my heart and don't want to lose that line, but LAGM has highlighted some limitations I have with time and work space so I may wait a bit to restart those. Just the 6 shoeboxes, 1 mini, and a handful of cakes is proving to be more of a hassle than I would like currently and would rather grow a few tubs well, rather than a bunch poorly. Each shoeboxes have roughly 2qt coir and varying amounts of spawn. Some are 1:4, some 1:2. Mini holds about 3qt coir (one of the first I ever made and holes are a bit lower than they should be so it's basically just a tall shoebox) and has a spawn ratio of around 2:3. The one shoebox started previously is about a week along (will check dates later) and is looking pretty close to pinning. My confidence in the cleanliness of the spawn is not as high as I would like. It was the 1 jar of three that didn't not show obvious signs of contams. However, all three jars were noc'd with the same plate. I gave it an extra shake and plenty of time for them to show up and did not see any deal breakers but still know that it is less than ideal. This is actually the first time I have not included verm in my sub. When I started, coir\verm seemed to be the standard so I just continued with what I knew. I just didn't have any verm this time so figured I would finally join the rest in omitting it. This is admittedly not as flashy or detailed as my usual updates. Used the other swab to start a few plates last week so I really hope to get a good plate from this one or I'm out. 01/01/21 Box is ready Plates are swabbed. 01/02/21 I've decided to add 2 more into the mix. I will also be doing Tat and p. allenii 1/22/21 Update, these are all the plates I got to germinate from pearly gates. Nothing from tat or allenii yet. Can be from a print or a MSS or a swab and can use as many different variants/species as you feel like keeping up with. Only 2 restrictions: Must start with spores on agar and Can't start before 00:00.01 on 01\01\2021 Starting late is totally fine as this experience is meant to be about learning together and don't want people to miss out on that because they weren't ready in time. ![]() Just to clarify, this is not my thing so I'm just following the rule laid out by those before me. Admins and TC can make any judgment calls that need to be made if it gets to that point. Varieties: Golden Teacher, Albino Penis Envy, Penis Envy 12/29/20 Poured my plates today. 400ml undiluted grain-soak water with 8g agar-agar (telephone brand). I need to start using a towel or something under my SAB floor because it dips in the middle, causing VERY uneven pours resulting in not many plates to be poured. 400ml got me 15 plates. SAB arm-holes are loosely covered with paper towels. I know it's considered un-necessary but the way my 'lab' is set up air from my central heating gets blasted straight into it from across the room. Poured at 118-120F and stacked poured plates 15 high in an attempt to beat condensation at its' own game. Pictures to come after inoculation. 1/1/21 Got my plates inoculated today. 5 each of GT, PE and APE. I think my sterile procedure was pretty good but I'm hungover and shaky as fuck so time will tell! These are my clearest plates yet with very little condensation. Baby steps! 1/7/21 We have germination! After nearly a week of checking on my plates several times a day, posting on this thread and undoubtedly pissing off some of the people following along with my complaining I finally noticed some baby mycelium on one of my APE plates! Out of my GT, PE and APE I wasn't expecting to get anything out of this one in particular. Sorry for the clarity of the pictures. I poured some crystal clear plates, got impatient so I moved my stack closer to my heat vent and that condensed them like a bastard...live and learn. The closeup was attempted to be taken through my jewelers loop. Cheers, mates! 1/15/21 Today was an interesting day. While my BRF pucks were in the PC I decided to give my plates one last look before tossing them after sitting blank for 14 days. The one APE plate I had that germinated has sprouted a tiny spec of myc in another spot and one of my PE plates finally germinated!! I need my jewelers loop to see it, but it's there! Still waiting on GT, but I think these BRF cakes will do the trick. Anyway, I inoculated the BRF cakes (3 GT, 3 PE and 1 APE), took 8 T-1 transfers off my APE plate and did a couple transfers for a non LAGM project. Busy day and it felt good! Shout out to d3monic for his agar TEK...absolute fucking GLASS! 1/26/21 After wasting a sleeve or two of plates that wouldn't germinate I said fuck it and made a batch of BRF pucks. This is LAGM and desperate times call for desperate measures! My GT and PE finally got off their lazy asses and germinated. I took transfers from the pucks to agar on 1/22/21 and just made the T-2 tonight. These are the T-1s from the pucks: GT PE PE 1/28/21 My APE is crawling along, to be expected. 31Dec20 16:30: For some time now my usual agar recipe has been: -10g agar -8g brf (home-ground, and poorly so might I add...) -500ml H2O I add my brf to the water and let it sit a while, occasionally agitating. I don't filter or decant or anything. Not here to make beauty plates for the Gram. Agar added cold after the brf soaks a little while, then brought to boil and poured. Dishes allowed to cool sans lids, assembled (almost all my prepours now are jars with unmodified lids), and PCd for 30min @15psi (or steamed an hour, depending on what's going on)... 3Jan21 00:30: FINALLY streaked all my plates tonight: -3x semilanceata -2xGHalo -2×caerulescens -2×AA+ ![]() Golden Halo and AA+ showing germination. Each GH and AA+ got 2x xfers (4x total of each). Semilanceata and caerulescens both showing clear germination. Making more prepours today. Ok... so last week I made some agar and somehow the fuck or another fucked up my nutes/math. The resultant plates showed clear, derpy growth that I had to wait until it grew out until I could proceed with photos. My semilanceata and caerulescens are just now growing back out, as is the AA+ (enough to show photos of). The GH was its usual bonkers resilient to my whoopsie, and started to throw its trademark rhizo growth after some time. Took 2 xfers from this plate and may go to grain with a couple wedges for fun. The blips you see in the center are bits of poorly ground brown rice, for the record. Long time no update, but I am almost ready to take the semilanceata to grain. Hard to see with prepours but you get the idea. We're looking good. Temps inside are... really low lately. so progress is painfully slow. Might buy some RGS for this project but the car's head gasket is toast. Have to make it up to H Depot and check er out or might just have to run barley. Hmmmm... ![]() 1st caerulescens plate that looked decent enough to drop so dropped it as well as the best semilanceata plate to 4 qts (2 each)of Barley. Prep was overnight soak, followed by 50min simmer/boil. Mixing up sub for bottle grows (semilanceata). Not sure the exact ratios but in order of amount: manure/straw/coir/wbs/bran. Mixed last night, PCing today. Dropped caerulescens and semi cultures to jars of barley (2x qts for both) night before last (10Feb21). Inoculated 8x pts of semilanceata substrate with ~4cc of liquid inoculant (made from tossing a puck in a jar of sterilized water and razor blade and agitating). Mix was well-hydrated and yet shook well/freely. One jar at least will contaminate I'm sure given that while I was wiping down my lid,I accidentally jerked and plunged my finger and iso rag inside and touched the substrate... Semilanceata and caerulescens jumping off onto barley. Didn't have propane cash for stove but I was itching to cult, so I made 14 WBR 1/2 pints. WBR Prep: water brought to ~200f and rice stirred in. Brought to boil and kept at low boil for maybe 8 -10 min. I always follow the initial draining with a boiling water rinse. You want it to swell but retain it's shape (not "bloom"). I made a LI with the best looking T3 AA+ plate and inoculated all with ~3cc. Photos taken maybe 2-3 days ago(?). Example of visible germination now evident on all semi bottle grows and grain jars. Cold snap (pretty darn chilly indoors, even for Alaska) really slowed everything down so I was happy to see this. Will monitor and report. Finally got more Halo cultures going and first quart taking off nicely. I'll expand to 4-5 more for a 64qt likely. 3 more plates will be ready for grain by Fridayish. Shook first qt of Halo today, and made a LI out of this plate to inoculate 5x bottle grows (4xqt and 1xpt): If they don't contaminate I'll be the luckiest guy in the world. Wife wanted the DSLR I was using in the SAB and got me irritated and I had an... angry air box session... during the LI making. Never know. Mix for bottles was something like 50% manure, 10% coir, 30% WBR, 10% wheat bran. Semi bottle grows are coming along finally after cold-weather lag. 8 of my (AA+) WBR half pints are colonized so going to a 16qt tonight likely. 6 more @90% or so. Might do a novelty. Made 3shoeboxes eventually from 13 cakes (4/4/5).Not enough room in my space for larger tubs or expansion. Getting submarine level cramped in here... Remaining two half pints will be used for a novelty. Halo qt and semi qts are chugging along.

made a couple transfers from germination plate and then inoculated a grass seed substrate. grass seed prep: put dry seed in container with a pinch of gypsum, add water, sterilize for 60 minutes. I waited until I got in vitro pins/fruiting bodies and cloned those. My pin clones made pins so I cloned those: Next step: grain spawn! Edited by Haywire (05/11/21 07:02 AM)

Im going to biopsy a few plates tomorrow and see if I can get the myc away. 1/28 Got into two transfers since last update. This plate looks like I'm getting away from the original contamination. Jan. 3 2021 10:30pm I had some really quick growth on my cyan plate. Had a large clump of spores shoot out of the syringe. The goal here is to get fruits as fast as possible so I decided to do only enough transfers to get clean myc for a LC. Sorry for the blurry pictures this time.
Jan. 11 2021 I mentioned doing a LC in my last update. I will only do that for the x7x. I only need 1qt of spawn at a time for the Pan Cyans.
Jan. 12 2021 Very fast growth on my Pan Cyan culture. Jan. 18 2021 This is current growth 2 days after the jar was shaken on Jan 16. I already have a tray of Pans going in my FC so I'm not sure what I'm going to do with this jar yet. I didn't anticipate such fast growth. Jan. 19 2021 Gave it one last shake. I'll mix up some poo/straw tomorrow and pasteurize. Jan. 20 2021
Jan. 21 2021 Finally got around to working with my x7x again.
Jan. 25 2021 The jar of pans didn't fully recover from the last shake which is pretty disappointing but I am so desperate to have a pan trip I decided to spawn it anyways. x7x T3s turned into crap so I made up a fresh batch of agar and went back to the germ plate to grab some of these rhizo strands. Jan. 28 2021 9g agar 6g lme 500ml distilled This batch of agar and the rhizo strands from the grown out germ plate got me this sexy T1 plate. Current growth on the Pans.
