Finally I have something to update! Up until now, the condensation of the plates has made it pointless to take any photos for you guys.
I have transferred from the germ plate of Gymnophilous, which had what I believe is very nice growth and I didn't see anything that looked like bacteria or mould. I could be terribly wrong, but I guess we'll fine out
I couldn't find my BRF for the no-pours, so I blended up a little grain I had soaking, then strained it and used that for the agar nutrient. Should be fine?

I had a new bottle of food colouring and underestimated its strength - the agar is VERY blue lol. Here's a photo before they went in the PC.

I got 3 transfers from the Gym (actually 4 but the first was a disaster) and swiped some TWC to two plates. There's a question mark on one because I wasn't labelling as I went along and I'm not 100% what it is.

I am low on jars and the places I'd usually get them have all sold out. I could buy them off Amazon but that's not a cheap option where I am. So, I went to a cheap supermarket, got 3 x 670g jars of pickled beetroot for 85p each, am eating my way through it all and hopefully they will be empty by the time the cultures are ready for grain!

Don't you cover the arm holes when not in use? Lots of cool ways to do that.
I cut two circles in agar, an inner and outer. Then like a pie I slice many times. Then over the grain jars I run the scalpel under the agar with slices falling off. I do two or three jars from that plate.
When I shake first, damn pieces of agar stick to the jar.
I wanna poke! And LC tooooooo! Save me Joesex!!!!!!
p.s. Oh and I'm glad you took this LAGM GypsyBastard, you're more representative of the voice of LAGM than an old fart like me. Plus my head is straining learning Python now. LOL Good job!

Those agar pieces sticking to the side of the jar is my biggest pet peeve... ill shake a jar for what feel like hours trying to get one fuckin piece to slide down or break loose into the grain... lol

Yeah i use a couple of shoebox lids and put a torch or a spray bottle against them to help keep it in place. I'm planning to build a flow hood this month so it's fine.
Do you have cats? Mine are determined buggers. If they want in bad enough its hag ppening. One is still fairly kitten ish so he's pretty rambunctious.
Roar i feel that lol drives me nuts especially if it's the whole damn puck because no-pours. I've found that carefully rolling and up turning the jar to cover the agar with grains helps a lot with it not getting stuck but sometimes it'll be stubborn. In that case you just gotta hold the L and put your grain jar on the shelf sideways so the grains reach the agar
-------------------- No one likes a naysayer It's probably fine. Even if it's not fine, it'll eventually be fine. So it's fine. PE Adventure

I know huh,LI, Liquid Inoculate , I have to fabricate the blender thingy, but sounds more direct. Or Blenderless Liquid Inoculate - Munchauzen
LC Liquid Culture, ah brew up some wicked liquid myc! Pretty advanced shit and still go to agar.
But I wanna do one of 'em!

LC stupid easy, I have little 250ml media bottles. I boil 2g light malt extract to 500ml distilled water or 4g to 1000ml and evenly distribute between bottles. PC for 20 min. Drop a punch circle in from a clean plate and swirl it daily till ready. Test a little on agar, if it looks good after a few days pour it over some grains

D3, do you notice any difference in colonization times when compared to wedging up a whole plate and dropping that to a jar? Or is it more about expansion

I do the same, sometimes I'll multiple pieces to one jar if I'm a hurry though. I'll add a piece, then roll my jar with the open mouth facing the hood to bury the wedge. Add another piece.. repeat til done so the agars all spread out. It tends to stick together and get unseperable otherwise IME. I've been trying to find a better way to noc a jar up with a whole plate and maximize noc points, best I've found that I haven't tried yet is that super soft almost liquid agar method from Caps McGee and Munch's EzLC where you take a plate, drop into sterile water then shake the shit out of it.
I want to try the blender LC method, where you drop a plate into sterile water then blend. That really sounds like the best use of a plate, but adds a contam vector..

Man... that would be tedious as hell... I usually nocc 40 jars ot more at a time... id be there forever! Lol... ive got a pretty good, fluid motion that I scrape all the pieces out in less than 2 seconds... but they always pile together... so I end up shaking the shit outta the jar to get them un-stuck... maybe your way isn't so tedious after all. .. hmm..

It's not like a replace the lids each time or anything, it really doesn't take long, but obviously takes longer then just scraping em in. I leave the lid open to my jar and plate the whole time. I drop a wedge in, rotate with my hand that's already on the jar.. drop another wedge in, etc. They don't really get dispersed perfectly at this point, but there's a wall of grain between them so they don't get stuck and I can easily disperse them proper when I'm done. I only flame my scalpel once per plate and just move quick/careful
I just found that when scraping multiple pieces onto the top, I couldn't always get them satisfactorily unstuck and that was kind of defeating the purpose of multiple innoc points (and as you mention, wasting time trying to seperate them). I'm still experimenting though.. I like the agar to LI ideas due to how fast they colonize (and compared to LC, you don't have to wait for the liquid to colonize) but haven't tried them yet. I still have some open wounds after my last run with LC that tested clean...

How long does it take from inoculating the LC to full colonization (of the LC)? I've got some clean plates I need to do somethin with and I think my options are to grow out the plate and do the blenderless method or the 'true' LC. It seems like they'd take almost equal times though so I'm not sure one is better than the other

21/1 Update
Finally got around to taking T2's. Growth is still disorganized as all hell. Hoping there's something nice that I can take to grain by T3. Need to whip up another batch of no pours in the next couple of days. Had a vendor for cheap plates on mercadolibre but they've all disappeared and been replaced with ludicrously priced petris so I may bike up to the science store this weekend.

Bonus pic, here's my little habanero. About two months old, just moved her to a new pot
