Because I'm not a barbarian. Also I only ever have to shake my jars once. Let the mushroom take hold, shake it then, and it'll recover much faster.

here's an APE jar, this was liquid culture admittedly, it got time to recover and grow before I shook it, about a week or so.
But an immediate shake causes trauma and we don't want to traumatize our myc before its even begun...

Mushhead, I hate shaking my jars after dropping agar, but I always thought it was majorly mandatory

When I go to agar with my LAGM cultures, I'm gonna test this out for sure. Always down to skip an unnecessary step.

I also only shake once during colonization but I'll usually give it a little toss at inoculation to cover the agar with grain for more surface area to colonize but I definitely prefer to shake early rather than late. I think it speeds things up a bit compared to when I waited longer and if any tams are present a shake usually makes them show up a lot faster so I know to ditch it instead of it eating up real estate on the shelf. I used to wait til like 30% but these days closer to 15%. I think I remember reading a RR post that advised shaking when "a few dozen" grains are colonized and that definitely put me in the early shake camp because it makes sense, that's like 30-50 new points of growth after the shake if you distribute it well. I plan to take the concept and apply it to grain to grain using pint jars with millet as masters because the small grains will be a shit ton of nocc points when spread to the quart jars and I can probably get away with splitting that pint into 10 quarts

I usually only let it have about a week to nine days to recover and then shake.
after which I don't bother to shake it up again, its a waste of time.
After the first shake usually contams present themselves, but if you know you're working with clean culture why plan for it to contaminate?

When I drop wedges I just shake them to bury them. Then you got the whole thing jumping to grain even from the bottom of the agar.


I also used to wait til about 30% until somebody recently told me to shake a jar I had posted that had only jumped to the immediate grain next to it, there’s definitely something to early shaking. I’m realizing it’s sort of an art form knowing how to work the jars for maximum efficiency.

not so much worrying about the culture as much as the grain or sterile technique as I'm fairly new to g2g and had some mold out on me the first couple tries but mostly successful. I did some experimenting with grain prep last year and a handful of jars had mold that was not from the culture but it didn't usually show up until after a shake because it was hiding in the grains. I upped my PC time and the problem went away so I'm fairly confident it was the grain. it happens.

also sometimes one of the cats will get into the SAB without me noticing and then everything is fucked (dusty bastards)

to each their own but I think mycelium is much hardier than people give it credit for.

I have always shaken right after noc'ing, both because I thought I was supposed to when first starting, and to spread the wealth bit within the jar so to speak. Just to smash the agar and myc into as many grains/kernels as possible. Most contacts = more jump off points.
No clue if that's right or not, just what I've always done.

Yeah it can rub myc off into the grains for sure.


Or like when you use super soft plates so you can shake the jar and liquefy them.

I didn’t streak many plates on day 1, I believe 3 of each variety.  I think 1 had germ’d and I did a transfer on it, and that’s taking it’s sweet ass time.  I have been busy with stuff started before the 1st, and diving into that LC.  I’m loving it and wish I’d started that sooner.  And poured some to grain jars earlier and my god it was seamless, I love it!  I do think I’m going to start splitting up into smaller jars though, so as to not waste it.

I almost mentioned that too. We were talking about agar recipes recently and that's at least in part why I like them slightly softer. The wedges usually get smashed into almost nothingness (unless it sticks to the glass).


While we're talking about myc jumping off.....
I put a pin onto agar earlier when doing transfers. I swear the myc had made visible progress onto the agar by the time I finished transfers. No bullshit like 10 mins and this damn thing had jumped to the plate. Maybe I just never paid attention or noticed before but I was pretty surprised when I picked it back up. At first I thought I had fucked up and dropped some schmootz on the plate.

That’s what I was talking about back before we started. It could just be that everybody’s in the boring phase waiting on plates still but last year I’d say maybe 5% of the sign ups stuck it out.


Lagms got a real New Years resolution vibe to it. Everybody goes hard for a couple weeks and then it fizzles lol.

I haven’t been updating like last year but it’s coming. I didn’t want to add them to lagm but I started 30+ types of spores the first week and a half of the month. Gonna be constant pic dumps in a few weeks :dancer:




Late to the party here, but I did basically the same thing on accident... started 10 new varieties on 28DEC... lol then said I was gonna do 4 for LAGM... ended up with 8... just streaked 4 more new ones last night... lol. Thats on top of all the PE/Melmac hunting ive been doing...

Update: PE transfers T2 have happened. I took about 20 transfers from my 7 original germination plates and they are all looking decent, very wispy but maybe it will straighten up after a couple transfers. Very organized in the way they formed though which is a plus.

Excited to drop some of the T2 to some grain after they grow out a little bit! Although I might just wait till T3 as the wispyness of it is throwing me off.

Update:

20 JAN 2021

Had to pour a sleeve of plates today, as well as do some other stuff in the SAB, so I figured I’d make some progress here too. Took a couple of transfers from the Melmac plate that’s looking solid. Also grabbed a couple from a sketchy looking APE plate.


My growth has been slow, and not all looking the way I’d like, so I also went back to my germ plates to take some more T1’s. The germ plates were a bit better looking now that they have grown out a bit.


I decided to try a little experiment while I’m waiting. I think I picked the slowest varieties at the time of year my house actually cools off. Maybe this will get me some fruits quicker, or it will be a total shit show.

Here are some better shots of my TWC and TOC T2’s from today. Looks like one of the sectors on the TWC has some bacteria, but the other ones look great. I expect its time to take a T3 either tomorrow or the next day.

I've been cutting my plates into 9 pieces like this.


Each piece goes to it's own jar. I shake and roll the jar until the agar chunk is close to the middle of the jar. After a few days when the mycelium has colonized a few pieces of grain, I shake it up (pretty violently) and roll the jar grain around again. I keep repeat this process until there's theres visible colonization throughout the jar and then I leave it alone until it's fully colonized, shake one last time and spawn when it recovers. Lotkid taught me this.

This got shook right after this picture.

Then, it got shook again after this picture.

After a few days, it looked something like this.

Then it turned into this, got shook one last time, recovered and spawned.

I think a third plate is a good amount.

When I first started I messed around with wedges and went all the way up to two whole plates in a jar. Surprisingly was not that much quicker than a quarter or third plate wedge.



That’s right it was lotkid that was talking about it maybe a couple months ago.

I'm hoping to make some transfers tonight. I've got PE that needs T-1, some APEs that can possibly go to T-2 but I should probably wait a little longer. The GT syringe I bought from a vendor is dead. I can't get it germinate on agar, BRF cakes or pucks :frown: and I have some Chitwan to work with. It's MS so I don't want to send it to LC or transfer too many times so I'm a little confused as to what I should even do lol
