You can sterilize your own qtip in foil in the PC... and I if you want to hold true to tradition, you can grab some spores... but I'd doesn't hurt to run the clone also!

Thanks roarkell! lol, my mind isn't as sharp and fast as it used to be... I thought of that after I had posted. I'm hoping microdosing will help with that.
I could sterilize in an Instant Pot on high pressure for 45 mins too, yes? It'd take so long for such a small batch in a 23 quart...

Whatvi used to do is package up 10 or so in easy to open foil pouches... put them in a pint jar with the regular metal lid flipped or a plastic lid so it doesn't seal... since the InstaPot doesn't hit the pressures like the presto, 45 minutes should work... best of luck!

I use the instant pot to sterilize syringes/agar/lc.
Mine hits 15psi but only on the chicken setting.
I was thinking just mop up some of the dropped spores on that plate with the swab but the cap would work too.

You can post an update at the end here with a new post, see if you get some cool comments.
Then reply to your post, then copy to clipboard, plus cool comments, exit that reply page. Go to your original page/grow log and paste your update plus comments if you like.
And that will create your very own grow log like the others. See mine in my sig.

Background - I'm no expert. I'm a retired rather shy grannie trying to do it right and share what I learn. I have little experience and a growing (albeit slowly) knowledge of shrooms. I have my first successful grow thanks to all you wonderful people and this awesome site, I’m working on my second (first fruit popped up today!), and hope to keep it going. I’ve worked with spores once before - APE - and gosh that took me months to get them to germinate and produce usable myc… my first little guy popped his head out yesterday. Fingers crossed for all my tubs!
Spore source: Back in December 2020 I was gifted a jar of LC which had both mycelium growth and pieces of shroom in it. In mid January 2021 I put all those pieces to agar and this particular one sprouted a pin which dropped spores. I’m using those spores.

05/21/21
Used the Instant Pot on Pressure cook High setting for 1 hour to sterilize several individually wrapped q-tips.
In SAB
- Carefully unwrapped to expose one end of a sterile q-tip
- Being careful to not let anything but the q-tip over the top of a dish (? I call them plate or HG):
- Opened HG and dabbed up some spores, closed the plate
- Opened new agar HG, swabbed and closed it
- Folded the foil back over q-tip thinking since this was done in the SAB and I probably dabbed up a zillion spores I might use it again later on?
- Left all in the SAB and will keep there until I need to use the SAB for something else
… and now we wait
… … … …
and hope I didn’t screw up by leaving a small window cracked open and the HEPA filter running while working in the SAB

Just ran across cult with cats! (will you look at that !!! I figured out how to make a link). Now it’s not to promote the idea that you don’t have to be cognizant of cleanliness but OMG, it’ll set you free to relax a bit, cause a smile to spread across your face and fill you with warm fuzzies
So… I went to work fully expecting any myc growth will need to be cleaned away from contams but then again, these did come from a pin that grew in a sterile plate so maybe not everything? Time will tell. Here’s what’s in this pic:

- Original Plate from 1/21/21
- 1st plate swabbed 5/21
- 2nd plate swabbed 5/22
All following plates are transfers from the original pin done today, 5/22. Since the pin is too tiny to clone normally, I just cut it and transferred the pieces
- Cap from original plate’s pin
- Top of stem black with spores
- Bottom of stem
- Remaining stem
- Swabs I used to pick up spores - I figure they could be used again later, spores last forever, right?
Here we go folks!
@Inthepit - you mean post an update at the end here with a new post like this? Not sure what you meant by the next steps. I'll just have to do them and see if they make sense as I go. It's a bit of a learning curve for me and I appreciate your help

So far so good (even though I left the window cracked and HEPA on) we’ve got takeoff folks in ALL these plates, even the original from last January looks like it might have some teeny tiny pinheads (either that or it’s contam).

For a n00b, I’m paranoid I’ll mess up and loose the ORIG master. I realize I shouldn’t be since I have all these other plates (spores, cap, top section of the stem (black with spores) as well as trunk and base sections) germinating in separate plates (plus a bunch of other various plates from the original LC culture).
This is how I originally received the ORIG plates' shroom piece (in that LC) and how it looked shortly after I transferred the piece to the plate it’s in now.

I haven't done anything with that plate until now, when I found it had pinned and that pin had dropped spores (been distracted with getting APE spores going - the little darlines are popping their heads up in their Pasty EZ monos as I write - FINALLY!)
I get overwhelmed with the wealth of information and teks on how to keep a master culture going. I’ve read myc on agar will dry out but then Fahtster has a tek where colonized pop corn (or grain) is dried out, sits on a shelf that way until he wants to use it, rehydrates it in that same jar and it produces… so maybe if mine dry out all I’d need to do is rehydrate it in some LC? I read where dried shrooms were rehydrated too, but they had to be naturally dried, not dehydrator dried. There’s soOooo much to learn and do with this hobby.
Wish me luck! In addition to having these spore plates going for LAGM2021
- I have my first batch of four Pasty’s EzLC which are getting thick with myc so I need to dive in and pour on prepped quart rye jars - I'm sure I have WAY more LC than I need.
- I have seven 1/2 pint Josex’ Poke LCs ready and waiting for these spores’ myc to be transferred out and be ready and clean for the poke. Again I've probably made more than I need. Anyway... I read in the comments of that tek this LC has a long shelf life with no need to be refrigerated (which I assume would apply to all LC's, even Pasty's EzLC?) so I should have plenty of time. I’m using non-modified grey Ball plastic lids though, which was not recommended in the tek and I KNOW n00b’s are to follow teks to a T before switching things up but… I don’t have the means to make the holes, don’t want the raised eyebrows to borrow the tools, etc. and I read in other posts other folks have used unmodified lids so - leap of faith -wish me luck!
- I've got twelve quarts of prepped rye and eight more coming to use with the above Pasty and Josex jars. Hope I'm not being overly ambitious
Time will tell...
- Also, from that original jar of LC I have: three other original agar plates of shroom flesh waiting for me to figure out what and how to do what with them (slants - don't have any supplies; LC on a shelf - have all those supplies; Transfer some to another thickly poured agar plate - have experience and supplies; Give Fahtster's tek a go - need to first make his lid); From it’s A2G I put some colonized grains to agar; I took a clone from it’s harvest and transferred out from that. I wanted to learn, try out different things, see what happens, but most of all I was driven by my desire to make sure in one form or another I keep that original master going... and then I read about senescence
more to learn and understand...
... my mind is like a lite-up pinball machine ... ping a thought flashes ... ping ping ping a question lights up ping ping ping ping - I really need to read up more and understand the what, how, why of saving/using/storing a master culture...
- when do you start to be concerned about a culture loosing it’s vigor/potency (it’s already been four months of sitting in an agar dish)…
- how do I know if I poured the recommended amount of LC without using something to measure…
- what if I pour too much…
- remember to close the windows and turn off the HEPA filter before working in the SAB…
- don’t forget to put a hat on and wear your long sleeved t-shirt with the thumb holes cuz then when you put your gloves on it’s sleeves are held down far enough on your hands that the pulling up of the gloves doesn’t rub off dry skin which is probably where those little orange flecks of contam have been coming from cuz it never happen when you wore those t-shirts (bet even after liberally spraying and rubbing ISO ALL over hands and wrists after that dries there’s a remaining fleck or two that flys off - that won't happen with those T-Shirts, they are held down via the thumb hole and the gloves cover it a good inch of more)…
- go back to putting the heavily ISO sprayed bath towel down as the bottom of the SAB, you didn’t have contams then either…
- damn this sunburn, maybe I should wait till it’s all peeled off…
- and so it goes...
roarkell: Its good to see you taking the time to read up on everything... but occasionally it can scare new peeps in a different way... I can't speak for everyone but ive been at this a good while and I haven't reached the point of senescence with any of my original cultures... im just getting my slant game going, but ive been using a thickly poured plate as my master in the fridge... pop it out, take a few xfers tape it back up and back to the fridge... other than that, looks like you've got it pretty well sorted…

On 5/29/21 I made three transfers from one of the swabbed plates. I would have made more but didn’t have any more plates to use. The first two pics is what they look like today, two days later.
The last picture is of the top part of the stem that was all black with spores that I had transferred. Kinda cool how it looks don’t you think with the different types of myc on the same plate?
These look clean to me now so do I really need to transfer again? Am I being niave to think that?
I'd sure like to try a Josex Poke on that last plate if you guys think it's good to go... somewhere from the left side... but that plate is a biopsy with no transfers made from it yet...
I don't quite know what I'm looking for in 'clean agar.' To me, clean is basically nothing else growing, no spots, no goo, all white. Lot's to read out there... I better get on it!

This is what the xfrs doner plate looks like today:

roarkell: Of those 3 lined up, the last one looks the best but I'd still do a few transfers and see what they do... you want a more uniform growth pattern. Or at least that's what everyone says... lol. I like to take as many xfers as I can to see how things work. But im also team Full Send... if its clean, get it on grain! Lol.
To roarkell (thank you!) - or anyone: [LIST]How long do you wait, how far do you let the myc grow out, before you transfer again? I mean is there an ideal time to transfer? Are the first two pics grown out enough to transfer? lol... that 3rd pic certainly is. Is it not good to let them grow out that much before you do a transfer?
roarkell: #1- It depends on your intentions... if you're looking for fast propagation, you'll want to select faster myc, earlier... in that scenario, faster = better... #2- yes... lol. You can take transfers as soon as you can see mycelium... #3- ill let em grow and over grow... I like to use plate pins... no sense in doing all the work and not seeing it thru til the *proverbial* end, right?
Well, I didn't get it done in 2021. Picked it back up in 2022 though...

Well that looks like a good start on your grow log. So when you post an update here, copy the update to your grow log by editing it and watch it grow into something you can refer back to.
If you make a hyperlink to that grow log and put it somewhere like your signature you will have an easy way to get back to it
For the hyperlink, come up with a title, LAGM21, go to the grow log post and copy the link, that bunch of numbers in blue at the top, then come back, highlight your grow log title, click the hyperlink button and paste your link in.
Like Grannies Awesome LAGM21! To see the code for this just quote this post and you will see the hyperlink I made. Feel free to change the title!

Well this one got a head of me a bit, I was busy and only just got round the checking the tray today and the dam thing was pinning without its casing! So I threw on some old casing mix that I had. It was in a sealed tub so I'm hoping it's fine.
 The pan cyan bvi clone is in the larger tray, the two jars are some pan jambo from a different grow.

Texas Orange Caps - T1 Plates

Welcome aboard, Threads! You are off to a great start.
Can you believe we are 20 weeks into the year? It's been a weird one at la hacienda -- strangers in and out of the house almost every day, SWMBO and I have been fighting like cats and dogs, and work notified me that I'll have a drug test next month. All of that made me edgy and I ended up harvesting WG Box 1, Flush 3 early and dumping Box 2, F3. F3 of Box 1 was 130g - almost as big as Flush 2 of Box 2. Since my grows are for personal / friend use, it didn't go to the dehydrator -- just straight to the composter. Also dumped the bulk of my stash and relocated the rest.
Four of the Texas Orange Cap T1 plates showed good growth, so I transferred and now have 6 T2 plates going. I'll probably just do agar work for the next couple of weeks - it's easier to hide a few agar plates than a bunch of shoe boxes and grain jars.

1. In Edit, for colors , click the red 'A', pick a color in the long bar on the right, then also pick somewhere in the big color square. You can also just type YELLOW or RED. I've assigned color codes for peeps who I've quoted so they always get the same color, OMG OCD!
2. For your signature, click on your screen name, look for Edit Profile, then stick stuff, stuff is good, in your signature box. You can click Preview to check it out! It's right after Adult Content, which scared me away for awhile! I like this cool list command... Always had a soft spot for grannies, even made a painting with a couple innit, guess because I was helped to survive by a couple Great Aunts...
Anyway feel free to ask, I'm just goofing on the PC and watching the surf...

That sounds wonderful…I’m off to the softball field all day today to watch my 11 yo granddaughter play in 3 games - she’s a hell of a short stop and catcher and can pitch, pretty much play any position and isn’t shy about keeping the rest of the team focused. Lol, I’m not too biased but she IS good.
