monokaryotic mycelium growth @ spore inoculation center.

According to psilocybin production Alan R.

Quote:

The
lid on each container is lifted slightly and with
a sterilized pipette or syringe, a drop of this
spore water is placed on three or four different
parts of the agar surface. The container is covered immediately and the dish is left to stand
at room temperature for 3-5 days.
Radial growths of monokaryotic mycelium
will occur at each inoculation point. When
any two mycelia have grown to the point of
making contact with each other, mating
(somatogamy) will take place. Within a few
days these united mycelia will have become
dikaryotic organisms. Any portion of this
mycelial tissue can now be used to seed new
cultures as described later.

I am pretty sure each drop will form a diakariotic bond long before reaching adjacent colonies am I correct?

Or does it take longer and is it harder to form dikariatoc bonds than I think ?

Yeah, the haploid hyphae are sure to encounter a spore soon after germination. A few drops of spore soln. has too many spores for monokaryotic growth to go on for long.

so if I inoculated a plate and  took a transfer 16 hours later grabbing only a few hyphae is it possible that this 4th or 5th transfer is mono carry out

Not really no, you can't even see the mycelia by the time the spores/hyphae have mated. Unless you're dropping a single spore to agar.
