Need help on agar wedge selecting !
    #27088260 - 12/14/20 04:58 AM (3 years, 1 month ago)

Hello there !

I am new here and I am experimenting with agar for the first time !

I have a bunch of 3 to 4gen agar plates that started from two clones.
I am pretty confused with the results. It is obviously mycelium but their shape doesn't seems good.
I have some photos and my conclusions and I would like a double check about all of this because I'm still learning

The first ones (green marker clone) shows big uneven rhizomorphic growth that does not seems to be sectoring (and the mycelium tends to crawl up going in the air and not staying in the agar surface):

The only good one seems to be the mycelium on the bottom left of the last picture which is thick and evenly distributed.

The other transfers (blue marker clone) seems puffy/cottony but nearly every transfers I have done with this one gave this kind of results:

The second one seems to be a strain isolation but the mycelium seems very thin in comparison with the myceliums from the green marker clone.

Do you think it's bad if the mycelium is that thin ?
Someone can give me some advice for the next transfers or resources about the different types of mycelial growth ?

Thanks a lot, this forum is dope

PS: It is cheap brf agar, sorry it is cloudy

For the first 3 snatch the smallest end of a few of the ropes and transfer to new plates...Small as humanly possible the better your transfers will be.

It will probably look better after a transfer or two and thicken up.

It might be extra rhyzomorphic in response to a little contam it senses or maybe just genetic.

I've had  really wispy cultures do just fine like that last one.
