Agar Culture Died after Refrigeration?
    #27074377 - 12/05/20 11:28 PM (3 years, 1 month ago)

Hi everyone i saved a couple plates a few months back. I took them out of refrigeration, waited 4-5 days, and then made transfers from those plates. The plates arent showing any signs of growth after over 24 hours.

I am wondering if the cultures died? or are they just taking forever to recover?

My process before refrigeration was: first wrapped the plates in cling wrap, and then vaccuum sealed the plates together in a pounch
, and placed in the fridge. I stored them there for about 6 months

I know for sure the plates didnt dry out, thats why it kept them air tight. could the vacuum sealing process have somehow killed the culture? Could the cultures have suffocated from not having enough air? I thought there would be enough enough air in the petri dish to sustain them for a while?

I also found that the myc culture that used to be a white color now look brown after refrigeration? is that abnormal?

Had mine in the fridge for 4 months, just placed them in a small plastic tub as is..first transfer plate is pinning..maybe it was because of no air.

were they in the back of your fridge? they could have frozen and died. Sometimes when stuff gets pushed to the back where the blower is things can get frozen.

Ya ever seen frozen sour cream? It is not salvageable lol.

I keep my slants in an insulated lunch box in the bottom/back of the fridge where it won't get bothered and is somewhat protected from temp changes

It's probably fine. Even if it's not fine, it'll eventually be fine. So it's fine.
