Well The plates I used were also poured about 3 months ago, though the Machine Elf didn't seem to care. The spore plate from it has completely covered the plate.
So anyway, I took transfers today, from the 1 plate that had the single colony, on to freshly made agar so we will see if it makes any difference. Just stoked to have it going. I'll let the streaked plate go a little longer before transferring though.
Also did a Josex poke on the single colony plate so will see how it does in LC.

I'll try to ask here about my Semp plates, instead on the AGAR topic, hoping you don't mind and you could help me out. I have these 3 plates, but they're not good looking... from where would you keep doing transfers? Should I dump some of those? And last thing: are those white dots mushroom knots?

I just took a first transfer from plates inoculated almost a month ago. About 2 weeks of that was just germination though. Glad to hear they should speed up. I was getting worried.

At least we're not alone lol.
Can anyone chime in that has fast agar work for these? I have always used a simple recipe. 10g agar, 10g LME, 500ml water.

I’ve just always used the same recipe for every species.
Around 8g of agar and lme to 450ml

I was trying to wait for knots on a new culture I am working with and it just sit there forever. I cased it and 1.5 week later I have the first pins coming up.
My current strategy is to spawn to coir with coir pseudocasing (on the dry side), wait for that to be 100% colonized, then case (and keep the case wet)

I’ve also had a few cultures that refused to do anything until cased. Casing might get things moving for ya

Since you're on the topic of casing... I know Sub should be on the dry side but should casing be regular field capacity when first applied?
