Agar transfer inconsitency

Hey!

So I did a transfer recently off an isolate that I've been using for some time now. I noticed that when  I do many plates I get some that are rhizo and some that begin rhizo and become cottony as the grow. I also get some that are cottony from the point of transfer. I'm worried that the isolate may be contaminated with something and that I've been transferring around without knowing. I've got pictures of 4 different transfer plates from the same isolate done in front of a flowhood.

At first glance, I thought you were growing in a server room.. lol. Those all look great; nice and clean. Some varieties will grow cottony (this is known as tomentose) and some more rhizomorphic. Generally, rhizomorphic growth is preferred since it's easier to see contams, but great mushrooms can come from either. RustyWhytes, for example, tend to have tomentose growth on agar.

I have taken transfers from an isolate and some were nice, circular and rhizomorphic and other plates have sectors of rhizomorphic and tomentose growth. Regardless, all the transfers did well in bulk. From the research I've done, the mycelium respond differently to the agar media depending on its conditions. So any inconsistencies in agar media, for example food supply (I use LME) can affect mycelial growth and cause it to have different appearances.

The growth can also change depending on the nutrients dissolved in agar. Again, rhizomorphic vs. tomentose growth isn't really a good indication of health.

Awesome! I was using a SAB prior and I chalked it up to possible contams from that but with the flowhood I was confused as to why it was still happening. If its normal then I'm relieved to hear it.
