Mold ? or myc?
    #27063898 - 11/30/20 02:35 AM (3 years, 2 months ago)

I started up some spores. Used a sterile swab to scrape a spore print, and swiped the agar and rubbed the swab around a bit.

They look quite a while to germinate, i think 10 -14 days or something.

And now im not sure what im seeing is mycelium. I just finished reading
Mad's write up on hidden contams and from what i understand is you want to always choose Rizo myc because you can distinguish a fluffy mold contam among the rizo easier.

That being said, will spore throw out rizo mycelium right away? or do they always start tomantose?

Do i got tomantose myc here or some sort of contaminants mycelium meshed with the cubes?

The beauty of agar is that it lets you see the growth with such clarity that it lets you make good choices as to where to transfer from, what to pitch and what to keep. From spores, be always on the lookout for the most uniform, fast and vigorous growth you can get, don't bother with the rest, just pitch it.

Getting that out of the way, I'd keep the plates in the 2nd and 4th picture, the rest look suspect so you can pitch them.

From the second pic you can transfer from 6 o'clock and 12 o'clock from the 4th.

Thanks for the tips, im restarting from spores because I believe my plates are fucked with hidden contams as lately some of my tubs will seem to be bacterial which stunts growth, and eventually leads to mold moving in. Plus small fruits and low yields.

How long until I start to see Rhizo growth off these plates? I understand Its prefered to isolate Rhizo because its easier to spot contams amongs the strandy myceliums since mold Myc is usually fluffy?

You shouldn't get hung up on getting rhizo vs tomentose growth, just clean growth. You probably are never going to see rhizo growth on agar from that culture, but that's ok.
Very rarely germ plates will show rhizo growth anyway and many cultures simply refuse to grow rhizo on agar, nothing wrong with that tho, tomentose does not mean weak or anything negative.

I don't think tomentose growth makes it difficult to see contams. Look out for organized, uniform growth and you are good. Don't trust growth that is patchy or isn't uniform.

Also and very important, not all contams come from our cultures or from technique. The importance of grain choice and grain prep gets massively overlooked I think, it can make or break a grow easily so it's not wise to always look at culture work as the sole cause for our troubles.
