I assume the drying temp was around 100°C - right?

I doubt it. I have no idea what the actual temp is but thats very hot. the presto dehydrator has no settings, its either on or off.

I googled presto and just found devices for cooking and assumed this is some crazy hack. But searching for "presto dehydrator" led me to the specs. According to those it's set to 74°C.

Nice tub, josex.
I had some really nice, rhizo growth on a few TYC plates. I was able to isolate some thick ropey strands from them. This is the result so far from one of those transfers. The rest of them look very similar.

I'm just doing this for fun to keep agar work from becoming a boring routine. I've read that rhizo growth doesn't perform any better than tomentose growth. But that is about as much as I know. Thanks for the response. How would I go about isolating a single strain? What would be the easier type of growth to isolate? Or does it not matter?

Do you have an update on any progress? I would post in your thread but I received a warning that the thread was 59 days old since the last post. Most of the plates in one of your updates back in September looked very uniform. Is uniformity a genetic trait that I should be looking for?

Hard to tell through the plastic but it looks like it could be myc. Spores tend to look weird but also my experience with ape has been a lot of puffy growth like that.

I'm updating the three main posts (setup, timeline, pictures) on a regular basis. So, next time I can just use that as recipe and improve on some details. So, actually my last update was few days ago. The tubs are slowly colonizing. No sign of contamination - I guess I'll see the first flush next week.
No idea if uniformity is a genetic trait. But I isolate as if it was - meaning, I select uniform looking mycelium for isolation and inoculation. What I found very surprising is that dishes 1 to 4 looked very nice (uniform, bit rhizomorph) in the beginning. So, I was already quite sure I'd select those for the inoculation. But after a while the development did a 180 and 1 to 4 turned into weird messy shapes while the others developed quite nicely. At some point 1 to 4 developed (all) small amber droplets while the left over dishes 7 and 9 didn't. Given that 1 to 4 were isolated from a visually different part of the initial mycelium versus the others I assume the development must have been genetic because chance would be very unlikely.
