Liquid culture contamination concern
    #27057354 - 11/25/20 04:08 PM (3 years, 2 months ago)

My question today is about a honey liquid culture, that was inoculated via a liquid culture syringe. The  procedure of making the LC and inoculation were carefully and fully followed. Nothing I’ve seen on the forums can seem to represent what I have. So I turn to the communities expertise to help shed some light on this predicament. This culture was made on 11/10/20. The growth does look fluffy but it sits on the bottom not sure if the other photos people post up were stirred up before taking it or it naturally lis supposed to look like that. There is a darker part on top of the whiter growth. It looks similar in shade to the caramelized bits initially produced when PCing the LC. There hasn’t been any noticeable growth for the last week even with daily stirring.

If you need any more info to help clear things up please ask.

These might help you.
Honestly that looks pretty turbid and honey water lc isn't the best method out there imo.
I used LME/distilled water.

I hope you didn't use an entire syringe of LC to inoculate one LC jar. A 10ml syringe should be good for several expansion jars.

4 LC jars and 3 brown rice jars were inoc’d. The 4 LC jars all look the same no difference in anything could be seen. 1cc of the LCS was put into each jar. Is it unusual that growth isn’t kinda floating in the middle of the liquid but instead it’s just collecting together at the bottom?

It is natural for the mycelium to rest at the bottom. That isn't the issue. The issue is the cloudiness of the water that is above the mycelium. You should always test your lc's on agar before using them to inoculate grain jars. That is the only sure fire way of making sure the lc is free from contaminates.
