Spores 13 years old, still good?

Gurus,

I resurrected some spore prints(B+) from 13 years ago.  They where printed on index cards, dried, sealed with tape and sealed in zip lock.  Performed, with sterile technique in SAB, inoculation to potato dextrose agar with flamed inoculation loop. Got good amount of spores as the black spots visible on agar.  Put in incubator at 80 deg F and about 75% humidity.
After 2 weeks, no signs of growth or contamination.

Started another set of agar plates with oat soak water with more B+ and GT prints 1 week ago.  Same technique and same incubation.  Also, no signs of growth or contamination.

Should I just wait?  Should I try and soak some spores in sterile water for 24 hours and try more plates?

Should I spring for a microscope and see if I can see if the spores are still viable?  I thought the spores are pretty dam tough.

Any suggestions appreciated.

A microscope (I have two) won't help you distinguish dead spores from viable ones. It isn't like they'll move around or something. Agar is the best way to make that differentiation, and you seem to already have your answer.

Last year a friend gave me a B+ print on foil. It looked great, but after 3 rounds of trying on different agar recipes, I could never get any growth out of it at all. Meanwhile, 3 other prints germinated and eventually led to :mushroom: :mushroom2:. It's probably coincidence that my dud was also a B+, but I'd suggest get another spore print.

Don't do what?  Incubate? Wrong temp? Wrong humidity? Don't use prints that are 13 years old?  Well those prints are the only ones I have.

I find it hard to believe I can't revive any spores.  I thought spores are tough as hell and designed for long term storage.  They have been sealed and indoors the whole time.  No extremes of heat or cold.

I have more prints from this time period.  I will inoculate more agar plates and just wait it out. If I get contaminants before mycelium then I will give up.

It does sound like a 1 to 14 day soak in sterile water might help, thoughts?

I am really just curious at this point.

I have had spores take 3-4 weeks to germinate, make up some soft agar and load it up..you never know.one thing was for sure, after that long I knew it was myc. edit..they weren't as old as yours though..good luck.

Sorry....don't use an incubator and try to keep temps closer to 70F.

If you want to try rehydrating you can....I've never had to do it so that's why I linked the thread with RR and Hyphae's comments about the distilled water.

I once read a tek stating that closing your syringe (which you would have to make) with a lewer lock, and then wrapping a rubber elastic band around the plunger and lock would lead to a higher succes rate.
This would be because the added pressure from the elastic band would help hydrate the spores.
Never tried it myself, but thought it was interesting when I read it.
Maybe it helps!

13 years is a long time, but this would be my suggestion as well. Rehydrating in a pressured syringe, then put to agar and hope for the best.

Thanks everyone for the great suggestions, offers and advice.  I will try to re-hydrate and wait.  Will post when I get results.
