Hi guys,
I have been running four varieties on agar and recently I have been finding trouble with pretty much all of them.
After 5,6,7 transfers I am not seeing signs of healthy looking mycelium, in fact even when I transfer from a snowflake ❄️ looking master dish I am seeing a degradation in mycelium quality.
One thing I can see is a very thin border of what looks like totally transparent mycelium around some of the plates and I’m wondering if it is possibly fungal or bacterial contamination or whether it’s just the leading edge and I have a nutrient problem.
I have been using 7g lme: 10g agar: 500ml water. I have recently upped my lme to 10g and the results have slightly improved.
Mycelial masters please have a look at the pictures I have attached and let me know of any thoughts, diagnoses or comments you may have.

Thanks

Are you using any dye or is that the color of you lie?
Red dye can be bad for your cultures.
They look bacterial to me

Humectant: (Glycerine - E422), Water, Colour: (Cochineal - E120), Bulking agent: (Maltodextrin). May contain small traces of sulphite and wheat (gluten)

Red food coloring is made from a bug with anti fungal qualities. Too much of it and the cultures will resist colonizing the agar. They can survive a little though but I've quit using it ever because; why put an anti fungal in my fungi culture?
Op. You know the cultures are toast once opened for a photo like that, right? Good pics, by the way.
A lot of your cultures look like they've got bacterial problems to me. The best looking culture, and the one i'd say to pursue, is the one you posted in the first pic with the clearish edge. I'd take a small (your transfers are too big) piece from that into a new dish and update this thread as it grows out.
I can't say that it's always the case but I've had clean cultures that do that and just figured it was the myc proceeding through the agar. A little practice with a scope and you'll realize that there is myc in many places that you wouldn't at first think.
