So long time lurker first time caller
I have recently knocked up some agar plates in a SAB and a week later I have nothing. So bad news I have nothing good news I have nothing.

about how long do spores take to produce mycelium on agar?

Can fumes from using bleach instead of 70% IPA have an effect?

I have run just 6 different spore syringes on agar, but most have colonized or showed signs of life within a few days of inoculation.
Two I did with inoculation loops show nothing yet, but I assume the spores just didn't pair up.

I heard it's no bueno to use any bleach near your myco setup.
ISO is definitely preferred!

I am new to the scene as well.  I have had 4 separate spore syringe > agar inoculations with two different syringes. The first two took ~ 15 days to show any signs of growth.  The last two took about 4 days. 

My conclusion is that my agar recipe on the first two was too hard (I made some dumb math mistakes).  After lessening the agar agar ratio, Spores were able to do their thing much faster.

so I probably should have provided more information

I bought premade agar plates off scamazon(I didn't wnat to answer questions from my room mate as to what and why) so there are a few unknowns in regards to the agar itself.

I did use an inoculation loop to spread spores across the plate

and fortunately I picked up some 70% IPA before the next covid scare so I will knock up some new plates using that for a sanitizer and not use the inoc. loop to see if either of those makes a difference

In this season I recently had to wait too weeks for germination, or maybe it was the 2 year old print. Anyway, no growth means at least there is no contam, so if nothing works in theory you could just reinoculate the same plates again with more spores or a different print or whatever after 4 weeks total, if nothing has showed by then.

I like that idea. found myself wondering if all the spores in a syringe could get stuck to the walls of a syringe so I'm not getting any onto my agar

I read somewhere that it COULD damage spores I personally don't see the harm but I also did 3 streaks per plate as per the laboratory way.


Any way I noc'd up some more plates today. This time I used 70%ipa for disinfectant and only placed one or two drops in the center of the agar. Those are the only two things I did different than last time. Lets see what happens this go around.

A lot of TEKs call for VIGOROUS shaking of the MSS to disperse the spores in the solution better before you flame sterilize the needle.

I kind of like when it takes longer for the sake of not seeing contam. Definitely something to put you a little more at ease.
