Agar Plates

I scraped B+ spores from foil to agar plate last Thursday (one week ago). I am noticing some growth and it is growing daily. The black specs were from the scrape, but now I am starting to wonder if I messed up. I used the old "open oven" tek to assure I had respectable air flow away from my plates as well as flaming and cooling the scalpel (in isso) prior to scraping. I also sprayed my work area with isso. I am pretty positive that my contam level should be low (don't we all say that). Can you guys take a look and see if I appear to be on track? This is my first agar project so I would appreciate it. That looks like a lot of spores. Most people here will tell you to use a loop or some other form than scraping. That being said:

You are gonna have to wait it out to see if you get any mycelium growth. If you do, then do a quick transfer to another agar plate. If you do not see healthy mycelium growth, then back to the drawing board.

I tried the oven way once and it was not pretty for me. I'm not saying it doesn't work, I just did not like it or the results. I built a an 'incubator' out of a small metal shelf and plastic warp they use in warehouses. Works great. Waiting for some LEDs to get here and I'll try doing a TEK on it. Good luck.

That looks like way too many spores. I would recommend building/buying a inoculation loop as well as building and using a SAB if you don't already. I can tell you watch that YouTube channel, which is not a bad thing, but it is better to stick with what works. The oven tek is outdated, stick with what is proven to work better.

Make a loop Blades can damage spores.  Best to use inoculation loop and do a streak plate.  Below is link to Bod's video's.  There is one for Spore print to Agar.  Also going from flame to IPA is wrong way.  Best of luck to you but make a SAB.

Yep, after reviewing how they use and inoculating loop, I know what I did wrong. Anyway, a mistake that didn't cost, but 5 bucks. I will do it right the next time. I'm learning! haha
