genes for psilocin psilocybin production known?

Just curious if they have figured out genes related to enzymes etc that produce psilocybin?

I am sure now that things are opening up for mushrooms it will get sorted and allow for some hyper potent strains being developed. I dunno if would help/speed up traditional "breeding"/ searching but imagine crispr could help. but perhaps isolation would be more expensive then synthetic route for medical purposes. Thanks!

Absolutely. Multiple metabolic pathways have been published. One of the more recent one published uses glycolysis inherit in Sacchromyces cerevisae (yeast) to produce psilocbin, baeocystin and other tryptamines derivatives.

The total exogenous (to yeast) genes necessary to convert glucose -> psilocybin are five. 4 of the 5 genes are native to Psilocybe cubensis and the 5th from Catharanthus roseus because it's more efficient at converting Tryptofan to Tryptamine in yeast. While crispr isn't used in the transformation, the Cas9 protein is used to integrate multiple copies of the genes into the yeast genome.

Here's a link to the paper on producing psilocybin with yeast. The authors of the paper have gone on to start a psilocybin yeast startup known as OctarineBio.

More recently, a synthetic approach was released into the domain that uses pure chemistry (no cells or organisms) to produce pure psilocybin in high volumes, high purity and low cost. This approach is what is currently being used to provide the drug for clinical trials and will likely be used to produce pharmaceutical-grade pure products for prescription / clinical use.

Man, the yeast psilocybin is wild! Wow! I looked around a little to find how they synthesize it. I can rent an article for 48 hours for $40 from ACS publications, any knowledge on that knowledge?

More modern? That paper was from Feb of this year.

If you are looking for a pure chemical synthetic approach, this one was released in April by the founders of Usona:

This is essentially the approach John Hopkins uses to produce the drug for their clinical trials.
