about using less water in my agar recipe, due to pooling innoculant.

I squirted some innoculant onto my dishes but I see the excess moisture from the innoculant seems to be preventing the mycelium from rapidly growing further, almost trapping them in a bubble lick coating of moisture: I can see however the spores have germinated, but that's about as far as it goes.
It appears to be excess moisture

I was wondering if I can reduce the moisture content of my agar recipe from say 500 ml to 400 ml, would that do something..

anyone have experience with this?

Solving the problem the wrong way. Don't just squirt inoculant onto a plate. Use an inoculation loop and do it the right way

When I inoculate with a syringe, I of course use as little as possible, firstly. Then I tilt the plate to get the droplet to roll in a zigzag pattern. Most importantly to your question, I leave my inoculated plates unwrapped in my sab for about 24 hours to get rid of excess moisture.

Hi, these plates of mine are still unwrapped after like a week. Sadly only one is left uncontammed.

My eyes have finally been opened to what sterile technique is... :smile: Quite happy to say I'm pretty sure I will make a success of it this time, slowly but surely...

Bodhisatta, how do you use an inoculation loop with a spore syringe, or a jar with spore water... I just read prior to your post about using an inoculation loop - I had used a simple blunt edged butter knife to scrape off spores off the print :rolls eyes:

Anyways, thank you for the advice, I was very clumsy in controlling the plunger of the syringe, I shall certainly take care from now on to only drop a drop or so... mistakes go a long way to edify us :smile:
