Liquid Culture
    #27031063 - 11/10/20 04:50 AM (3 years, 2 months ago)

HEEY, I'm still a noob at working with cultures.

I made a liquid culture with Natal Super Strength (Cube from Kwazulu Natal South Africa) with a Spore print, liquid culture was water and 4% Honey.

There was nice mycelium growth in the jar but quite stringy or wavy, not really too solid, at the same time I made LC with some Puerto Rican Agar that looked better and more solid.

I inoculated a few jars with each and after 2 weeks the Puerto Ricans colonized great almost finished but the Natal Super Strength mycelium was barely visible.

There was growth but barely anything.

Was it perhaps because it was from a Spore Print and it had to germinate? Is that mycelium just weak or will it maybe be better after a few transfers?

I made another liquid Culture and inoculated it with that same Natal LC but I pressure cooked it too long and I believe it caramelized the honey since there were brown/gold chunks after PC'ing. It still grew though , I haven't tossed it yet, but the growth was more like a few solid balls clinging to those golden chunks. Is that proof that the culture just needs another transfer because it was straight from spores the first time?

Sorry for this crazy unstructured question

Any questions to clarify or advice would be appreciated.

I will attach pictures a bit later since I'm not home at the moment.

Maybe I'm missing it but did you go spores -> broth? Or did you germinate them on agar? Going spores straight to broth could work but iirc there's a pretty high contam risk. Going via agar gives you a better chance to start your LC with clean myc.
