Grow room setup and using Ez Dial MonoTubs?
    #27024486 - 11/06/20 06:39 AM (3 years, 3 months ago)

Hello everyone,
I am a new member and completely new to mycology but have been lurking and reading on here for a good while.  I have been doing my best to educate myself on everything related over the past couple of months and have spent many hours reading here, watching YouTube videos and reading the psilocybin growers Bible in preparation to begin growing myself. I had been considering several different teks like martha, automated monotubs, unmodified monotubs etc and was leaning towards landing somewhere in between and using modified monotubs with micropore tape in a grow room and was trying to answer a few of my lingering questions last night when I came across Pastywhytes ez dial monotub thread https://www.shroomery.org/forums/showflat.php/Number/26140339/fpart/20/vc/1#26140339 and had my mind completely changed. So, I have now decided that easy dial tubs will be the first method I experiment with.  I only found the thread last night and spent a couple of hours reading through it but only made it to Page 20 so far. I am very happy to have found it though and after having deliberated and researched other methods pretty extensively have decided that I would like to use this due to the simplicity and massive yields being reported by the OP and most of the others who have tried it.

Even with the extensive amount of research I have done so far I very much feel that I have barely scratched the surface as far as the knowledge and experience required to become an experienced and effective cultivator and I fully understand the level of dedication and hundreds of hours of experience, building knowledge and trial and error required to even come close to calling oneself an experienced cultivator and am perfectly willing to put in the time.  I have plans to start fairly small with 64 quart monotubs in a grow room I am putting together in a small space in my attic. I understand how variable yields can be and how easy it is for things to go wrong so I am trying not to rush too much and learn as much as possible before really leaping into things, however my ultimate goal is to try and produce at least a couple of pounds a month out of the space that I have for myself and the other people I know who are engaging in psychedelic therapy.

I do have quite a number of questions still though and I'm very much hoping that you someone can help me answer them.

Some of the most important things I am trying to figure out at this moment are regarding my grow room and the way that it should be set up for the best contaminant free growing conditions. I will attach pictures of the space I am using below but it is a triangle or a room with a height of 6.5’ width of 8’and length of 20’ divided into two sides. For the time being I am planning on setting up only one of them and will be working on it today. My plan is to start by doing the walls and ceiling out of reflective foam insulation and then enclose the entire thing including the floor in heavy duty 5mil plastic sheeting and add a zippered greenhouse door on the front. The adjacent room is heated but this attic area is not so I will be using an oil-filled type of space heater as I have seen recommended here and running a germguardian hepa air filter like this one that I will be picking up today https://www.homedepot.com/p/GermGuardian-Elite-5-in-1-Pet-Pure-True-HEPA-Air-Purifier-with-UV-Sanitizer-and-Odor-Reduction-22-in-Tower-AC4300BPTCA/300353737?source=shoppingads&locale=en-US.  There is currently not any mold or contamination to be concerned about within the attic that I can see, however I do plan on doing my best to disinfect and clean the area prior to building my room probably by using an ozone generator I have and running it for a couple hours before beginning the build in addition to spraying everything with 99% isopropyl alcohol or bleach and running the HEPA filter inside for a Time.  I still have some questions about what if anything I should be doing to maintain humidity and regulate fresh air exchange within my grow room as well as type of lighting and orientation.  I have actually purchased quite a lot of supplies for various methods over the past month or two, not knowing exactly what I was going to end up using and supplies not being common in my area, I grabbed up whatever I could with plans to either use it or sell it later. I have a small reptile humidifier as well as a House of hydro three disc and 12 disc ultrasonic humidifier and all supplies required to build a large or small humidifier with fans and humidity controller if needed. I also have oscillating fans, programmable timers and temperature controllers and various styles of LED lights ranging from 1,000 watt grow lights two smaller adhesive light strips that can be placed inside individual containers or above shelves if I were using that method and would love some recommendations on all of these things if you wouldn't mind.  I live in the Northeast where it gets very cold and dry in the winter so it may be tricky for me to regulate temperature and humidity (if needed) within this room, this is why I am planning on adding the additional reflective insulation and plastic sheeting in addition to trying to maintain a sterile environment.  I know the ez dial tub design is intended to maintain its own humidity within but because of the climate here I am thinking I may need to supplement the outside environment with something like a fogger or ultrasonic unit. I did see one response from somebody in a similar situation on the thread saying that they were using an ultrasonic in a small grow room in much the same climate with good results.

I do have some bags of colonized grain spawn that are ready to go into tubs now and more on the way, however I started these over a month ago when I still had very little knowledge and both the bags and the spores I started with were purchased online from sellers that seemed to have good reputations but I am unsure of the quality of either. I ended up with numerous contaminated bags some that are colonizing slowly and some that just didn't do anything at all either due to the bags and grain being poor quality or not sterilized correctly, my sterile technique not being good or possibly having bad spores or a combination of all three. I was using 3 lb bags of millet with self-healing injection ports and liquid cultures of spores of various strains of cubensis from sin City spores and injected all bags with 5 cc's and stored them in a closet at around 70° to colonize. Of the multiple bags I started I had to trash most bc of contamination and only had two that fully colonized, one of which just began to show a small amount of contamination at the end of the bag.  I am concerned about the contaminated bag but have been told that if I cut around the contamination within a margin of at least an inch and spray the area with isopropyl alcohol I should be able to effectively remove it and still use this bag. I am not sure where I am at with the remaining bags I still have colonizing as progress is very slow on them and I do not know if I should just try and use them now, give them more time to continue colonizing, or try and do something to speed up the process like add more spores, put them in a small greenhouse with a heater keeping them at a constant 75 and a light on etc.  I will attach photos of all of the bags in question below. The bag that did the best was started on 9/22 with Jedi mind fuck spores and I believe is as completely colonized as it possibly can be possibly even having been left for too long . The whole thing is a solid block of mycelium with some small spots of exudate present. I plan on shredding this and the other block both weighing 3 lb with a cheese grater and adding each to a monotub with somewhere between around 6 lb of substrate each in an attempt to try and get as much growth as I can out of them. A couple things I am wondering is whether or not I have let these go to the very limit or possibly waited too long on them as well as how big of a risk I am taking by trying to remove the contaminant from the one bag that just started showing some mold. I am also wondering if there would be any benefit to splitting these bags up between multiple mono tubs due to the fact that they are so heavily colonized, much more so than most photos of colonized grain spawn I have seen others using thus far.  I will be using some substrate I just purchased from a local grower and provider. I have 40 lb of it and it consists of lobster compost, manure, compost, forest humus, coco coir, rice hulls, vermiculite, straw, coffee and gypsum, and he claims to get very high yields from it. I have read many other substrate teks and it seems to me that the most popular option is simply 50/50 verm and cocoa however despite the fact that I have multiple pressure cookers, bags sealer buckets etc I was not prepared to make my own at this juncture and decided that if I was going to buy something I might as well try this.  would you have any predictions as to the outcome or effectiveness of a substrate of this nature? Is it probable that it will outdo any of the standard subs other people are running or maybe not do as well? I do plan on preparing my own substrate in the future with a large bulk sterilizer or pasteurization unit that I will put together out of a 16 quart pressure cooker and 60 quart cooler and I'm considering purchasing a large bulk sterilizer utilizing 55 gallon drums from someone in the area as I am interested in growing culinary mushrooms in my garage as well as potentially producing and selling grain bags and bulk substrate both online and locally.

Some things that I am unsure about is how I should go about spawning and fruiting my bulk substrate. I'm planning on setting up my tubs with the extra hole and likely taping that off during colonization and removing the tape during fruiting as I have seen others working in a similar climate suggesting to do this.  I think this would probably be my best bet or maybe I should tape all of the holes during the initial colonization time, especially since there is a chance I could experience some contamination in the tub that I am planning to use the block from which I will surgically remove the small contaminated area.  I could also just tape off the holes on that tub with either micropore or packing tape or just leave them open and hope for the best? One thing I am curious about is what I should do if I encounter any contamination during the additional colonizing or fruiting process in any of my tubs and how it is that such a contaminant would not quickly spread to every other tub seeing as they have open holes for fresh air exchange. If I do end up with a contaminated tub what are the possibilities of renewing the contaminant and saving the tub? I have seen some others here say that they have been able to do this with good success when it is isolated and caught early enough. If a tub has any significant degree of contamination or contamination that is scattered throughout should I completely trash that batch and get it out of the room immediately or is there any chance that it will still yield usable mushrooms and not put the rest of my grow in danger?

Also I am looking for recommendations as far as misting and fanning on my tubs and how often or when during the process I should do this or if I can possibly add an external ultrasonic humidifier and not need to do this as I have seen suggested by one or two others? From what I have seen from pastys posts it seems that he is usually only missing and fanning once during the spawning and fruiting process and have seen something similar being said by others with some saying that by adding an ultrasonic humidifier they did not end up having to miss or fan at all but would like to know what is best.

I would love some advice on what to do with the additional grain bags I have which are still colonizing and how I should go about starting my next batch for the highest odds of success. I purchased multiple strains of spores based on a lot of reading I did on some of the fastest easiest growers with the highest yields and have golden teachers, Jedi mind fuck, treasure coast, pesa, z strain, b+ and am unsure of the quality of them due to the high degree of contamination with my first batch of grain bags as well as some accidental mishandling of my own and suspended matter visible in the spore cultures.when I first got the spore cultures I did not know that they were supposed to be refrigerated in order to preserve them and had them at room temperature for several days before putting them in the refrigerator where they have been since. In all of them I can see small black particles suspended and floating and I'm not sure if this is typical or an indication that they might already be contaminated. I will attach some photos.

I do have Petri dishes and agar and have watched videos and read on making agar plates, syringes and things like agar slurry suggested by Willie Myco on YouTube for some of the fastest colonization in growth possible but still feel pretty unprepared to do this and I'm not sure what is going to be the best method for me to speed up my process and isolate a live strain which from what I have been reading seems can be quite difficult and take quite some time to get right. I have everything I need to put together a glove box or still air box with a small HEPA filter at one end and have put a down payment on a large laminar flow Hood but will not be getting it for a while so will be limited to one of the former methods for the time being. What would your suggestion be for this? Is a still air box built out of a tote and desktop HEPA filtration unit sufficient or should I go with the glove box for now instead? I do have several different styles of blowers and inline carbon filters that I could also modify for use with a HEPA filter  for this purpose as well if it would make sense. I have 10 more 3 lb grain bags consisting of corn, millet, oats, and coffee that I just purchased from the same person who made the substrate which I would like to get going and colonizing as soon as possible and am unsure what is going to be my best bet for these.although I have no experience isolating live strains I would like to do this for these if possible and inoculate from a liquid Master culture, however I have heard it can take time to get these right.  I was also considering making some agar plates, trying to figure out which strains are live and contaminant free and just using the whole 10cc syringe per bag this time around if it would be easier and save me time for this batch, although this might not be a good idea, im not sure.

If you made it this far, thank you for even taking the time to read all of this I know it is a lot.  I do have other questions but I am reading and watching constantly and trying to figure them out as I go. I very much hope that you are willing and able to help me out with some of this I would be eternally grateful if so. Regardless, thank you for even considering answering any of these questions and for your time.

That’s a lot of text but I did get the gist of the first half at least. When growing in monotubs or shoe boxes, the grow room conditions don’t matter much as long as the air isn’t totally stagnant. The tubs hold their own humidity and you just adjust how much air you let in to them.

It’s good to have a somewhat clean space to grow in but not necessary. You do all your clean work in a SAB or in the flow. And as long as you’re producing clean spawn the tubs will take care of themselves. And I just read a little more, go with a still air box with just two big arm holes, no fans or anything attached.

I see some sketchy grain bags at the end. I’d get into your agar right away. The only things to really worry about in the beginning is producing clean inoculate and clean spawn. Cubensis basically grow themselves if you just keep it clean. Syringes are rarely clean. Once you get things going then worry about finding good clones etc.

The way I do it is pretty quick and reliable. If starting a new variety: spore print to agar, transfer 2-3 times until definitely clean, let plate grow out to about 4/5, make Blenderless LI, pour about 10-20ml per qt of grain and shake really well to distribute the mycelium, 10-14 days later spawn my grain to coir/verm in shoe boxes (or a mono), wait for mushrooms.
